LF data were also validated by western blot and MRM based experiments. On standard mixtures, both CBN methods showed an excellent behavior in terms of reproducibility and coefficients of variation (CVs) in comparison to the other SpCs approaches. Overall, the CBN(P) method was demonstrated to be the most reliable and sensitive in detecting small differences in protein amounts when applied to biological samples.This study was designed to explore the role of local knowledge (LK) in enhancing the resilience of Dinki watershed in the central highlands of Ethiopia. The data were collected through focus group discussions, key informant interviews, and household surveys. The qualitative data were analyzed through thematic analysis. Whereas descriptive statistics and logistic regression were used for quantitative data analyses. The result showed that the majority of the respondents have local knowledge and used in their daily activities. Furthermore, the LK is reported to have the adaptive capacity in managing land resource degradation, treating disease, mitigating food insecurity, and enhancing social capital, among others. Likewise, the logistic regression analysis showed that age, gender, and education status of the household head were significantly correlated (p less then 0.05) to knowledge level, indicating their predictive power for the acquisition of LK. Accordingly, old-aged (60+ years) male respondents who attended primary education were higher to acquire LK than their counterparts. The result realized that the adaptive roles (land resource management, medication, climate change adaptation, etc.) of local knowledge systems could contribute to enhancing resilience. More importantly, the presence of social mechanisms is insurance to maintain LK. Thus, both intergenerational and intragenerational information gaps are filled with education and promotion on the roles of local knowledge systems. Besides, local-decision options should participate in custodians to share their experiences, that could contribute to sustaining ecosystem resilience.Bio-asphalt binders have been proposed as replacements for traditional asphalt binders, owing to advantages such as environmental protection, low costs, and abundant resources. However, a limitation of bio-asphalt binders is that their high-temperature performance is not suitable for pavement construction. In recent years, nano-particles have been widely used to improve the pavement performance of asphalt binders, particularly the high-temperature performance. Thus, the nano-particles might also provide a positive modified effect on the high-temperature performance of bio-asphalt binders. Based on this, five types of nano-particles including SiO2, CaCO3, TiO2, Fe2O3, and ZnO are selected for the preparation of modified bio-asphalt binders, using different dosages of nano-particles and bio-oil. The high- and low-temperature performances, aging resistance, workable performance, and water stability of the nano-modified bio-asphalt binders and mixtures are investigated. The results reveal that, the high-temperature performance and aging resistance of the nano-modified bio-asphalt binders and mixtures are improved at increased nano-particle dosages, whereas their low-temperature performance is slightly weakened. The effects of the nano-particles on the workable performance and water stability are insignificant.The introduction of transgenic cotton (Bt-cotton) for controlling bollworms has resulted in increased production; however, the residual effects of mulches from Bt-cotton are poorly understood. Therefore, the current study evaluated the impact of Bt and non-Bt cotton mulches on soil properties, weed dynamics and yield of winter crops sown after cotton. Three different winter crops, i.e., wheat (Triticum aestivum L.), canola (Brassica napus L.) and Egyptian clover (Trifolium alexandrinum L.) and two mulch types, i.e., Bt mulch (obtained from Bt-cotton cultivars, i.e., 'CIM-616' and 'GH-Mubarik') and non-Bt mulch (obtained from non-Bt cultivars, i.e., 'CIM-620' and 'N-414') were included in the study. The mulches were applied at a rate of 2 t ha-1 before planting the winter crops. The Bt and non-Bt mulches differentially affected soil properties, weed dynamics and productivity of winter crops. The non-Bt mulches decreased the soil bulk density and penetration resistance, while increased the soil porosity. CD532 in vitro Wheat crop increased the soil porosity, pH, available N and soil organic matter content. Overall, non-Bt mulches improved the productivity of winter crops compared with Bt mulches. The toxins released by Bt mulches lowered the weed density; however, it negatively influenced soil properties (bulk density and available nitrogen) and productivity of winter crops. Therefore, appropriate crop rotation measures may be opted for the soils cultivated with Bt-cotton to conserve soil and achieve yield sustainability for the crops sown after cotton. Nonetheless, non-Bt mulches can be used for improving soil properties and productivity of winter crops.
False-negative malaria rapid diagnostic test (RDT) results amongst symptomatic malaria patients are detrimental as they could lead to ineffective malaria case management. This study determined the nationwide contribution of parasites with Pfhrp2 and Pfhrp 3 gene deletions to false negative malaria RDT results in Ghana.
This was a cross sectional study where whole blood (~2 ml) was collected from patients presenting with malaria symptoms at 100 health facilities in all the regions in Ghana from May to August 2018. An aliquot of the blood was used to prepare thin and thick blood smears, filter paper blood spots (DBS) and spot a PfHRP 2 RDT kit. The remaining blood was separated into plasma and blood cells and stored at -20°C. Plasmodium parasite density and species identity was estimated from the blood smears. Plasmodium falciparum specific 18S rRNA PCR, merozoite surface protein (msp 1) and glutamate rich protein (glurp) gene PCR were used to identify P. falciparum positive samples, which were subjected toia diagnosis requires continuous surveillance of parasites with Pfhrp 2 gene deletions.
