Unfortunately, current instrumental methods to evaluate muscle hardness are expensive, time-consuming, and wasteful. This study sought to develop classification models for differentiating the muscle hardness of crisp grass carp on the basis of blood analysis. Out of the total 264 grass carp samples, 12 outliers from crisp grass carp group were removed based on muscle hardness ( less then 9 N), and the remaining 252 samples were used for the analysis of seven blood indexes including hydrogen peroxide (H2O2), glucose 6-phosphate dehydrogenase (G6PD), malondialdehyde (MDA), glutathione (GSH/GSSH), red blood cells (RBC), platelet count (PLT), and lymphocytes (LY). Furthermore, six machine learning models were applied to predict the muscle hardness of grass carp based on the training (152) and testing (100) datasets obtained from the blood analysis random forest (RF), naïve Bayes (NB), gradient boosting decision tree (GBDT), support vector machine (SVM), partial least squares regression (PLSR), and artificial neural network (ANN). The RF model exhibited the best prediction performance with a classification accuracy of 100%, specificity of 93.08%, and sensitivity of 100% for discriminating crisp grass carp muscle hardness, followed by the NB model (93.75% accuracy, 91.83% specificity, and 94% sensitivity), whereas the ANN model had the lowest prediction performance (85.42% accuracy, 81.05% specificity, and 85% sensitivity). These machine learning methods provided objective, cheap, fast, and reliable classification for in vivo crisp grass carp and also prove useful for muscle quality evaluation of other freshwater fish.Previous studies have reported that pitchers with glenohumeral internal rotation deficit (GIRD) may increase the risk of shoulder injury. However, limited information is available regarding the specific effects of GIRD in baseball pitching. The purpose of this study was to investigate whether baseball pitchers with GIRD change their pitching mechanism. Fifteen baseball pitchers with GIRD and 15 pitchers without GIRD were recruited from university or senior high-school teams. A three-dimensional motion analysis system (Eagle System, Motion Analysis Corporation, Santa Rosa, CA, USA) was used to capture the pitching motion while performing fastball pitches. The kinematics and kinetics of the throwing shoulder and trunk were analyzed based on motion captured data. The Mann-Whitney U test was used to test the differences of the analyzed parameters between two groups. At the instant of ball release, the GIRD group showed lower shoulder external rotation and trunk rotation, and larger shoulder horizontal adduction. In addition, the GIRD group exhibited a significantly larger shoulder inferior force in the cocking and acceleration phase, and a significantly larger internal rotation torque in the acceleration phase. The present results suggested that pitchers with GIRD need stretch training to enlarge joint range of motion, and to improve trunk strength and flexibility to alleviate potential problems associated with pitching in GIRD pitchers.Ras is the most frequently mutated oncogene and recent drug development efforts have spurred significant new research interest. Here we review progress toward understanding how Ras functions in nanoscale, proteo-lipid signaling complexes on the plasma membrane, called nanoclusters. We discuss how G-domain reorientation is plausibly linked to Ras-nanoclustering and -dimerization. We then look at how these mechanistic features could cooperate in the engagement and activation of RAF by Ras. Moreover, we show how this structural information can be integrated with microscopy data that provide nanoscale resolution in cell biological experiments. Synthesizing the available data, we propose to distinguish between two types of Ras nanoclusters, an active, immobile RAF-dependent type and an inactive/neutral membrane anchor-dependent. We conclude that it is possible that Ras reorientation enables dynamic Ras dimerization while the whole Ras/RAF complex transits into an active state. These transient di/oligomer interfaces of Ras may be amenable to pharmacological intervention. We close by highlighting a number of open questions including whether all effectors form active nanoclusters and whether there is an isoform specific composition of Ras nanocluster.Nucleotide-based intergenomic similarities are useful to understand how viruses are related with each other and to classify them. Here we have developed VIRIDIC, which implements the traditional algorithm used by the International Committee on Taxonomy of Viruses (ICTV), Bacterial and Archaeal Viruses Subcommittee, to calculate virus intergenomic similarities. When compared with other software, VIRIDIC gave the best agreement with the traditional algorithm, which is based on the percent identity between two genomes determined by BLASTN. Furthermore, VIRIDIC proved best at estimating the relatedness between more distantly-related phages, relatedness that other tools can significantly overestimate. In addition to the intergenomic similarities, VIRIDIC also calculates three indicators of the alignment ability to capture the relatedness between viruses the aligned fractions for each genome in a pair and the length ratio between the two genomes. The main output of VIRIDIC is a heatmap integrating the intergenomic similarity values with information regarding the genome lengths and the aligned genome fraction.
