By measuring allele-specific expression in F1 hybrids from multiple crosses, we found that the majority of expression divergence between species was explained by trans-regulatory mechanisms. We also found strong evidence for two cis-regulatory alleles affecting expression divergence of two genes with putative effects on skeletal development (dync2li1 and pycr3). These results suggest that SNPs and structural variants contribute to the evolution of novel traits and highlight the utility of the San Salvador Island pupfish system as an evolutionary model for craniofacial development.
Very light daily smoking is increasingly common among young adults. Evidence suggests that levels of nicotine dependence vary significantly among young adults who engage in very light daily smoking. However, the links between dependence and clinically relevant outcomes (eg, lapse) in this population remain unclear. The goal of this study was to address this gap by evaluating how well different nicotine dependence scales predict lapse behavior among very light daily smoking young adults.
Very light daily smokers (1-5 cigarettes/day) aged 18-25 participated in an initial laboratory session, during which nicotine dependence was assessed using four commonly used measures the Fagerstrӧm Test for Cigarette Dependence (FTCD), the Hooked On Nicotine Checklist (HONC), the Transdisciplinary Tobacco Use Research Centers (TTURC) Nicotine Dependence Inventory, and the Wisconsin Inventory of Smoking Dependence Motives (WISDM). After a baseline period, eligible participants (n = 40) completed a 10-day abstinence incentink self-reported nicotine dependence with the ability to achieve and maintain abstinence among very light daily smoking young adults. 4-Methylumbelliferone solubility dmso The results may aid clinicians in selecting among variable measures of nicotine dependence when assessing and treating this population.
Intracarotid injection of mannitol has been applied for decades to support brain entry of therapeutics that otherwise do not effectively cross the blood-brain barrier (BBB). However, the elaborate and high-risk nature of this procedure has kept its use restricted to well-equipped medical centers. We are developing a more straightforward approach to safely open the BBB, based on the intra-arterial (IA) injection of NEO100, a highly purified version of the natural monoterpene perillyl alcohol.
In vitro barrier permeability with NEO100 was evaluated by transepithelial/transendothelial electrical resistance and antibody diffusion assays. Its mechanism of action was studied by western blot, microarray analysis, and electron microscopy. In mouse models, we performed ultrasound-guided intracardiac administration of NEO100, followed by intravenous application of Evan's blue, methotrexate, checkpoint-inhibitory antibodies, or chimeric antigen receptor (CAR) T cells.
NEO100 opened the BBB in a reversible and nont approach to enhance brain entry of any therapeutic.
Changes in motivational orientation across adulthood affect cognitive processes. The purpose of this research was to investigate if and how motivational incentives (gains or losses) affect prospective memory for intended actions in younger, middle-aged, and older adults.
The consequences of memory hits and misses and the framing of the memory tasks were experimentally manipulated between participants In a gain-framing condition, participants accumulated rewards, dependent on the proportion of target events to which they responded accurately. In a loss-framing condition, participants received an initial endowment from which losses were deducted, dependent on the proportion of targets they missed. We measured memory accuracy, perceived task importance, and ongoing-task performance.
Gains and losses had different effects on memory across age groups Age × Motivational Valence interactions emerged across two studies. Older adults showed relatively better memory performance to avoid losses than to achieve gains. Moreover, higher age was associated with lower memory performance (Study 1) and slower but more accurate decisions in an ongoing activity (Study 2).
The findings reveal that motivational incentives and the framing of consequences as gains or losses moderate the relation between age and memory performance. Older adults' memory performance may benefit when messages encourage the avoidance of losses. This may also help to design age-tailored interventions in applied settings (e.g., health-related behavior).

Phylogenetic models that disregard this heterogeneity are prone to systematic errors, which can lead to severe long-branch attraction artifacts. State-of-the-art models accounting for across-site compositional heterogeneity include the CAT model, which is computationally expensive, and empirical distribution mixture models estimated via maximum likelihood (C10-C60 models). Here, we present a new, scalable method EDCluster for finding empirical distribution mixture models involving a simple cluster analysis. The cluster analysis utilizes specific coordinate transformations which allow the detection of specialized amino acid distributions either from curated databases or from the alignment at hand. We apply EDCluster to the HOGENOM and HSSP databases in order to provide universal distribution mixture (UDM) models comprising up to 4,096 components. Detailed analyses of the UDM models demonstrate the removal of various long-branch attraction artifacts and improved performance compared with the C10-C60 models. Ready-to-use implementations of the UDM models are provided for three established software packages (IQ-TREE, Phylobayes, and RevBayes).
