Background Traumatic brain injuries represent a significant cause of morbidity and mortality worldwide and road traffic crashes account for a significant proportion of these injuries. It is one of the leading causes of death, especially among young adults, and, according to the World Health Organization, this will surpass many diseases as the major cause of death and disability by the year 2020 and lifelong disability is common in those who survive. It is also known as the silent epidemic. Many CT scan scoring systems for brain injury have been developed but none of them are validated. These scores are based on structural findings of CT scan to predict the prognosis. Marshall and Rotterdam are the two most widely used scoring systems. Method This was an observational study with prospectively collected data.923consecutive patients with TBI this study aimed to compare the Helsinki CT scoring system with the Rotterdam scoring system to find out the better score for the prognostic purpose by using the Glasgow outcome score. Results Helsinki CT scoring system was found to be the most accurate score among the others in predicting the outcome of the patient based on morphological findings in CT scan. These scores also guided us regarding the treatment plan and patients with the good prognostic outcome are usually treated aggressively. Also these scores guide clinicians whether to opt for a conservative approach or any intervention. Conclusion In this study we compared, Rotterdam vs. Helsinki CT Scoring System. We observed that the Helsinki score was better outcome predictors for TBI than the Rotterdam score and can be used as a single validated score in predicting outcome and guiding the treatment.Serotonin (5-HT) receptors have been shown to homodimerize and heterodimerize with other G protein-coupled receptors (GPCRs), although the details of this process have not yet been elucidated. Here we use coarse-grained molecular dynamics on monomeric 5-HT2C receptors to predict the transmembrane (TM) helices involved in such associations. All these simulations were carried out both in 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC) lipid bilayers and in mixed composition POPC-Cholesterol ones, to show whether the presence of cholesterol could directly influence and drive the dimeric association. The goal is to get insights on the self-assembly pathway leading to GPCRs 5-HT2C oligomerization, which is supposed to be the basis of its constitutional activity. From the analysis of the molecular dynamics trajectories, we observed the formation of 5-HT2C oligomers through self-assembly and we identified the main domains involved in the receptor dimerization. In particular, dimers and oligomers from the two different environments show TM4-TM5 and TM1-TM7-H8 as the preferential dimerization interfaces. Nevertheless, substantial differences arise for oligomers in POPC and in POPC-Chol membranes in POPC-Chol the variability of dimers interfaces is strictly limited to the TM1-TM7-H8 and TM4-TM5 interfaces and the dimorphism depends on cholesterol that directly participates in its formation. The aim was to assess whether stimulation of salivation affects the physiological flow of cortisol during a stressing day as compared to an ordinary day. The second aim was to show how the normalising factor affects the outcome of the study. Methods Stimulated saliva was taken from 42 children at 800 a.m. and 1200 a.m. on two separate days one month apart. During the first day, the children were exposed to stress situation, while the second day was considered a control day. The concentration of cortisol was analysed using ELISA. Results The highest level of cortisol was observed in the morning of the stress day (p 0.99). Conclusion Based on our results, the examination of the cortisol diurnal rhythm is not reliable in stimulated saliva. Moreover, the effect of saliva stimulation has to be taken into account for every marker individually (Fig. 2, Ref. Selleckchem A939572 22).
