Electrophoretic NMR has the capacity to provide unique physico-chemical information but is limited by a variety of experimental artifacts, such as thermal convection and electrolytic products in the sample. Here we present some simple modifications to the experimental hardware and protocol that, in a significant number of cases, can much improve experimental accuracy and precision. We show that one can strongly reduce artifacts in a symmetric sample cell with an appropriate feeding of current and with a porous plug suitably inserted. This latter feature requires that the electric field pulses across the sensitive volume are implemented as current-controlled pulses applied to the sample. Measurements with current-controlled pulses have the additional advantage of not requiring calibration with samples of known electrophoretic mobility.
To present a case series of women with borderline ovarian tumours (BOTs) who underwent oocyte vitrification in addition to fertility-sparing surgery.

Observational study of all women referred to a French fertility preservation unit between 2015 and 2019 for counselling regarding a fertility preservation (FP) strategy after BOT fertility-sparing surgery. All eligible women underwent one or more cycles of controlled ovarian stimulation (COS) using an antagonist protocol, followed by oocyte retrieval. Metaphase II (MII) oocytes were vitrified.

Twenty-five women with BOTs were referred during the study period. Among them, 11 women underwent at least one cycle of COS. One hundred and seven MII oocytes were vitrified. The mean number of vitrified MII oocytes per woman was 9.7 (standard deviation 5.2). Five live births were reported during follow-up of four women with vitrified oocytes three spontaneous pregnancies, one in-vitro fertilization cycle with fresh embryo transfer, and one live birth after return ofe available short-term data are reassuring, further long-term studies evaluating the safety and cost-effectiveness of this systematic FP strategy after BOT fertility-sparing surgery are required.
Severe hypoxia exists in placentas during early pregnancy, with reoxygenation during mid-gestation. Hypoxia-inducible factor-1α (Hif1α), an oxygen sensor, initiates placental vascular development. We have shown that the placental vasculature in Hmox1-deficient (Hmox1
, Het) pregnancies is impaired, with morphological defects similar to Hif1α-deficient placentas.

Whole wild-type (WT) and Het mouse placentas were collected at E8.5 (1%-3% O
) and E9.5-15.5 (8%-10% O
). mRNA levels were determined using real-time RT-PCR or PCR arrays and protein levels using Western blot. Bone marrow-derived macrophages (BMDMs) from WT, Het, and Hmox1 knockout (KO) mice, representing different Hmox1 cellular levels, were generated to study the role of Hmox1 on Hif1α 's response to hypoxia-reoxygenation and gestational age-specific placental lysates.

Hif1α was expressed in WT and Het placentas throughout gestation, with protein levels peaking at E8.5 and mRNA levels significantly upregulated from E9.5-E13.5, but significa caused by any genetic defect or environmental influence in early pregnancy could be the root cause of pregnancy disorders.
To investigate associations between placental volume (PV) at 11 weeks' gestation and offspring bone outcomes at birth, 6 years and 8 years.

3D ultrasound scanning was used to assess 11weekPV in a subset (n=236) of the Southampton Women's Survey (a prospective mother-offspring cohort). Maternal anthropometric measures and lifestyle information were obtained pre-pregnancy and at 11 weeks' gestation. Offspring dual-energy x-ray absorptiometry scanning was performed within 2 weeks postnatally and at 6 and 8 years. Linear regression was used to assess associations between PV and bone outcomes, adjusting for offspring age at DXA and sex, and maternal age, height, smoking status, walking speed and triceps skinfold thickness. β are SD change in bone outcome per SD change in PV.

In adjusted models, 11weekPV was positively associated with bone area (BA) at all time points, with evidence of persisting associations with increasing childhood age (birth n=80, β=0.23 [95%CI=0.03, 0.42], 6 years n=110, β=0.17 [-0.01, 0.36], 8 years n=85, β=0.13 [-0.09, 0.36]). Similar associations between 11weekPV and bone mineral content (BMC) were observed. Associations with size-corrected bone mineral content were weaker at birth but strengthened in later childhood (birth n=78, β=0.07 [-0.21, 0.35], 6 years n=107, β=0.13 [-0.08, 0.34], 8 years n=71, β=0.19 [-0.05, 0.43]).

11weekPV is associated with DXA bone measures at birth, with evidence of persisting associations into later childhood. Further work is required to elucidate the contributions of placental morphology and function to gestational influences on skeletal development.

In this study, preliminary results in vitro showed that Fr-75 eluted from RA extract could not only completely inhibit the tracheal ring contraction raised by KCl in 20 min, but also effectively affect the tracheal ring contraction induced by KCl-, Ach- and His in a concentration-dependent manner (3.91-250 μg/mL). Further results on cells exhibited that Fr-75 could decrease the concentration of intracellular Ca2+ as well. selleckchem These results revealed the underlying mechanism in vitro that the inhibition of tracheal ring contraction might be due to the decline of the intracellular Ca2+ concentration, which caused by suppressing calcium channel, antagonizing the muscarinic and histamine receptors. Also, results in vivo exhibited that Fr-75 could distinctly ease the symptoms of ovalbumin-sensitized mice, including relieving the pathological injury, increasing the latency to preconvulsive dyspnea and to enhanced pause, reducing the inflammatory cells, chemokines and cytokines in BALF and lung tissue.
