The size distribution graphs showed a decreasing trend in the mean diameter of composite nanofibers with the increase in GO-COOH content, from 141.40 nm for CS/PG 0.1% to 119.88 nm for CS/PG 0.5%. The dispersion of GO-COOH led to composite scaffolds with increased elasticity; the Young's modulus of CS/PG 0.5% (84 ± 4.71 MPa) was 7.5-fold lower as compared to CS/PEO (662 ± 15.18 MPa, p less then 0.0001). Contact angle measurements showed that both GO-COOH content and crosslinking step influenced the surface wettability of scaffolds, leading to materials with ~1.25-fold higher hydrophobicity. The in vitro cytocompatibility assessment showed that the designed nanofibrous scaffolds showed a reasonable cellular proliferation level after 72 h of contact with the fibroblast cells.Recently, promising results of the antitumor effects were observed in patients with metastatic castration-resistant prostate cancer treated with 177Lu-labeled PSMA-ligands. Radionuclide therapy efficacy may even be improved by using the alpha emitter Ac-225. Higher efficacy is claimed due to high linear energy transfer specifically towards PSMA positive cells, causing more double-strand breaks. This study aims to manufacture [225Ac]Ac-PSMA-I&T according to good manufacturing practice guidelines for the translation of [225Ac]Ac-PSMA-I&T into a clinical phase 1 dose escalation study. Quencher addition during labeling was investigated. Quality control of [225Ac]Ac-PSMA-I&T was based on measurement of Fr-221 (218 keV), in equilibrium with Ac-225 in approximately six half-lives of Fr-221 (T½ = 4.8 min). Radio-(i)TLC methods were utilized for identification of the different radiochemical forms, gamma counter for concentration determination, and HPGe-detector for the detection of the radiochemical yield. Radiochemical purity was determined by HPLC. The final patient dose was prepared and diluted with an optimized concentration of quenchers as during labeling, with an activity of 8-12 MBq (±5%), pH > 5.5, 100 ± 20 μg/dose, PSMA-I&T, radiochemical yield >95%, radiochemical purity >90% (up to 3 h), endotoxin levels of less then 5 EU/mL, osmolarity of 2100 mOsmol, and is produced according to current guidelines. The start of the phase I dose escalation study is planned in the near future.A central characterization of retinal immunobiology is the prevention of proinflammatory activity by macrophages. The retinal pigment epithelial cells (RPEs) are a major source of soluble anti-inflammatory factors. This includes a soluble factor that induces macrophage apoptosis when the activity of the immunomodulating neuropeptide alpha-melanocyte-stimulating hormone (α-MSH) is neutralized. In this manuscript, isolated extracellular soluble membranes (ESMs) from primary RPE were assayed to see if they could be the soluble mediator of apoptosis. Inhibitor Library Our results demonstrated that RPE ESMs mediated the induction of macrophage apoptosis that was suppressed by α-MSH. In contrast, the RPE line ARPE-19, cultured under conditions that induce similar anti-inflammatory activity to primary RPEs, did not activate apoptosis in the macrophages. Moreover, only the ESMs from primary RPE cultures, and not those from the ARPE-19 cell cultures, expressed mFasL. The results demonstrate that RPE ESMs are a soluble mediator of apoptosis and that this may be a mechanism by which the RPEs select for the survival of α-MSH-induced suppressor cells. In Ethiopia, land, water, energy and food (LWEF) nexus resources are under pressure due to population growth, urbanization and unplanned consumption. The effect of this pressure has been a widely discussed topic in nexus resource literature. The evidence shows the predominantly negative impact of this; however, the impact of these factors is less explored from a local scale. As a result, securing nexus resources is becoming a serious challenge for the country. This necessitates the identification of the driving factors for the sustainable utilization of scarce LWEF nexus resources. Our study provides a systemic look at the driving factor indicators that induce nexus resource degradation. We use the Analytical Hierarchical Process (AHP) to develop the indicators' weights, and use a Path Analysis Model (PAM) to quantitatively estimate the effect of the driving factor indicators on the LWEF nexus resources. The results indicate that social (48%), economic (19%), and policy and institutional changes (14%) are theotection of LWEF nexus resources, which have close links with human health and the environment. In order to alleviate the adverse effects of driving factors, all stakeholders need to show permanent individual and collective commitment. Furthermore, we underline the necessity of applying LWEF nexus approaches to the management of these drivers, and to optimize the environmental and social outcomes.A framework for optimal sensor placement (OSP) for virtual sensing using the modal expansion technique and taking into account uncertainties is presented based on information and utility theory. The framework is developed to handle virtual sensing under output-only vibration measurements. The OSP maximizes a utility function that quantifies the expected information gained from the data for reducing the uncertainty of quantities of interest (QoI) predicted at the virtual sensing locations. The utility function is extended to make the OSP design robust to uncertainties in structural model and modeling error parameters, resulting in a multidimensional integral of the expected information gain over all possible values of the uncertain parameters and weighted by their assigned probability distributions. Approximate methods are used to compute the multidimensional integral and solve the optimization problem that arises. The Gaussian nature of the response QoI is exploited to derive useful and informative analytical expressions for the utility function. A thorough study of the effect of model, prediction and measurement errors and their uncertainties, as well as the prior uncertainties in the modal coordinates on the selection of the optimal sensor configuration is presented, highlighting the importance of accounting for robustness to errors and other uncertainties.
