This experiment was performed to evaluate the effect of vitamin C and methionine hydroxy analog (MHA) on performance, blood parameters, liver enzymes, activity of selected hormones, antioxidant activity of blood plasma, intestine morphology, and HSP70 gene expression in broilers under heat stress. Four hundred 1-day-old male chickens of commercial Ross 308 strain were used in a completely randomized design with 4 treatments and 5 replicates of 20 chickens each. The experimental treatments were as follows (1) base diet based on maize soybean (the control), (2) base diet with 0.2% vitamin C, (3) base diet with 0.46, 0.36, and, 0.32% methionine hydroxy analog, in the starter, grower, and finisher periods, respectively, and (4) base diet with a mixture of 0.2% vitamin C and 0.46, 0.36, and 0.32% of methionine hydroxy analog in the starter, grower, and finisher periods respectively. Although numerically higher than control, the effect of the experimental diets on performance was not significant across the experimental period (P>0.05). Glucose and LDL concentrations under all experimental treatments were significantly reduced compared to the control (P less then 0.05). The concentrations of liver enzymes (AST and ALT) and antioxidant activity (SOD, GPx, and MDA) of broilers were significantly different from the control under combined MHA and vitamin C supplementation (P less then 0.05). All experimental diets led to significantly increased T4 concentrations, compared to control (P less then 0.05). The length and width of intestinal villi of broilers were significantly increased in all of the experimental treatments compared to the control (P less then 0.05). The difference in HSP70 gene expression was significantly reduced in all of the treatments compared to the control (P less then 0.05). The results of this experiment showed that the use of vitamin C and methionine hydroxy analog (MHA) can be effective in reducing the negative effects of heat stress.In order to reduce the thermal stress in high chromium cast iron (HCCI) matrix composites reinforced by zirconia toughened alumina (ZTA) ceramic particles, finite element simulation is performed to optimize the geometric configuration of ceramics perform. The previous model simplifies the overall structure of the ceramic particle preform and adds boundary conditions to simulate the particles, which will cause uncontrollable error in the results. In this work, the equivalent grain models are used to describe the actual preform, making the simulation results closer to the actual experimental results. The solidification process of composite material is simulated, and the infiltration between molten iron and ceramic particles is realized. Thermal stress in solidification process and compression stress distribution are obtained. The results show that adding 10-mm round holes on the preform can improve the performance of the composite, which is helpful to prevent the cracks and increases the plasticity of the material.Pharmaceutical consumption is increasing worldwide as it is essential to treat and prevent health issues but they end up in the environment. However, in many African countries like Ethiopia, the status of these compounds in various environmental samples is not very well known. In this study, a simple method for the extraction and determination of thirteen pharmaceutical compounds of different therapeutic classes in water samples using solid-phase extraction and HPLC-DAD was developed. Different parameters affecting extraction were optimised and obtained as hydrophilic-lipophilic balance (HLB) extraction cartridge, water sample pH of 5, elution solvent of 2% formic acid in water with methanol (2080%, v/v), a sample volume of 150 mL and addition of 0.5% w/v EDTA in the sample. The limits of detection and quantification of the optimised method were in the range of 0.1-0.8 µg/L and 0.2-2.6 µg/L, respectively. The relative recovery in the spiked environmental water sample was in the range of 70-117% except for amoxicillin and acetylsalicylic acid in influent wastewater. The precision for all ranged from 0.3 to 11%. The proposed method was successfully tested for the detection and quantification of different environmental water samples collected from Addis Ababa, Ethiopia. Trimethoprim, caffeine and albendazole concentrations of 7.8 (1.1), 3.2 (0.4) and 2.1 (0.1) µg/L were quantified in hospital wastewater, respectively. The concentration of norfloxacin was found to be below the limit of quantification in the same water. Trimethoprim and ciprofloxacin were also found in the sewage treatment plant influent sample at a concentration of 0.5 (0.02) and 0.3 (0.01) µg/L, respectively.Hypochlorous acid (HOCl) is a strong oxidant produced by myeloperoxidase. Previous work suggested that HOCl modifies the corrin ring of cobalamins to yield chlorinated species via mechanisms that are incompletely understood. Herein, we report a mechanistic study on the reaction between cyanocobalamin (CNCbl, vitamin B12) and HOCl. Under weakly acidic, neutral and weakly alkaline conditions, the reaction produces the c-lactone derivative of CNCbl chlorinated at the C10-position of corrin ring (C10-Cl-CNCbl-c-lactone). Formation of C10-Cl-CNCbl-c-lactone was not observed at pH ≥ 9.9. Binding and reactivity studies of C10-Cl-CNCbl and C10-Cl-CNCbl-c-lactone with relevant proteins of the cobalamin pathway and with cultured cells are necessary to elucidate the potential physiological effects of these species.
