Here, we determined the role of the mitochondrial pyruvate carrier (MPC) in the occurrence of this metabolic inflexibility in wild-type (WT) and MPC1-deficient (MPC1def) flies that were exposed to diets with different sucrose concentrations for 15-25 days (Standard Diet SD, Medium-Sucrose Diet MSD, and High-Sucrose Diet HSD). Our results showed that MPC1def flies had lower mitochondrial respiration rates than WT flies on the SD and MSD. However, when exposed to the HSD, WT flies displayed decreased mitochondrial respiration rates compared to MPC1def flies. WT flies exposed to the HSD also displayed increased proline contribution and slightly decreased MPC1 expression. Surprisingly, when fed the MSD and the HSD, few metabolites were altered in WT flies whereas MPC1def flies display significant accumulation of glycogen, glucose, fructose, lactate, and glycerol. Overall, this suggests that metabolic inflexibility starts to occur in WT flies after 15-25 days of exposure to the HSD whereas the MPC1def flies display metabolic inflexibility independently of the diet provided. This study thus highlights the involvement of MPC as an essential protein in Drosophila to maintain proper metabolic homeostasis during changes in dietary resources.Lechwe are specialised wetland antelope that can have a strict social hierarchy or perform lekking during breeding. The southern lechwe (Kobus leche) and the Nile lechwe (K. megaceros) are both found in zoos globally, but little research is available to support husbandry decisions. The aim of this research was to investigate current housing and husbandry used for these lechwe across North American and European zoos. A survey was distributed to holders in 2018 and information on 33 herds (18 Nile and 15 southern) was collected. The survey focussed on population demographics, enclosure size, biologically relevant exhibit features, mixed-species holdings, nutrition, use of environmental enrichment and performance of abnormal repetitive behaviours. Results showed that lechwe were housed in herds with similar sex ratios to wild counterparts but with a potential lack of opportunity to lek. Many zoos provided wetland, but this was rarely actively managed, and not all zoos provided cover for hiding and retreat. Current feeding practice differed significantly compared to available antelope husbandry guidelines. No consistency in amounts of pellet, forage or produce provided to lechwe across institutions was found. Abnormal repetitive behaviour was noted by several zoos, but no significant predictor of such behaviour could be identified. Despite some identifiable recognition of ecology informing lechwe management, it is important that evidence-based husbandry decisions are made based on a species' evolutionary pathway and ecological needs and some fundamental features of lechwe husbandry do not always correlate with the adaptive traits of a specialised wetland ungulate.This study evaluates whether serum phospholipids fatty acids (PL-FAs) and markers of their endogenous metabolism are associated with breast cancer (BC) subtypes. EpiGEICAM is a Spanish multicenter matched case-control study. A lifestyle and food frequency questionnaire was completed by 1017 BC cases and healthy women pairs. Serum PL-FA percentages were measured by gas chromatography-mass spectrometry. Conditional and multinomial logistic regression models were used to quantify the association of PL-FA tertiles with BC risk, overall and by pathological subtype (luminal, HER2+ and triple negative). Stratified analyses by body mass index and menopausal status were also performed. Serum PL-FAs were measured in 795 (78%) pairs. Women with high serum levels of stearic acid (odds ratio (OR)T3vsT1 = 0.44; 95% confidence interval (CI) = 0.30-0.66), linoleic acid (ORT3vsT1 = 0.66; 95% CI = 0.49-0.90) and arachidonic to dihomo-γ-linolenic acid ratio (OR T3vsT1 = 0.64; 95% CI = 0.48-0.84) presented lower BC risk. Participants with high concentrations of palmitoleic acid (ORT3vsT1 = 1.65; 95% CI = 1.20-2.26), trans-ruminant palmitelaidic acid (ORT3vsT1 = 1.51; 95% CI = 1.12-2.02), trans-industrial elaidic acid (ORT3vsT1 = 1.52; 95% CI = 1.14-2.03), and high oleic to stearic acid ratio (ORT3vsT1 = 2.04; 95% CI = 1.45-2.87) showed higher risk. These associations were similar in all BC pathological subtypes. Our results emphasize the importance of analyzing fatty acids individually, as well as the desaturase activity indices.
