Recent evidence suggests that in addition to classical glucose transporters, sodium-glucose transporters (SGLTs) are expressed by cancers. We therefore investigated the possibility that SGLT inhibitors, which are used in treatment of type 2 diabetes, may exert antineoplastic activity by limiting glucose uptake. We show that the SGLT2 inhibitor canagliflozin inhibits proliferation of breast cancer cells. Surprisingly, the antiproliferative effects of canagliflozin are not affected by glucose availability nor by the level of expression of SGLT2. Canagliflozin reduces oxygen consumption and glutamine metabolism through the citric acid cycle. The antiproliferative effects of canagliflozin are linked to inhibition of glutamine metabolism that fuels respiration, which represents a previously unanticipated mechanism of its potential antineoplastic action.The tumor microenvironment (TME) is an important mediator of breast cancer progression. Cancer-associated fibroblasts constitute a major component of the TME and may originate from tissue-associated fibroblasts or infiltrating mesenchymal stromal cells (MSCs). The mechanisms by which cancer cells activate fibroblasts and recruit MSCs to the TME are largely unknown, but likely include deposition of a pro-tumorigenic secretome. The secreted embryonic protein NODAL is clinically associated with breast cancer stage and promotes tumor growth, metastasis, and vascularization. Herein, we show that NODAL expression correlates with the presence of activated fibroblasts in human triple-negative breast cancers and that it directly induces Cancer-associated fibroblasts phenotypes. We further show that NODAL reprograms cancer cell secretomes by simultaneously altering levels of chemokines (e.g., CXCL1), cytokines (e.g., IL-6) and growth factors (e.g., PDGFRA), leading to alterations in MSC chemotaxis. We therefore demonstrate a hitherto unappreciated mechanism underlying the dynamic regulation of the TME.Transition and lanthanide metal complexes have rich photophysical properties that can be used for cellular imaging, biosensing and phototherapy. In this brief review, we survey the recent literature on the use of luminescent metal complexes (including ReI, RuII, OsII, IrIII, PtII, EuIII and TbIII) as DNA optical probes, including examples of compounds that bind selectively to non-duplex DNA topologies such as quadruplex, i-motif and DNA mismatches. Their applications in biosensing and phototherapy are briefly mentioned in the relevant sections.Mitigating the risk of mercury (Hg) contamination in rice soils using environmental friendly amendments is essential to reducing the probable daily intake (PDI) of MeHg via rice consumption. Here, we examined the impacts of different doses (0% (control), 0.6% and 3%) of rice hull-derived biochar (RHB) and mixture of wheat-rice straw-derived biochar (RWB) on the fractionation, phytoavailability, and uptake of total (THg) and methyl Hg (MeHg) by rice in Hg-polluted soil (THg = 78.3 mg kg-1) collected from Wanshan Hg mining area. Both biochars increased rice biomass up to 119% as compared to control. Dihydroartemisinin cell line Application of RHB and RWB significantly (P ≤ 0.05) decreased bioavailable Hg (soluble and exchangeable and specifically-sorbed fractions) concentrations by 55-71% and 67-72%, respectively. The addition of RHB significantly decreased MeHg concentrations in the soil. However, RWB (particularly at 3%) increased significantly MeHg concentrations in the soil as compared to the control and RHB treatments, likely due to the increased abundance of Hg-methylation microorganisms (e.g., Geobacter spp., Nitrospira spp.) in the RWB treatments. Both RHB and RWB significantly decreased MeHg concentrations in the rice grain by 55-85%. We estimated a reduction of the PDI of MeHg from 0.26 μg kg-1 bw d-1of control to below the reference dose (0.1 μg kg-1 bw d-1) of two biochar treatments. Our results highlight the potentiality of RWB and RHB for mitigating MeHg accumulation in rice and reducing PDI of MeHg via rice consumption, which offers a sustainable approach for management of Hg-polluted soils.High levels of arsenic in drinking water and food materials continue to pose a global health challenge. Over 127 million people alone in Bangladesh (BD) and West Bengal (WB) state of India are exposed to elevated levels of arsenic in drinking water. Despite decades of research and outreach, arsenic awareness in communities continue to be low. Specifically, very few studies reported arsenic awareness among low-income farming communities. A comprehensive approach to assess arsenic awareness is a key step in identifying research and development priorities so that appropriate stakeholder engagement may be designed to tackle arsenic menace. In this study, we developed a comprehensive arsenic awareness index (CAAI) and identified key awareness drivers (KADs) of arsenic to help evaluate farmers' preferences in dealing with arsenic in the environment. The CAAI and KADs were developed using a questionnaire survey in conjunction with ten machine learning (ML) models coupled with a hybrid feature selection approach. Two questionnaire surveys comprising of 73 questions covering health, water and community, and food were conducted in arsenic-affected areas of WB and BD.
