Based on available resources, laboratories used different RNA extraction methods during the COVID-19 pandemic. To evaluate the efficiency of different methods, this study compared Ct values in qRT-PCR, time taken for sample processing and PCR loading, the manpower involved, and the costs of consumables between manual and automated approaches. A cross-sectional study was performed on 120 nasopharyngeal/oropharyngeal swabs collected at VRDL for RT-PCR analysis. Based on the automated RNA extraction (Genetix, HT 96 Purifier) and RT-PCR (Trivitron PCR Kit) results, which analyzed both the E gene (screening) and ORF gene (confirmation), samples were assigned to four groups, namely Group-I (Ct 15-22), Group-II (Ct 23-29), Group-III (Ct 30-36), and Group-IV (Ct >36). Magnetic beads (Lab system, Trivitron) were utilized in the manual RNA extraction protocol. SPSS software, version 19.0, was used to perform the statistical analysis of the data. The two methods' Ct values were scrutinized using a paired t-test, in a group-wise fashion. A Z-test was utilized for evaluating variations among the other parameters. There was a statistically significant (p<0.05) difference in Ct values for target genes among Groups I, II, and III; nevertheless, no variation in the interpretation of the results was evident. The observed variations in time, manpower, and cost were statistically significant, with a p-value less than 0.005. The manual method demanded twice the workforce; it also consumed 40 minutes more, and the automated approach incurred 35 times the expense for consumables. Automated RNA extraction procedures consistently outperformed their manual counterparts, as evidenced by the study's findings regarding RNA yield. Despite the vaccine's efficacy, circulating false information on social media platforms regarding its safety led to hesitancy, particularly amongst women with young children, expecting mothers, or those who were breastfeeding. This research proposes to assess Saudi mothers' knowledge, attitude, and practice (KAP) concerning the COVID-19 vaccine, while simultaneously investigating how personal traits impact these aspects.
From April 2022 to July 2022, a cross-sectional, observational study investigated 293 Saudi mothers attending primary healthcare clinics at King Abdulaziz Medical City (KAMC) in Riyadh, Saudi Arabia. Participants responded to a pre-validated, self-administered questionnaire. This questionnaire consisted of 39 closed-response questions, grouped into four sections: participant characteristics, their understanding of, their feelings towards, and their practices concerning the COVID-19 vaccine. The English questionnaire was translated into Arabic and then retranslated into English by a different translator, finally being compared to the original English version to validate the translation's precision. Prior to the survey's deployment for data collection, a pilot study was undertaken with 20 individuals. The statistical analysis was executed via SPSS version 230, distributed by IBM Corporation in Armonk, NY. An analysis of the association between the four questionnaire parts was undertaken using the Chi-square test of proportion. Of those who took part in the research, 64% had ages below 40. 56% of the sampled population have graduated with a bachelor's degree or a higher academic attainment. A study revealed that 41% considered the official channels of the Ministry of Health to be the most important source of information concerning COVID-19 vaccines. Among the respondents, 45% demonstrated an extensive familiarity with the COVID-19 vaccine, and 62% had a positive view. A notable 40% of survey participants revealed that they withheld taking the COVID-19 vaccine until its mandatory status was enforced. In the survey conducted on parents of children between 12 and 18 years old, 78% reported that their children received the COVID-19 vaccine. Mothers under 40 years of age displayed markedly better vaccine awareness in contrast to the older demographic. The COVID-19 vaccine's KAP was notably impacted by variables including a younger demographic, higher educational levels, previous flu vaccine administration within the last three years, and adherence to childhood immunization schedules.
