Participants were 90 adolescents 12.0-17.5 years (50% female, 30% Hispanic/Latinx), at risk for adult obesity by virtue of above-average body mass index (BMI; kg/m2 ≥ 70th percentile) or parental obesity (BMI ≥ 30 kg/m2 ). ACEs were determined as presence (vs. absence) based upon the Schedule for Affective Disorders and Schizophrenia for School-Aged Children. Physiological stress response was measured as heart rate/blood pressure response to the Trier Social Stress Test. Homeostatic model assessment of insulin resistance was determined from fasting glucose/insulin. Sixty-one percent of adolescents reported positive ACE history. The presence of ACEs predicted greater heart rate (p .08). Findings suggest heightened sympathetic stress response in adolescence could be explanatory in how ACEs increase the risk for later cardiometabolic disease. Future studies should characterize ACEs in relationship to day-to-day variations in adolescents' stress physiology and glucose homeostasis.

) are the most frequently prescribed first-line systemic treatment for moderate-to-severe plaque psoriasis in Germany. Risankizumab (Skyrizi
) is a humanized IgG1 monoclonal antibody that specifically binds to the p19 subunit of interleukin-23.

This study compared risankizumab treatment with FAEs in psoriasis patients.

This phase 3, randomized, active-controlled, open-label study with blinded assessment of efficacy was conducted in Germany. Patients were randomized (11) to subcutaneous risankizumab 150mg (weeks 0, 4, 16) or oral FAEs at increasing doses from 30mg/d (week 0) up to 720mg/d (weeks 8-24). Enrolled patients were adults naive to and candidates for systemic therapy, with chronic, moderate-to-severe plaque psoriasis. Phototherapy was not allowed within 14 days before or during the study.

Key efficacy endpoints were met at week 24 for risankizumab (N=60) versus FAEs (N=60) (P<.001) achievement of PASI 90 (primary endpoint; 83.3% vs 10.0%), PASI 100 (50.0% vs 5.0%), PASI 75 (98.3% vs 33.3%), PASI 50 (100% vs 53.3%), and Static Physician's Global Assessment of clear/almost clear (93.3% vs 38.3%). The rates of gastrointestinal disorders, flushing, lymphopenia, and headache were higher in the FAEs group. One risankizumab patient reported a serious infection (influenza requiring hospitalization). There were no malignancies, tuberculosis, or opportunistic infections in either treatment arm.

The evolution of immune defense is determined by a complex interplay of costs and benefits of parasite infection and immune response, all of which may differ for male and female hosts in sexual lineages. Here, we examine fitness costs associated with an inducible immune defense in a fish-cestode host-parasite system. Cestode infection induces peritoneal fibrosis in threespine stickleback (Gasterosteus aculeatus), constraining cestode growth and sometimes encasing and killing the parasite. Surveying two wild populations of stickleback, we confirm that the presence of fibrosis scar tissue is associated with reduced parasite burden in both male and female fish. However, fibrotic fish had lower foraging success and reproductive fitness (reduced female egg production and male nesting success), indicating strong costs of the lingering immunopathology. Consistent with substantial sexually concordant fitness effects of immune response, we find alignment of multivariate selection across the sexes despite sexual antagonism over morphological shape.
