Catalytic, enantioselective synthesis of stereogenic silicon compounds remains a challenge. Herein, we report a rhodium-catalyzed regio- and enantio-selective intermolecular hydrosilylation of alkene with prochiral dihydrosilane. This new method features a simple catalytic system, mild reaction conditions and a wide functional group tolerance.Artificial intelligence algorithms based on principles of deep learning (DL) have made a large impact on the acquisition, reconstruction, and interpretation of MRI data. Despite the large number of retrospective studies using DL, there are fewer applications of DL in the clinic on a routine basis. To address this large translational gap, we review the recent publications to determine three major use cases that DL can have in MRI, namely, that of model-free image synthesis, model-based image reconstruction, and image or pixel-level classification. For each of these three areas, we provide a framework for important considerations that consist of appropriate model training paradigms, evaluation of model robustness, downstream clinical utility, opportunities for future advances, as well recommendations for best current practices. We draw inspiration for this framework from advances in computer vision in natural imaging as well as additional healthcare fields. We further emphasize the need for reproducibility of research studies through the sharing of datasets and software. LEVEL OF EVIDENCE 5 TECHNICAL EFFICACY STAGE 2.
Measurement of greenhouse gas (GHG) concentrations and isotopic compositions in the atmosphere is a valuable tool for predicting their sources and sinks, and ultimately how they affect Earth's climate. Easy access to unmanned aerial vehicles (UAVs) has opened up new opportunities for remote gas sampling and provides logistical and economic opportunities to improve GHG measurements.
This study presents synchronized gas chromatography/isotope ratio mass spectrometry (GC/IRMS) methods for the analysis of atmospheric gas samples (20-mL glass vessels) to determine the stable isotope ratios and concentrations of CO
, CH
and N
O. To our knowledge there is no comprehensive GC/IRMS setup for successive measurement of CO
, CH
and N
O analysis meshed with a UAV-based sampling system. The systems were built using off-the-shelf instruments augmented with minor modifications.
The precision of working gas standards achieved for δ
C and δ
O values of CO
was 0.2‰ and 0.3‰, respectively. The mid-term pble isotope analysis of these three important GHGs. In contrast to previously reported findings, the presented method enables successive analysis of all three GHGs from a single ambient atmospheric gas sample.Nucleotide repeat expansions in the C9orf72 gene cause frontotemporal lobar degeneration (FTLD) and amyotrophic lateral sclerosis (ALS). Transcribed repeat RNA accumulates within RNA foci and is also translated into toxic dipeptide repeat proteins (DPR). The mechanism of repeat RNA accumulation, however, remains unclear. The RNA exosome complex is a multimeric ribonuclease involved in degradation of defective RNA. Here, we uncover the RNA exosome as a major degradation complex for pathogenic C9orf72-derived repeat RNA. Knockdown of EXOSC10, the catalytic subunit of the complex, enhanced repeat RNA and DPR protein expression levels. RNA degradation assays confirmed that EXOSC10 can degrade both sense and antisense repeats. Furthermore, EXOSC10 reduction increased RNA foci and repeat transcripts in patient-derived cells. Cells expressing toxic poly-GR or poly-PR proteins accumulate a subset of small nucleolar RNA precursors, which are physiological substrates of EXOSC10, as well as excessive repeat RNA, indicating that arginine-rich DPR proteins impair the intrinsic activity of EXOSC10. Collectively, arginine-rich DPR-mediated impairment of EXOSC10 and the RNA exosome complex compromises repeat RNA metabolism and may thus exacerbate C9orf72-FTLD/ALS pathologies in a vicious cycle.Emerging infectious diseases are major drivers of global and local amphibian biodiversity loss. Therefore, developing effective disinfection methods to manage the impact of diseases in wild and captive "ark" populations are an important goal in amphibian conservation. While chemical disinfectants have been used safely and effectively in larval and adult amphibians infected with pathogenic microbes, their applicability to amphibian egg masses has remained untested. To bridge this gap, we exposed embryos of the common toad (Bufo bufo) and agile frog (Rana dalmatina) experimentally to three widely used disinfectants voriconazole, chloramphenicol and chlorogen-sesquihydrate. For 3 days we exposed portions of egg masses to these disinfectants at 1×, 2×, 5× and 10× the concentration recommended for the disinfection of tadpoles and adults. Subsequently, we recorded embryonic and larval survival, as well as larval body mass and the incidence of abnormalities 12 days after hatching. Application of voriconazole had species- and concentration-dependent negative impacts on survival and body mass, and caused marked malformations in the viscerocranial structure of B. bufo tadpoles. Exposure to chlorogen-sesquihydrate also resulted in significant mortality in B. bufo embryos and negatively affected body mass of R. dalmatina larvae. Chloramphenicol had little negative effects on embryos or larvae in either species. Based on these results, the application of voriconazole and chlorogen-sesquihydrate cannot be recommended for the disinfection of amphibian eggs, whereas treatment with chloramphenicol appears to be a safe method for eliminating potential pathogens from anuran egg masses and their immediate aquatic environment. The majority of individuals over an age of 60 have hypertension. Elevated blood pressure and older age are associated with very similar changes in brain structure and function. We review the parallel brain changes associated with increasing age and blood pressure. This review focuses on joint associations of aging and elevated blood pressure with neuropsychological function, regional cerebral blood flow responses to cognitive and metabolic challenges, white matter disruptions, grey matter volume, cortical thinning, and neurovascular coupling. Treatment of hypertension ameliorates many of these changes but fails to reverse them. Treatment of hypertension itself appears more successful with better initial brain function. LY303366 order We show evidence that sympathetic and renal influences known to increase blood pressure also impact brain integrity. Possible central mechanisms contributing to the course of hypertension and aging are then suggested. An emphasis is placed on psychologically relevant factors stress, cardiovascular reactions to stress, and diet/obesity.
