To assess the reliability and construct validity of a self-paced, submaximal run test (SRTRPE) for monitoring aerobic fitness. The SRTRPE monitors running velocity (v), heart rate (HRex), and blood lactate concentration (B[La]), during three 3-minute stages prescribed by ratings of perceived exertion (RPEs) of 10, 13, and 17.
Forty (14 female) trained endurance runners completed a treadmill graded exercise test for the determination of maximal oxygen consumption (VO2max), v at VO2max (vVO2max), and v at 2mmol·L-1 (vLT1) and 4mmol·L-1 (vLT2) B[La]. Within 7 days, participants completed the SRTRPE. Convergent validity between the SRTRPE and graded exercise test parameters was assessed through linear regression. Eleven participants completed a further 2 trials of the SRTRPE within a 72-hour period to quantify test-retest reliability.
There were large correlations between v at all stages of the SRTRPE and VO2max (r range = .57-.63), vVO2max (.50-.66), and vLT2 (.51-.62), with vRPE 17 displaying the strongest associations (r > .60). Intraclass correlation coefficients (ICC3,1) were moderate to high for parameters v (range = .76-.84), HRex (.72-.92), and %HRmax (.64-.89) at all stages of the SRTRPE. The corresponding coefficients of variation were 2.5% to 5.6%. All parameters monitored at intensity RPE 17 displayed the greatest reliability.
The SRTRPE was shown to be a valid and reliable test for monitoring parameters associated with aerobic fitness, displaying the potential of this submaximal, time-efficient test to monitor responses to endurance training.

Jerk-based measures have been utilized to examine dynamic balance because of their ability to quantify changes in acceleration and may provide an understanding of the postural corrections that occur during stabilizing following a jumping task. The purpose of this study was to compare acceleration and jerk following a jump stabilization task between individuals with CAI and the uninjured controls. Adavivint chemical structure Thirty-nine participants volunteered to participate in this case control study. Participants completed a jump stabilization task requiring them to jump off 2 feet, touch a marker set at 50% of their maximal vertical jump height, land on a single limb, and maintain balance for 3 seconds. Acceleration was calculated as the second derivative, and jerk was calculated as the third derivative of the displacement of the resultant vector position. Participants with CAI had greater acceleration (mean difference = 55.6 cm/s2; 95% confidence interval, 10.3 to 100.90; P = .017) and jerk compared with the uninjured controls (mean difference = 1804.5 cm/s3; 95% confidence interval, 98.7 to 3510.3; P = .039). These results suggest that individuals with CAI made faster and more frequent active postural control corrections to regain balance following a jump compared with the uninjured controls.
Elder abuse, neglect, and exploitation are under-detected and under-reported. The purpose of this qualitative study was to describe out-ofhospital emergency care providers' experiences of identifying elder abuse.
Individual theme interviews were conducted with nine prehospital emergency care providers and three community paramedics in spring 2019. The transcribed data were analyzed using inductive content analysis.
Although the short duration of care contacts made the identification of elder abuse challenging, the emergency care providers detected indicators of physical, psychological and social abuse, unethical action, material exploitation, and self-neglect/self-abuse. The professionals based their observations on patient and family interviews, on clues in the home environment, on caregiving quality combined with the patient's medical history, and on physical signs, which were the easiest to identify.
The identification of elder abuse may be improved by multiprofessional collaboration, by increased attention given to risk groups and common indicators of abuse, and by adoption or creation of screening tools to assist detection and reporting. Training on the detection of elder abuse should be included in nursing and social work curricula and in the continuing professional development of emergency care providers.

Such efforts are challenged by metabolite transport limitations, which arise when complex plant pathways distributed across organelles and tissues are reconstructed in unicellular hosts without concomitant transport machinery. We recently reported an engineered yeast platform for production of the tropane alkaloid (TA) drugs hyoscyamine and scopolamine, in which product accumulation is limited by vacuolar transport. Here, we demonstrate that alleviation of transport limitations at multiple steps in an engineered pathway enables increased production of TAs and screening of useful derivatives. We first show that supervised classifier models trained on a tissue-delineated transcriptome from the TA-producing plant Atropa belladonna can predict TA transporters with greater efficacy than conventional regression- and clustering-based approaches. We demonstrate that two of the identified transporters, AbPUP1 and AbLP1, increase TA production in engineered yeast by facilitating vacuolar export and cellular reuptake of littorine and hyoscyamine.
