Deep-water renewal (DWR) events are characterized in the Strait of Georgia, Canada using 11 years of real-time physical and chemical oceanographic data and seafloor videos. At least 6 DWRs occur per year at 300 m water depth and each event continues for over 3 days. They initiate during neap tides and are associated with increased turbidity. In the spring, DWRs introduce cold, oxygenated and nutrient-poor waters, and in the fall they introduce warm, oxygen-depleted, nutrient-rich and saline waters. Although the timing and magnitude of DWRs differ from year to year, we demonstrate that they are not restricted to two seasons, but continue throughout the year. High-resolution videos of DWRs show that these events comprise a plume of high suspended sediment concentration that flows parallel to the basin axis and deposits approximately 1.5 cm per event.The establishment of the Sanjiangyuan National Park (SNP) favours implementation of strictest ecological protection on the Tibetan Plateau, thus firmly ensuring national ecological security. To understand ecological background in the SNP, spatial and temporal variations of ecosystem, and its services during the period 2000-2015 and significance identification were analysed by using the methods of remote sensing, GIS and model simulation. The results showed that (1) Area with extremely important ecosystem services accounted for approximately 51.4% of the SNP's total area, of which extreme importance water regulation, soil conservation and sand fixation regions contributed 15.3%, 13.7% and 22.4%, respectively. (2) The SNP had formed a spatial pattern of ecosystem services with water regulation as core in the eastern part, soil conservation as core in the central part and sand fixation as core in the western part. (3) For the period 2000-2015, water regulation service generally improved in the SNP. this website Soil conservation service also improved overall; and sand fixation service exhibited a decreasing trend due to reduction in wind speed and vegetation coverage. (4) Climate warming and humidification, combined with the implementation of ecological protection project in the SNP were the primary reasons for ecosystem services improvement. However, grassland degradation had not yet been fundamentally suppressed, and vegetation coverage was still declining in regional areas. For strict protection and sustainable use of the SNP and its natural resources, overall planning and scientific layout should be paid more attention, and classification and subarea protection should be implemented based on natural ecosystem laws.Poly(N-isopropylacrylamide) (PNIPAm) is widely used to fabricate cell sheet surfaces for cell culturing, however copolymer and interpenetrated polymer networks based on PNIPAm have been rarely explored in the context of tissue engineering. Many complex and expensive techniques have been employed to produce PNIPAm-based films for cell culturing. Among them, spin coating has demonstrated to be a rapid fabrication process of thin layers with high reproducibility and uniformity. In this study, we introduce an innovative approach to produce anchored smart thin films both thermo- and electro-responsive, with the aim to integrate them in electronic devices and better control or mimic different environments for cells in vitro. Thin films were obtained by spin coating of colloidal solutions made by PNIPAm and PAAc nanogels. Anchoring the films to the substrates was obtained through heat treatment in the presence of dithiol molecules. From analyses carried out with AFM and XPS, the final samples exhibited a flat morphology and high stability to water washing. Viability tests with cells were finally carried out to demonstrate that this approach may represent a promising route to integrate those hydrogels films in electronic platforms for cell culture applications.Fluorescent proteins with varying colors are indispensable tools for the life sciences research community. These fluorophores are often developed for use in mammalian systems, with incremental enhancements or new versions published frequently. However, the successful application of these labels in other organisms in the tree of life, such as the fungus Candida albicans, can be difficult to achieve due to the difficulty in engineering constructs for good expression in these organisms. In this contribution, we present a palette of Candida-optimized fluorescent proteins ranging from cyan to red and assess their application potential. We also compare a range of reported expression optimization techniques, and find that none of these strategies is generally applicable, and that even very closely related proteins require the application of different strategies to achieve good expression. In addition to reporting new fluorescent protein variants for applications in Candida albicans, our work highlights the ongoing challenges in optimizing protein expression in heterologous systems.It is widely recognized that monocytes-macrophages adopt a wide variety of phenotypes, influencing the inflammatory activity and demyelination in Multiple Sclerosis (MS). However, how the phenotype of human monocytes evolves in the course of MS is largely unknown. The aim of our preliminary study was to analyse in monocytes of relapsing-remitting and progressive forms of MS patients the expression of a set of miRNAs which impact monocyte-macrophage immune function and their communication with brain cells. Quantitative PCR showed that miRNAs with anti-inflammatory functions, which promote pro-regenerative polarization, are increased in MS patients, while pro-inflammatory miR-155 is downregulated in the same patients. These changes may indicate the attempt of monocytes to counteract neuroinflammation. miR-124, an anti-inflammatory marker but also of myeloid cell quiescence was strongly downregulated, especially in progressive MS patients, suggesting complete loss of homeostatic monocyte function in the progressive disease phase. Profiling of miRNAs that control monocyte polarization may help to define not only the activation state of monocytes in the course of the disease but also novel pathogenic mechanisms.
