From September 2015 to December 2021, a retrospective review of our pathology and radiology databases identified 95 patients. These patients, confirmed to have lung neuroendocrine neoplasms (NENs, 74 cases) and hormone-producing adenomas (HAs, 21 cases), had all undergone a pre-operative computed tomography (CT) scan. From the cohort of cases studied with biphasic CECT, a selection of 55 cases (18 HAs and 37 NENs) was subjected to meticulous review by three radiologists, each possessing different levels of experience. Following attack by natural killer (NK) cells, GSDMB3-expressing cells underwent pyroptosis, in contrast to GSDMB4-expressing cells, which exhibited a mixed pyroptotic and apoptotic death; GSDMB1/2-expressing cells, however, only experienced apoptosis. Despite NK cell-mediated cleavage attempts, GSDMB4 displayed limited resistance, indicating that GSDMB3 alone maintains complete functionality. Interferon and the chemotherapy drug methotrexate similarly induced GSDMB1-3 isoforms, which were the most abundant isoforms in the tested tumor cell lines. Expression of the cytotoxic GSDMB3/4 isoforms in bladder and cervical cancers, in contrast to frequently upregulated GSDMB1/2 isoforms, correlated with improved outcomes, indicating a protective effect of GSDMB3/4-mediated pyroptosis in these malignancies. Our findings suggest that tumors might impede and escape the pyroptotic response induced by killer cells, a process facilitated by the generation of non-cytotoxic GSDMB isoforms. Conditional inactivation of Bcl11b resulted in an increased count of intestinal tissue-resident memory (TRM) cells and their progenitors, in contrast to a decrease in both splenic effector and circulating memory lymphocytes and their progenitors. Circulating memory cell loss was partly attributable to Bcl11b-/- circulating precursors exhibiting increased homing to the intestines, without significant modifications to their respective programs. Bcl11b-/- TRM cells experienced alterations in transcriptional programming, with reduced levels of multipotent/multifunctional (MP/MF) program genes, including Tcf7, and augmented expression of effector program genes, such as Prdm1. The differentiation of intestinal CD8+ TRM cells, a process influenced by Ahr, a transcription factor, is further constrained by the regulatory actions of Bcl11b. Deregulation of TRM programs, while associated with TRM cell accumulation in the intestine, nevertheless resulted in a poor recall response. In Bcl11b-deficient TRM cells, the expression of genes belonging to the MP/MF program was observed to be lower, mirroring the reduced chromatin openness and decreased concentration of activating histone modifications at these specific genomic regions. Conversely, effector program genes exhibited a heightened state of activating epigenetic modification. These findings pinpoint Bcl11b as a leading participant in the intestinal memory cell tissue residency program, acting upstream of Tcf1 and Blimp1, thereby promoting multipotency and constraining the effector program.
