Altering the NTD structure in both spike variants adversely affects the binding of all NTD-specific antibodies assessed, located both inside and outside of the NTD supersite. One of the observed variants displayed a change to the signal peptide cleavage site facilitated by P9L, leading to the loss of a critical disulfide bridge. This represents a unique escape mechanism of substantial antigenic impact. Although the occurrences of deletions and disulfide mutations are infrequent, similar modifications have arisen independently in multiple other lineages, suggesting a probability of greater dominance in future iterations. With the unrelenting spread of SARS-CoV-2, the observed adaptability of the NTD foreshadows its broad capability to circumvent the immune system.
