Our previous study found that nicotinamide (NIC), the precursor of nicotinamide adenine dinucleotide (NAD), could inhibit the endothelial-mesenchymal transition and accelerate healing the wound to the corneal endothelium in the rabbit. Here we hypothesize that NIC may possess the capacity to protect the cornea from UVB-induced endothelial apoptosis. Therefore, a mouse model and a cultured cell model were used to examine the effect of NAD+ precursors, including NIC, nicotinamide mononucleotide (NMN) and NAD, on the UVB-induced apoptosis of corneal endothelial cells (CECs). The results showed that UVB irradiation caused apparent corneal edema and cell apoptosis in mice, accompanied by reduced levels of NAD+ and its key biosynthesis enzyme, nicotinamide phosphoribosyltransferase (NAMPT), in the corneal endothelium. However, the subconjunctival injection of NIC, NMN or NAD+ effectively prevented UVB-induced tissue damage and endothelial cell apoptosis in the mouse cornea. Moreover, pretreatment using NIC, NMN and NAD+ increased the survival rate and inhibited the apoptosis of cultured human CECs irradiated by UVB. Mechanistically, pretreatment using NIC recovered the AKT activation level and decreased the BAX/BCL-2 ratio. In addition, the capacity of NIC to protect CECs was fully reversed in the presence of the AKT inhibitor LY294002. Therefore, we conclude that NAD+ precursors can effectively prevent the apoptosis of the corneal endothelium through reactivating AKT signaling; this represents a potential therapeutic approach for preventing UVB-induced corneal damage.Sirtuin 6 (Sirt6) is predominantly expressed in epithelial cells in intestinal crypts. It plays an important role in protecting intestinal epithelial cells against inflammatory injury. Previously, we found that colitis is associated with the down-regulation of Sirt6 protein in the intestines. Here, we report that murine interferon γ (Ifnγ) inhibits Sirt6 protein but not mRNA expression in YAMC (a mouse colonic epithelial cell line) in a dose- and time-dependent manner. Using miRNA array analysis, we showed that Ifnγ induces expression of miR-92b in YAMC cells. Valproic acid cost With in silico analysis, we found that the Sirt6 3' UTR contains a putative binding site for miR-92b. Luciferase assay showed that Ifnγ inhibited Sirt6 3' UTR activity and this effect was mimicked by miR-92b via directly targeting the miR-92b seed site in the 3' UTR of Sirt6 mRNA. Furthermore, Western blot demonstrated that miR-92b down-regulated Sirt6 protein expression in YAMC cells. Blocking miR-92b with a specific inhibitor attenuated the inhibitory effect of Ifnγ on Sirt6 protein expression in the cells. Collectively, our data suggest that Ifnγ inhibits Sirt6 protein expression in intestinal epithelial cells via a miR-92b‒mediated mechanism. miR-92b may be a novel therapeutic target for rescuing Sirt6 protein levels in intestinal epithelial cells, thereby protecting against intestinal mucosal injury caused by inflammation.Synthetic superhydrophobic (SH) surfaces were developed after 1990s, and the number of publications in this field is around 13 500 at present. However, the industrial production of SH coatings is very unsatisfying after the intensive research activity in the last two decades. The main reason is the loss of the water repellence properties when SH surfaces are exposed to outdoor conditions due to their weak mechanical properties and contamination from the medium which removes the initial SH properties. In this Feature Article, we focus on the scientific and technical reasons which prevent the application of the SH surfaces in our daily lives by highlighting some well-known but mostly overlooked problems in this area. (The synthesis methods of SH surfaces are not the subject of this article since they were reviewed previously in very good articles.) The basic contact angle science and the issue of the cancellation of the Wenzel and Cassie-Baxter equations are reviewed in the first part. The issues of the expensive and small-scale SH surface preparation problems, the difficulties in obtaining a transparent SH surface, the troubles arising from the water vapor condensation on an SH surface, the lack of robustness and abrasion resistance of most of the SH surfaces, the drawbacks of the fabricated self-healing SH surfaces, the short useful service life of self-cleaning SH surfaces due to surface contamination, and the ineffective anti-icing SH coatings are reviewed in the following text. Some important problems affecting the unsuccessful industrial applications of the SH surfaces are discussed critically in the Conclusions and Outlook section. Finally, some proposals are presented for future directions on the synthesis and applications of SH surfaces.
