METHODS Clascoterone cream was applied twice daily for up to nine months to the face and/or trunk. Treatment Emergent Adverse Events (TEAE) and Local Skin Reactions (LSR) were evaluated at Months 1, 3, 6, and 9, and at any unscheduled visit(s). All statistical analysis was performed using SAS®, Windows Version 9.3. RESULTS There were 609 screened and enrolled subjects (n=317 clascoterone, n=292 vehicle from original studies); n=347 completed the study (n=179 clascoterone, n=168 vehicle). Overall, 110 (18.1%) subjects experienced 191 TEAEs. The most frequently reported TEAE was nasopharyngitis (n=20). Fourteen subjects had a total of 19 test article related TEAEs. Nine subjects experienced 9 TEAEs leading to discontinuation. Six subjects experienced seven serious TEAEs, none treatment related; one serious TEAE led to study discontinuation. Overall treatment emergent LSRs=18.1% (110/607). The most frequent LSRs on the face and trunk were erythema, scaling/dryness, and pruritus; most were trace/minimal or mild in severity. LIMITATIONS Long-term efficacy was not a primary endpoint CONCLUSIONS A low frequency of TEAEs over 9-months of clascoterone treatment was observed. Managing the balance between accurately identifying early-stage melanomas while avoiding biopsies of benign lesions (i.e. over-biopsy) is the major challenge of melanoma detection. Decision-making can be especially difficult in patients with extensive atypical nevi. Recognizing that the primary screening modality for melanoma is subjective examination, studies have shown a tendency towards over-biopsy. Even low-risk routine surgical procedures are associated with morbidity, mounting healthcare costs, and patient anxiety. Recent advancements in noninvasive diagnostic modalities have helped to improve diagnostic accuracy, especially when managing melanocytic lesions of uncertain diagnosis. Breakthroughs in artificial intelligence have also shown exciting potential in changing the landscape of melanoma detection. In part I of this continuing medical education article, we review novel diagnostic technologies such as automated 2D and 3D total body imaging with sequential digital dermoscopic imaging, reflectance confocal microscopy, and electrical impedance spectroscopy, and we explore the logistics and implications of potentially integrating artificial intelligence into existing melanoma management paradigms. This paper briefly describes objectives and outcome of the EuroMix project as well as recent findings from OECD, WHO and EFSA addressing combined exposure to multiple chemicals. Building on this, five steps addressing further development needs and implementation of existing tools especially for risk managers and policy makers are proposed. This study investigated the effect of two communication strategies (informational and affect-based) in reducing chemophobia, the irrational fear of chemicals. In an online experiment, participants (N = 448) were randomly assigned to one of three groups ("control", "knowledge", or "affect" group). The following dependent variables were assessed chemophobia, knowledge of basic toxicological principles, affect towards chemicals, benefit perception of the use of chemicals, and preference for natural substitutes in consumer products. The results showed that only the informational approach, which conveys knowledge of basic toxicological principles, significantly decreased chemophobia and the preference for natural substitutes in consumer products. The affect-based approach significantly increased positive affect towards chemicals and the benefit perception of their use, but did not decrease chemophobia. This suggested that the provision of relevant information about basic toxicological principles is a more effective strategy than merely addressing laypeople's affect towards chemicals to reduce chemophobia. Relevant knowledge could be taught in schools or disseminated by toxicologists and scientists who are trusted by the public. The toxic effect of zearalenone (ZEA) is not fully understood and there is an urgent need for the development of effective agents to protect against the toxic effects of ZEA. In this study, we detected whether curcumin (CUR) can reduce Leydig cells apoptosis induced by ZEA. The Effects of ZEA and CUR on cell viability was evaluated using a Cell Counting kit-8 assay (CCK-8). Apoptosis was detected by terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay and flow cytometry. The protective effect of CUR on oxidative stress induced by ZEA was determined by ROS, MDA, T-SOD, GSH and GSH-Px levels. In addition, we also determined effects of key signaling pathways and proteins involved in the apoptotic, PI3K-AKT, Nrf2 and endoplasmic reticulum stress signaling pathways by Western blotting. The expressions of proteins (PTEN, AKT, p-AKT, Bax, Bcl-2, GRP78, CHOP, JNK, P-JNK, Caspase-12, Caspase-9, Caspase-3, Nrf2, Keap1 and HO-1) were measured. The experimental results showed that CUR can alleviate oxidative stress and apoptosis caused by ZEA through the PI3K-AKT, Nrf2 and endoplasmic reticulum stress signaling pathways.
