This study focuses on understanding of the Escherichia coli release from different particle fractions (0.25-, 0.5-, 1-, and 2-mm) of solid manure and evaluating different influent boundary conditions during cell release from manure and when a solid manure is applied to the soil. The 0.25-mm and 2-mm particle sizes resulted a greater cell release compared to 0.5-mm and 1-mm fractions (p less then 0.05). The shape and magnitude of the cell release curves (CRCs) from the original manure bulk were mainly influenced by the two 0.25-mm and 2-mm fractions, respectively. The arithmetic mean for normalizing the CRCs and the time variable- based normalized CRCs for the manure-treated soil were the robust variables in evaluation of the experimental data. However, a single maximum bacteria concentration could provide the realistic dataset for the modeling process. Evaluation of the root-mean-squared-error and Akaike criterion showed that the two- and three-parametric models are recommended for simulating the cell release from solid manure in comparison with one parametric models. This study also suggests considering separate microbial release evaluations, with regards to influent concentration, for manure and manure-treated soils to propose best management practices for controlling bacteria pollution. Further research will reveal the key roles of different woody components and soluble material ratios for the various solid manures in bacteria release.A quantitative structure-activity relationship (QSAR) study was conducted using nineteen previously synthesized, and tested 1-aryl-6-hydroxy-1,2,3,4-tetrahydroisoquinolines with proven in vitro activities against Plasmodium falciparum. In order to computationally design and screen potent antimalarial agents, these compounds with known biological activity ranging from 0.697 to 35.978 μM were geometry optimized at the B3LYP/6-311 + G(d,p) level of theory, using the Gaussian 09W software. To calculate the topological differences, the series of the nineteen compounds was superimposed and a hypermolecule obtained with s ¯ = 17 and 20 vertices. Other molecular descriptors were considered in order to build a highly predictive QSAR model. These include the minimal topological differences (MTD), LogP, two dimensional polarity surface area (TDPSA), dipole moment (μ), chemical hardness (η), electrophilicity (ω), potential energy (Ep), electrostatic energy (Eele) and number of rotatable bonds (NRB). By using a training set composed of 15 randomly selected compounds from this series, several QSAR equations were derived. The QSAR equations obtained were then used to attempt to predict the IC50 values of 4 remaining compounds in a test (or validation) set. Ten analogues were proposed by a fragment search of a fragment library containing the pharmacophore model of the active compounds contained in the training set. The most active proposed analogue showed a predicted activity within the lower micromolar range.Sale of controlled drugs without prescription is a burning issue in developing countries like Pakistan. Illicit sale practices lead towards drug abuse and misuse among youngsters and negatively impact the health of youth and economy of any country. Present study aims to highlight the illicit sale practices at community pharmacies/drug stores of Punjab, Pakistan. Study was conducted at community pharmacies/drug stores (n = 200) of Punjab, Pakistan. Sales men at pharmacies/drugs stores were interviewed and then their statements were cross verified by sending fake customers at their community pharmacies to check the extent of illicit sale practices by them. Gathered data was analyzed using SPPS-22. Out of 200 pharmacies, pharmacists were physically present at 5% of pharmacies (n = 200), rest of the 95% pharmacies (n = 190) were being run by non-qualified persons and were found to be engaged in illicit sale practices. Controlled drugs were being provided to customers without prescription, which is a dilemma and need to be addressed for effective policy making.
