Background The tilapia aquaculture industry is facing heavy economic losses due to Streptococcus agalactiae (S. agalactiae) infections. While progress has been made in past years, the lack of a high-quality tilapia genome and transcript annotations makes systematic and comprehensive exploration for a non-coding RNA regulatory network associated with the infection process unfeasible, and it stunts further research focused on disease defense and treatment. Herein, single molecular real time sequencing (SMRT-Seq) and RNA-seq data were utilized to generate a high-quality transcript annotation. In addition, Changes in mRNA and non-coding RNA expression were also analyzed during a S. agalactiae infection in tilapia. Findings In total, 16.79 Gb of clean data were obtained by sequencing on six SMRT cells, with 712,294 inserts (326,645 full-length non-chimeric reads and 354,188 non-full-length reads). A total of 197,952 consensus transcripts were obtained. Additionally, 55,857 transcript sequences were acquired, with pendent characteristics. Finally, lncRNA-mRNA (trans and cis), lncRNA-miRNA-mRNA, and circRNA-miRNA-mRNA regulatory networks were constructed and revealed that non-coding RNA is involved in the regulation of immune-related genes. Conclusions This study generated a greatly-improved transcript annotation for tilapia using long-read PacBio sequencing technology, and revealed the presence of a regulatory network comprised of non-coding RNAs in Nile tilapia infected with S. agalactiae.This study aimed to identify major phytochemical constituents, as well as compare the immunomodulatory effects of Psidium guajava L. and Phyllanthus amarus Schun and Thonn crude ethanol extracts and their fractions on striped catfish (Pangasianodon hypophthalmus) head kidney leukocytes (HKLs). Methods We contacted all PBS patients 14 years-old or older treated and followed at our institution. Age at orchiopexy, type of orchiopexy (with or without ligation of gonadal vessels), testicular volumes and positions were evaluated. Pelvic MRI (to assess prostate size, seminal vesicles and vas) and hormonal profile were ordered. Sperm analysis and analysis of urine after masturbation were performed after informed consent. Results Fifteen patients were included in this study. Mean age was 19.2 years. Mean age at orchiopexy was 18 months. Fourteen patients (93.3%) had normal and orthotopic testes. Mean testicular volume was 6.9 cc. Eight patients collected semen, five of them (62.5%) had spermatozoa in the specimen and motile sperm was found in 4 (50%). Our study highlights a high prevalence of prostate and seminal vesicle abnormalities that may represent an important cause for their infertility.Objective To investigate abnormal intrinsic connectivity of striatum in lifelong premature ejaculation (PE) patients compared with healthy controls (HCs). Methods Forty-seven lifelong PE patients and 30 healthy controls were enrolled in the present study and underwent resting-state functional magnetic resonance imaging. The functional connectivity (FC) analysis and 2-sample t tests were applied to investigate the alterations of striatum-related connectivity in patients compared with HCs (significant threshold at P less then .05, false discovery rate corrected), and during which Fisher's r-to-z transformation was adopted and the resulting z values were used as the statistical FC values. Correlation analysis was performed to test possible relationships between the imaging findings and clinical characteristics in the patient group (P less then .05, Bonferroni correction). Results The results showed that compared with HCs, lifelong PE patients had significantly decreased FC between the right caudate and insula, superior temporal pole (STP) and orbitofrontal cortex (OFC) as well as decreased FC between the bilateral putamen and insula, STP and middle cingulate cortex (MCC). Meanwhile, patients had significantly increased FC between the left caudate and OFC, and increased FC between the right putamen and fusiform. The mean FC value from the caudate-OFC, caudate-insula, and caudate-STP connectivity negatively correlated with the Premature Ejaculation Diagnostic Tool score, separately. Conclusion The current study showed the functional abnormality of lifelong PE in multiple brain regions implicated in sensation, motivation, and ejaculation-related inhibitory control, which may improve our understanding of the abnormal striatum-related neural mechanisms in lifelong PE patients.
