Background Secondary metabolites of plants, the poly phenols, play vital role in protection from many health problems in human beings. Structurally favored phytochemicals may be studied to protect multi organ injury. At pharmacological doses, gallic acid is nontoxic to mammals and is generally absorbed in the intestine. Aims In this present study, gallic acid was evaluated for its protective efficacy against lipo polysaccharide (LPS) and dgalactosamine (D-GalN) induced multiorgan injury, i.e., liver, kidney and brain. Methods Three different doses of gallic acid (5, 10 and 20 mg/kg p.o.) were administered to the experimental animals for 6 consecutive days followed by exposure to LPS (50 µg/kg I.P.) and D-GalN (300 mg/kg I.P.) on 6th day. Results Exposure to LPS and D-GalN resulted in increased oxidative stress and proinflammatory cytokines. Altered hematology and serology due to LPS and D-GalN were restored towards control by gallic acid. Declined antioxidants such as reduced glutathione, superoxide dismutase and catalase due to injurious effects of LPS and D-GalN were rejuvenated by gallic acid. Exposure to LPS and D-GalN severely increased lipid peroxidation, CYP2E1 activity and tissue lipids while lowered protein content. Gallic acid restored all these parameters towards control in dose dependent manner and 20 mg/kg dose provided the best protection. Histological study showed improved histoarchitecture of liver, kidney and brain that supported biochemical endpoints. Conclusion Gallic acid minimized oxidative stress and provided best protection at 20 mg/kg dose against LPS and D-GalN induced multi organ acute injury.Aim and Objectvie Intake of Stachys sieboldii MIQ. has been associated with relieving inflammation and maintaining optimal gut health function. We investigated the diversity and composition of microflora in feces of S. sieboldii MIQ.-fed mice. In addition, we evaluated the production of major cytokines (Interleukin-6 and -10) related with inflammation and fatty acid composition of several tissues. Materials and methods 16S ribosomal DNA sequencing-based microbiome taxonomic profiling analysis was performed using EzBioCloud data base. The total RNA from mesenteric lymph node was isolated and then synthesized with prime script 1st strand cDNA synthesis Kit. Quantitative real-time PCR was performed on cDNA samples using the SYBR™ Green PCR Master Mix. Results Mice fed on S. sieboldii MIQ. showed significantly reduced counts of aerobic and coliform in the feces compared with control. 16S rDNA sequencing analysis of fecal samples showed that supplementation with S. sieboldii MIQ. increased beneficial intestinal mi decreased harmful microflora. Moreover, lower mRNA expression of pro-inflammatory cytokine IL-6, and antiinflammatory cytokine IL-10 in mesenteric lymph node of supplemented mice might be associated with the lower abundances of harmful fecal microflora. This work is based on various bio-reduction of graphene oxide into reduced graphene oxide and their applications in organic synthesis and group transformations. Graphene oxide, with abundant oxygencontaining functional groups on its basal plane, provides potential advantages, including excellent dispersibility in solvents and the good heterogeneous catalyst. This manuscript reviews various methods of synthesis of graphene and graphene oxide and a comparative study on their advantages and disadvantages, how to overcome disadvantages and covers extensive relevant literature review. In the last few years, investigation based on replacing the chemical reduction methods by some bio-compatible, chemical/impurity-free rGO including flash photo reductions, hydrothermal dehydration, solvothermal reduction, electrochemical approach, microwave-assisted reductions, light and radiation-induced reductions has been reported. Particularly, plant extracts have been applied significantly as an efficient reducing agent due to toupling reaction and a wide number of multicomponent reactions have been highlighted. Finally, the aim of this study is to provide an outlook on future trends and perspectives for graphene-based materials in metal-free carbo-catalysis in green synthesis of various pharmaceutically important moieties. Background Ghrelin, a unique 28 amino acid peptide hormone secreted by the gastric X/A like cells, is an endogenous ligand of the growth hormone secretagogue receptor (GHSR). learn more Ghrelin-GHSR signaling has been found to exert various physiological functions including stimulation of appetite, regulation of body weight, lipid and glucose metabolism, and increase of gut motility and secretion. This system is thus critical for energy homeostasis. Objective The objective of this review is to highlight the strategies of ghrelin-GHSR based intervention for therapy of obesity and its related metabolic diseases. Results Therapeutic strategies of metabolic disorders targeting the ghrelin-GHSR pathway involve neutralization of circulating ghrelin by antibodies and RNA spiegelmers, antagonism of ghrelin receptor by its antagonists and inverse agonists, inhibition of ghrelin O-acyltransferase (GOAT), as well as potential pharmacological approach to decrease ghrelin synthesis and secretion. Conclusion Various compounds targeting the ghrelin-GHSR system have shown promising efficacy for intervention of obesity and relevant metabolic disorders in animals and in vitro. Further clinical trials to validate their efficacy in human being are urgently needed. Background Cerebellar and brainstem symptoms are common in early stages of multiple sclerosis (MS) yet their prognostic values remain unclear. Objective The aim of this study was to investigate long-term disability outcomes in patients with early cerebellar and brainstem symptoms. Methods This study used data from MSBase registry. Patients with early cerebellar/brainstem presentations were identified as those with cerebellar/brainstem relapse(s) or functional system score ⩾ 2 in the initial 2 years. Early pyramidal presentation was chosen as a comparator. Andersen-Gill models were used to compare cumulative hazards of (1) disability progression events and (2) relapses between patients with and without early cerebellar/brainstem symptoms. Mixed effect models were used to estimate the associations between early cerebellar/brainstem presentations and expanded disability status scale (EDSS) scores. Results The study cohort consisted of 10,513 eligible patients, including 2723 and 3915 patients with early cerebellar and brainstem symptoms, respectively.
