Since their discovery at the beginning of this century, riboswitches have been regarded as potential antibacterial targets. Using fragment screening, high-throughput screening and rational ligand design guided by X-ray crystallography, lead compounds against various riboswitches have been identified. Here, we review the current status and suitability of the thiamine pyrophosphate (TPP), flavin mononucleotide (FMN), glmS, guanine, and other riboswitches as antibacterial targets and discuss them in a biological context. Further, we highlight challenges in riboswitch drug discovery and emphasis the need to develop riboswitch specific high-throughput screening methods.The focus of this paper is on examining the mechanical behavior of spark plasma sintered WC-Co based composites doped with Cr3C2, TaC-NbC, TiC, and VC, as well as defining some parameters characterizing deformation and fracture processes during hardness measurement. The calculated microhardness of WC-Co cemented carbides for all the studied compositions is found to be higher than the results obtained during hardness testing. Therefore, the ratio of the experimental and calculated values of microhardness is shown to be an approximate indication of WC-Co cemented carbide sensitivity to damage processes during indentation. Some parameters characterizing the microstructure-microhardness relationship are defined, and the nanomechanical properties of WC-Co cemented carbide phases are examined in order to separate the deformation and fracture processes during the indentation process. Strain gradient linear function parameters are calculated for 10-cycle nanoindentation. It was found that the nanoindentation curve after 10 cycles shows anomalous behavior of the WC grains, which indicates their fracture processes.A simple, mild and efficient method to prepare HSi- or HOSi-telechelic, high-molecular-weight polydimethylsiloxane polymers (to 41,600 g·mol-1) using the one-shot hydrolysis of MHMH is reported; titration of the water allowed for higher molecular weights (to 153,900 g·mol-1). The "living" character of the chain extension processes was demonstrated by adding a small portion of MHMH and B(C6F5)3 (BCF) to a first formed polymer, which led to a ~2-fold, second growth in molecular weight. The heterogeneous reaction reached completion in less than 30 min, much less in some cases, regardless of whether it was performed neat or 50 wt% in dry toluene; homogeneous reactions in toluene were much slower. The process does not involve traditional redistribution, as judged by the low quantities ( less then 3%) of D4 produced. However, it is not possible to avoid Chojnowski metathesis from MHDDMH giving D3, which occurs competitively with chain extension.The global obesity epidemic continues to rise worldwide. In this context, unraveling new interconnections between biological systems involved in obesity etiology is highly relevant. Dysregulation of the endocannabinoidome (eCBome) is associated with metabolic complications in obesity. This study aims at deciphering new associations between circulating endogenous bioactive lipids belonging to the eCBome and metabolic parameters in a population of overweight or obese individuals with metabolic syndrome. To this aim, we combined different multivariate exploratory analysis methods canonical correlation analysis and principal component analysis, revealed associations between eCBome subsets, and metabolic parameters such as leptin, lipopolysaccharide-binding protein, and non-esterified fatty acids (NEFA). Subsequent construction of predictive regression models according to the linear combination of selected endocannabinoids demonstrates good prediction performance for NEFA. Descriptive approaches reveal the importance of specific circulating endocannabinoids and key related congeners to explain variance in the metabolic parameters in our cohort. Analysis of quartiles confirmed that these bioactive lipids were significantly higher in individuals characterized by important levels for aforementioned metabolic variables. In conclusion, by proposing a methodology for the exploration of large-scale data, our study offers additional evidence of the existence of an interplay between eCBome related-entities and metabolic parameters known to be altered in obesity.Nowadays, there are many commercial products from natural resources on the market, but they still have many additives to increase their biological activities. On the other hand, there is particular interest in natural sources that would have antimicrobial properties themselves and would inhibit the growth and the reproduction of opportunistic microorganisms. Therefore, a comparative antimicrobial study of natural samples of aloe and its commercial products was performed. Qualitative and quantitative determination of antimicrobial efficiency of Aloe arborescens and Aloe barbadensis and its commercial products on fungi, Gram-negative, and Gram-positive bacteria was performed. Samples exhibited antimicrobial activity and slowed down the growth of all tested microorganisms. Research has shown that natural juices and gels of A. arborescens and A. barbadensis are at higher added concentrations comparable to commercial aloe products, especially against microbial cultures of Bacillus cereus, Candida albicans, and Pseudomonas aeruginosa, whose growths were completely inhibited at a microbial concentration of 600 μg/mL.
