These results indicated that the effect of spermidine and spermine on anti-aging is related with improving autophagy and mitochondrial function.BACKGROUND Forkhead box K1 (FOXK1) is a transcription factor belonging to the Forkhead box (FOX) family and is closely related to the development of various cancers, but the functional mechanism through which FOXK1 regulates autophagy and epithelial-mesenchymal transition (EMT) in the acidic microenvironment of gastric cancer (GC) remains unclear. RESULTS Our results indicated that the inhibition of FOXK1 induced autophagy and thus exerted antimetastatic effects in an acidic microenvironment. The dual inhibition of mammalian target of rapamycin (mTOR) and FOXK1 enhanced autophagy and reversed EMT of acidic GC cells. In addition, FOXK1 activated transcription in conjunction with the MAZ promoter. CONCLUSION Together, our results suggest that FOXK1 can be used as an independent prognostic indicator for GC patients. We also revealed a new strategy involving the cotargeting of FOXK1 and autophagy to reverse the effects of EMT. MAZ is involved in the development and progression of GC as a downstream target of FOXK1. METHODS Here, the cellular responses to the inhibition of FOXK1 in GC were studied in vivo and in vitro through wound healing assays, transwell assays, Western blotting, laser confocal microscopy and transmission electron microscopy. The molecular mechanisms of FOXK1 and Myc-associated zinc finger protein (MAZ) were studied via chromatin immunoprecipitation sequencing (ChIP-seq), bioinformatics, Western blotting, and quantitative real-time PCR (q-PCR).BACKGROUND The precision of adult height prediction by bone age determination in children with idiopathic growth hormone deficiency (IGHD) is unknown. METHODS The near adult height (NAH) of patients with IGHD in the KIGS database was compared to adult height prediction based on the Bayley Pinneau (BP) in 315 children and based on the Tanner-Whitehouse 2 (TW2) method in 121 children. Multiple linear regression analyses adjusted for age at GH start, mean dose and years of of GH treatment, and maximum GH peak in stimulation test were calculated. RESULTS The mean underestimation of adult height based on the GP method was at baseline 4.0±0.5cm in girls and 4.4±0.4cm in boys, at 1 year of GH treatment 2.0±0.3cm in girls and 0.5±0.3cm in boys, while at last bone age determination adult height was overestimated in mean by 0.4±0.4cm in girls and 3.7±0.3cm in boys. The goal of this position paper on peptide hormone analysis was to establish laboratory guidelines that may contribute to improve optimal diagnosis and treatment control of DSD. The essential peptide hormones used in the management of patients with DSD conditions are follicle-stimulating hormone, luteinising hormone, anti-Müllerian hormone, and Inhibin B . In this context, the following position statements have been proposed serum and plasma are the preferred matrices; the peptide hormones can all be measured by immunoassay, whilst use of LC-MS/MS technology has yet to be implemented in a diagnostic setting; sex- and age-related reference values are mandatory in the evaluation of these hormones; and except for Inhibin B, external quality assurance programs are widely available.To implement effective mitigation measures to stop the flow of litter from land-based sources into the sea, it is important to identify key pollution sources and to monitor litter trends over time. We sampled plastic and other anthropogenic debris in urban stormwater run-off in Cape Town, South Africa, from three catchment areas representing different land-use types residential, industrial and a mixed commercial/residential area. Sampling was conducted in 2018-19 by placing nets over stormwater outlets during rain events. Organic matter constituted 79% of material by dry mass (industrial 51 ± 22%, commercial/residential 86 ± 4%, residential 88 ± 13%). The nets caught 5-576 anthropogenic litter items·ha-1·day-1 (2-377 g·ha-1·day-1) with significantly higher densities in the industrial and commercial/residential areas than the residential area. Among anthropogenic litter items, 40-78% were made of plastic (52-64% by mass). Most plastic items were single-use packaging, but industrial pellets washed out of the industrial outlet during every rain event. Compared to a similar study conducted in 1996, the number of litter items decreased by ~20% in the industrial and residential areas but tripled in the commercial/residential area. The proportion of plastics in the litter stream was broadly similar to 1996. We extrapolate that some 60-570 t of plastic are released from Cape Town stormwater outlets annually, which is orders of magnitude less than predicted by global models. It is nevertheless clear that stormwater outlets are a significant source of litter into the sea. Intercepting this waste before it reaches the sea would greatly reduce litter loads in coastal waters around Cape Town.
