Background Deregulation of the PI3K signalling pathway is frequent in squamous cell carcinoma of the head and neck (SCCHN) and may be implicated in radioresistance. We report on the results from a phase I 3 + 3 dose escalation study of alpelisib, a class I α-specific PI3K inhibitor in combination with concurrent cisplatin-based chemoradiation (CRT) in patients with locoregionally advanced SCCHN (LA-SCCHN). Methods Eligible patients had previously untreated LA-SCCHN and were candidates for CRT. The primary objective was to evaluate safety and determine the recommended phase II dose (RP2D). Alpelisib was given orally once daily at two dose levels 200 mg and 250 mg. CRT consisted of cisplatin 100 mg/m2 IV every three weeks and standard fractionation radiotherapy (IMRT) 70 Gy in 35 fractions. Results Nine patients were enrolled (six alpelisib 200 mg, three 250 mg). Oropharynx was the primary site in all patients (seven p16-positive; five T1-2N2M0, four T3-4N2-3M0 [AJCC 7th edition]). All patients completed CRT within seven weeks. Grade 3 alpelisib-related toxicities occurred in four patients. No dose-limiting toxicity (DLT) was observed at 200 mg among three DLT-evaluable patients. Two of two DLT-evaluable patients treated at 250 mg experienced DLTs (inability to complete ≥75% alpelisib secondary to radiation dermatitis and febrile neutropenia). Thus, RP2D was declared at 200 mg. After median follow-up of 39.7 months, two patients developed pulmonary metastases despite locoregional control. Three-year overall survival was 77.8% (95% CI 36.5%-93.9%). Conclusion Alpelisib at 200 mg has a manageable safety profile in combination with cisplatin-based CRT in LA-SCCHN.The determinants of biodiversity patterns in the subterranean habitat called Mesovoid Shallow Substratum (MSS) are not well-understood. https://www.selleckchem.com/products/CP-690550.html In this study, thirty-three scree slopes at high altitudes were selected across the Sierra de Guadarrama National Park in central Spain to investigate the effect of ten environmental variables on the abundance and species diversity of the spider and springtail assemblages from the colluvial MSS. In each locality, a multiperforated PVC tube with a pitfall trap inside was buried up to 1 m deep, and generalized linear models and Mantel tests were used to analyze the effect of mainly climate- and habitat-related variables on the diversity patterns of both taxa. A total of 1143 individual spiders belonging to 54 species and 40 811 springtail individuals belonging to 62 species were collected. The analyses indicated that cold temperatures and the presence of forest cover on the surface significantly enhance richness and abundance in the two taxa. Environmental similarity also had a small positive effect on faunistic similarity. However, the effects of temperature and habitat detected on spider richness and abundance were stronger than on springtails, whereas the reverse was found regarding faunistic similarity. These results indicate that subterranean dwellers respond differently to the same environmental factors, which in turn, points to a different degree of affinity for the MSS. The MSS plays an important role in the survival of high mountain arthropod species, acting as a climate refuge, so the protection of this habitat should be prioritized.In vertebrates, the olfactory bulb (OB) is the zone of the brain devoted to receiving the olfactory stimuli. The size of the OB relative to the size of the brain has been positively correlated to a good olfactory capability but, recently, this correlation was questioned after new investigation techniques were developed. Among them, the isotropic fractionator allows to estimate the number of neurons and non-neurons in a given portion of nervous tissue. To date, this technique has been applied in a number of species; in particular the OB was separately analyzed in numerous mammals and in a single crocodile species. Thus, a quantitative description of the OB's cells is available for a small portion of vertebrates. Main aim of this work was to apply isotropic fractionator to investigate the olfactory capability of elasmobranch fishes, whose traditional concept of outstanding olfaction has recently been scaled down by anatomical and physiological studies. For this purpose, the OB of two elasmobranch species, Galeus melastomus and Scyliorhinus canicula, was studied leading to the determination of the number of neurons vs non-neurons in the OB of the specimens. In addition, the obtained cell quantification was related to the olfactory epithelium surface area to obtain a new parameter that encapsulates both information on the peripheral olfactory organ and the OB. The analyzed species resulted in an overall similar quantitative organization of the peripheral olfactory system; slight differences were detected possibly reflecting different environment preference and feeding strategy. Moreover, the non-neurons/neurons ratio of these species, compared to those available in the literature, seems to place elasmobranch fishes among the vertebrate species in which olfaction plays an important role.Two laboratory-scale up-flow anaerobic sludge blankets (UASB) reactors, one with and one without granular activated carbon (GAC), were operated for municipal sewage treatment at low temperatures (16.5 ± 2.0 °C). During the 120-day operation, the GAC-amended reactor significantly enhanced COD removal (from 62% to 75%, P less then 0.05) and methane production (from 87 to 218 mg CH4-COD/reactor/d) than the non-GAC reactor. Bacterial communities were significantly different between the two reactors (P less then 0.05). Geobacter, a key indicator for direct interspecies electron transfer (DIET), had the highest differential score (LEfSe analysis), showing significantly higher abundances in the GAC-amended reactor (3.7-8.8%) than the non-GAC reactor (0.9-4.0%). GAC also enriched syntrophic bacteria, Syntrophomonas, Syntrophus and sulfate reducing bacteria. Methanobacterium dominated the archaeal community in the GAC-amended reactor sludge (35.7%) and GAC-biofilm (75.3%), and was less abundant in the non-GAC reactor (9.9%). It indicates that GAC enriched microbial syntrophic partners with potential electro-activities in the anaerobic digestion process.
