ARIA landmarks, an important aspect of ARIA, lets screen reader users access different sections of the webpage quickly, by enabling them to skip over blocks of irrelevant or redundant content. However, these landmarks are sporadically and inconsistently used by web developers, and in many cases, even absent in numerous web pages. Therefore, we propose SaIL, a scalable approach that automatically detects the important sections of a web page, and then injects ARIA landmarks into the corresponding HTML markup to facilitate quick access to these sections. The central concept underlying SaIL is visual saliency, which is determined using a state-of-the-art deep learning model that was trained on gaze-tracking data collected from sighted users in the context of web browsing. We present the findings of a pilot study that demonstrated the potential of SaIL in reducing both the time and effort spent in navigating webpages with screen readers.COVID-19 challenged higher education to rapidly shift to remote course delivery. This study surveyed community college students (N = 356) about their confidence in completing learning related tasks before and after the shift, access to technologies used in in remote learning, and disruptions that impacted their learning. Results indicated notable declines in confidence across all demographics with significant changes in those age 18-21and for those without prior online course experience. Technology use for remote courses was primarily laptops and smartphones. Students reported the most significant changes to work-life balance came through employment changes and mental health issues. Instructional changes were both positive and negative in workload organization, course delivery, communication and technology. Institutions can use this study's findings to enact contingency planning, expand online and blended course options, refine academic and social support, and allocate resources to mental health.Demineralized bone matrix (DBM), a potential alternative to autologous bone-graft, has been increasingly used for clinical bone repair; however, its application in larger defects isn't successful partly due to the rapid dispersion of DBM particles and relatively lower osteoinductivity. Here, a novel strategy is created to complement the osteoinductivity of DBM by incorporating DBM in biopolymer hydrogel combined with the abrogation of BMP antagonism. Combined treatment of DBM + noggin-suppression displays increased osteogenic potency of human bone marrow mesenchymal stem cells (hBMSCs) in vitro. Injectable chitosan (MeGC)-based hydrogel with heparinization (Hep-MeGC) is further developed to localize and stabilize DBM. Noggin-suppression reveals the significant increase in osteogenesis of hBMSCs in the photopolymerizable Hep-MeGC hydrogels with the encapsulation of DBM. Moreover, the combination of DBM + noggin-suppression in the injectable Hep-MeGC hydrogel displays a robust bone healing in mouse critical-sized calvarial defects in vivo. The mechanistic analysis demonstrates that noggin-suppression increased DBM osteoinductivity by stimulating endogenous BMP/Smad signals. These results have shown promise in DBM's ability as a prominent bone grafting material while being coupled with gene editing mechanism and a localizing three-dimensional scaffold. Together, this approach poses a significant increase in the efficiency of DBM-mediated craniofacial bone repair and dental osteointegration.Previous studies showed that human cell line HEK293 lacking mitochondrial superoxide dismutase (MnSOD) exhibited decreased succinate dehydrogenase (SDH) activity, and mice lacking MnSOD displayed significant reductions in SDH and aconitase activities. Since MnSOD has significant effects on SDH activity, and succinate is a key regulator of TET enzymes needed for proper differentiation, we hypothesized that SOD2 loss would lead to succinate accumulation, inhibition of TET activity, and impaired erythroid precursor differentiation. To test this hypothesis, we genetically disrupted the SOD2 gene using the CRISPR/Cas9 genetic strategy in a human erythroleukemia cell line (HEL 92.1.7) capable of induced differentiation toward an erythroid phenotype. Cells obtained in this manner displayed significant inhibition of SDH activity and ~10-fold increases in cellular succinate levels compared to their parent cell controls. Furthermore, SOD2-/- cells exhibited significantly reduced TET enzyme activity concomitant with decreases in genomic 5-hmC and corresponding increases in 5-mC.
