This study investigated the effects of minocycline microinjections, into the midbrain periaqueductal gray (PAG), on morphine withdrawal and the expression of pannexin-1 (panx1), phosphorylated mammalian target of rapamycin (p-mTOR), protein kinase A (PKA), and cAMP response element-binding protein (CREB). Rats were injected with morphine, intraperitoneally, at increasing doses, twice per day, to establish animal models of morphine exposure. Minocycline was administered into the PAG before the first intraperitoneal (i.p.) injection of morphine each day, on days 1-4. On the last day of the experiment, all rats were injected with naloxone, and morphine withdrawal was observed, and then changes in the expression levels of ionized calcium-binding adaptor molecule 1 (Iba1) and its downstream factors, panx1, p-mTOR, PKA, and CREB were evaluated by western blot and immunohistochemistry analyses. Morphine withdrawal increased microglial activation, whereas minocycline could inhibit microglial activation and withdrawal and the downregulation of panx1, p-mTOR, PKA, and CREB expression, reducing the effects of morphine withdrawal.
Extremes in water availability, either exceptionally wet or dry conditions, can damage crops and may detrimentally affect the livelihood and well-being of people engaged in agriculture. We estimated the effect of water availability on suicide in rural India, a context where the majority of households are dependent upon agriculture.
We used data from a nationally representative sample of 8.5 million people who were monitored for causes of death from 2001 to 2013. Water availability was measured with high-resolution precipitation and temperature data (i.e., the Standardized Precipitation Evapotranspiration Index). We used a fixed effects approach that modeled changes in water availability within districts (n=569) over time (n=13 years) to estimate the impact on suicide deaths. We restricted our analysis to rural areas and to deaths occurring during the growing season (June-March) among adults aged 15 or older, and controlled for sex, age, region, and year. RAD1901 We used Poisson regression with standard errors clustered at the district level and total deaths as the offset.
There were 9456 suicides and 249,786 total deaths in our study population between 2001 and 2013. Compared to normal growing seasons, the percent of deaths due to suicide increased by 18.7% during extremely wet growing seasons (95% CI 6.2, 31.2) and by 3.6% during extremely dry growing seasons (95% CI -17.9, 25.0). We found that effects varied by age.

Abnormally wet growing conditions may play an important, yet overlooked, role in suicide among rural Indian adults.Cyanobacterial harmful algal blooms (Cyano-HABs) in water resources involving algal species such as Microcystis aeruginosa have become a serious environmental issue due to their severely negative effects. In the present study, an adsorption-based strategy was employed to control M. aeruginosa, with industrial waste-derived Escherichia coli biomass valorized to produce polyethylenimine-modified polyacrylonitrile-E. coli biomass composite fiber (PEI-PANBF). PEI-PANBF removed approximately 80% of M. aeruginosa cells from an aqueous solution without causing any cell damage. Interestingly, the thickness of PEI-PANBF had a strong influence on the efficiency of M. aeruginosa cell removal. In addition, PEI-PANBF simultaneously removed M. aeruginosa cells and their toxic secondary metabolite, microcystin-LR, from aqueous media. Thus, our proposed fiber represents a feasible utilization method of industrial waste biomass as a biosorbent for the control of Cyano-HABs.The risk levels of heat-related extreme events need to be estimated for prediction and real-time monitoring to mitigate their impacts on air quality, public health, the ecosystem, and critical infrastructure. Many countries have adopted meteorological variable base thresholds for assessing the risk level of heat-related extreme events. These thresholds provide an approximate risk level for a specific event but do not consider its intensity and duration in the risk assessment. The current study provides a statistical tool to assess the risk of heat-related extreme events while concurrently considering their intensities and durations based on the wet-bulb globe temperature (WBGT). To this end, the intensity-duration-frequency (IDF) relationship of the extreme WBGT in South Korea was derived. Regional frequency analysis was employed to understand the IDF relationship. Return levels of heat-related extreme events in South Korea were calculated and their characteristics were investigated based on the annual maximuh Korea might become more severe in the future.
Neoadjuvant chemotherapy has become a powerful tool to convert borderline resectable (BRPC) and locally advanced pancreatic cancers (LAPC) into a resectable scenario. However, data analyzing the optimal type of therapy are scarce. In the present multicenter retrospective study, we evaluated the influence of FOLFIRINOX (FFX) and gemcitabine (GEM)-based neoadjuvant therapy on patient prognosis.
Data on 239 patients from 7 centers across Europe was gathered using an online database. Patients having received their first cycle of chemotherapy for BRPC/LAPC before 06/2017, with a minimum follow-up of 12 months, were included in the intention-to-treat analysis.
Patients treated with neoadjuvant FFX (n=135) or gemcitabine+nab-paclitaxel (GNP) (n=38) had significantly improved radiological response according to RECIST criteria as compared to single-agent GEM (n=16), with a partial/complete response of 59.3%, 55.3% and 6.25% respectively (p=0.001). Treatment with FFX (n=135) and GNP (n=38) resulted in higher resection rates compared to GEM (73.3%, 81.6% and 43.8%; p=0.01 and p=0.005). Regardless of regimen, patients who were resected had significantly prolonged overall survival compared to non-resected patients (p<0.01). Complete pathological responses (ypT0 ypN0) were predominantly observed with FFX (p=0.01). Adjuvant GNP in addition to successful neoadjuvant therapy and surgery resulted in a trend towards improved median survival as compared to postoperative observation (47.0 vs. 30.1 months, p=0.06).
Representing one of the largest studies published so far, our results reveal that patients with BRPC/LAPC should be offered either FFX or GNP to improve chances of resection and with this also survival.
