Specific taste receptors that are expressed in the lower gastrointestinal tract may respond to alteration in gut microbiota composition, detecting bacterial molecules, and regulate immune responses. Given the importance of brain-gut-microbiota axis and gene-environment interactions in PD, we investigate the associations between the genetic variants of TAS2R38 and gut microbiota composition in 39 PD patients. The results confirm that the majority of PD patients have reduced sensitivity to 6-n-propylthiouracil (PROP) and are carriers of at least one non-functional TAS2R38 AVI haplotype. Moreover, we found this correlation to be associated with a reduction in bacteria alpha-diversity with a predominant reduction of Clostridium genus. We hypothesised that the high frequency of the non-taster form of TAS2R38 associated with a diminuition of Clostridium bacteria in PD might determine a reduction in the activation of protective signalling-molecules useful in preserving gut homeostasis. This pilot study, by identifying a decrease in specific bacteria associated with a reduced sensitivity to PROP, adds essential information that opens new avenues of research into the association of PD microbiota composition and sensory modification.Along with advancements in both protein and chemistry science, the chemical modification of proteins is attracting more and more attention. More specifically, the attachment of polymers or reactive moieties into collagen offers a method to add novel functions to this protein. However, the fibrillogenesis of the modified collagen with high grafting density cannot always be achieved. Here, inspired by the hybrid fibrils of xenogeneic collagen, fibrillogenesis of acrylic acid-grafted-collagen (AAc-g-Col) without self-assembly property was achieved by the induction of natural collagen (Col). The step-by-step co-assembly process of AAc-g-Col and Col was confirmed by turbidity assay. The formation of Col/AAc-g-Col hybrid fibrils was verified by TEM since the acryloyl groups of the hybrid fibrils were labelled using HS-AuNPs based on the Michael addition. Moreover, rheology, SEM, and MTT assays revealed that the fibrillary structures and biocompatibility of the Col/AAc-g-Col hydrogel were comparable to that of the Col hydrogel, although they presented a lower viscoelasticity.To maintain the quality of postharvest fruits continuously and meet the health requirements of consumers, a high barrier and long-lasting antibacterial polylactic acid film as packaging material was developed in this study. Polylactic acid-based antibacterial films incorporated with Cinnamaldehyde inclusions were used to achieve long-lasting antibacterial activity and improve the barrier properties. Cinnamaldehyde inclusions were prepared via the inclusion method and used as a sustained-release antibacterial agent and reinforcement to be incorporated into polylactic acid-based films within a concentration range of 0-30 wt%. The FT-IR spectrum demonstrated that the Cinnamaldehyde inclusions was physically interacting with PLA. The XRD results showed that the cinnamaldehyde inclusions at a concentration of 10 wt% enhanced the crystallinity of the antibacterial film. The oxygen and water vapor barrier properties of the film were respectively 14.29% and 12.38% higher than those of a pure PLA film. The tensile strength of the antibacterial film increased by 20%. And the antibacterial activity against Escherichia coli and Listeria monocytogenes was 100%. The release rate of cinnamaldehyde of the antibacterial film was slow and varied smoothly for 20 d.A new amphiphilic pullulan derivative (DBAP-PO) was obtained by grafting tertiary butyl amine and octanoyl groups on the pullulan backbone as cationic and hydrophobic moieties, respectively. The structural characteristics of the modified polymer were investigated by FT-IR and 1H and 13C NMR spectroscopy. The self-association ability in aqueous solution of DBAP-PO was studied by viscosity and fluorescence methods. The intrinsic viscosity of the polymer was determined by Wolf model. The critical aggregation concentration (CAC) value of 0.028 g/dL, determined by fluorescence measurements in the presence of pyrene, was confirmed by capillary viscosimetry and dynamic laser scattering (DLS). Dialysis method was used to demonstrate the capacity of the pullulan derivative to form spherical nanoparticles (d ~ 200 nm) loaded with model drug, sodium diclofenac (DF) (74% entrapment efficiency). The DF release was sustained and pH-dependent. In vitro cytotoxicity as well as morphological studies conducted on the human skin fibroblasts showed that DBAP-PO/DF nanoparticles do not exhibit cytotoxic effects at the pharmacologically relevant concentration of DF, maintaining the typical morphology of the cells.In this study, a phosphorylated derivative of Cyclocarya paliurus polysaccharide (CP) was synthesized using sodium tripolyphosphate and sodium trimetaphosphate. Antioxidant activities and cytoprotective effects of unmodified polysaccharide and phosphorylated derivatives were investigated employing various in vitro systems. Results showed that phosphorylated modification caused a significant change in chemical composition and the apparent structure of CP. Phosphorylated Cyclocarya paliurus polysaccharide (P-CP) showed excellent scavenging capacities against DPPH and hydroxyl radical in a dose-dependent manner. P-CP provided a more significant protective effect against hydrogen peroxide-induced oxidative stress in RAW264.7 cells by a compositive oxidation defense mechanism than CP. P-CP could reduce oxidative stress by significantly enhancing the contents of antioxidant enzyme superoxide dismutase (SOD) and decreasing malondialdehyde (MDA) content in oxidative damaged cells, in addition to scavenging ROS directly and regulating cell cycle distribution, reducing apoptosis capacity and improving cell viability, consequently achieving the intracellular antioxidant activity.
