This study establishes a cell line from prostate tumour tissue derived from the Pten-/-/trp53-/- mouse, termed DVL3 which when subcutaneously implanted in immunocompetent C57BL/6 mice, forms tumours with distinct glandular morphology, strong cytokeratin 8 and androgen receptor expression, recapitulating high-risk localised human PCa. Compared to the commonly used TRAMP C1 model, generated with SV40 large T-antigen, DVL3 tumours are immunologically cold, with a lower proportion of CD8+ T-cells, and high proportion of immunosuppressive myeloid derived suppressor cells (MDSCs), thus resembling high-risk PCa. Furthermore, DVL3 tumours are responsive to fractionated RT, a standard treatment for localised and metastatic PCa, compared to the TRAMP C1 model. RNA-sequencing of irradiated DVL3 tumours identified upregulation of type-1 interferon and STING pathways, as well as transcripts associated with MDSCs. Upregulation of STING expression in tumour epithelium and the recruitment of MDSCs following irradiation was confirmed by immunohistochemistry. The DVL3 syngeneic model represents substantial progress in preclinical PCa modelling, displaying pathological, micro-environmental and treatment responses observed in molecular high-risk disease. Our study supports using this model for development and validation of treatments targeting PCa, especially novel immune therapeutic agents.Roots typically grow downward into the soil where they anchor the plant and take up water and nutrients necessary for plant growth and development. While the primary roots usually grow vertically downward, laterals often follow a gravity set point angle that allows them to explore the surrounding environment. These responses can be modified by developmental and environmental cues. This review discusses the molecular mechanisms that govern root gravitropism in flowering plant roots. In this system, the primary site of gravity sensing within the root cap is physically separated from the site of curvature response at the elongation zone. Gravity sensing involves the sedimentation of starch-filled plastids (statoliths) within the columella cells of the root cap (the statocytes), which triggers a relocalization of plasma membrane-associated PIN auxin efflux facilitators to the lower side of the cell. This process is associated with the recruitment of RLD regulators of vesicular trafficking to the lower membrane by LAZY proteins. PIN relocalization leads to the formation of a lateral gradient of auxin across the root cap. Upon transmission to the elongation zone, this auxin gradient triggers a downward curvature. We review the molecular mechanisms that control this process in primary roots and discuss recent insights into the regulation of oblique growth in lateral roots and its impact on root-system architecture, soil exploration and plant adaptation to stressful environments.Communication protocols are evolving continuously as the interfacing and interoperability requirements are the foundation of Industry 4.0 and Industrial Internet of Things (IIoT), and the Open Platform Communication Unified Architecture (OPC UA) protocol is a major enabling technology. OPC UA was adopted by the industry, and research is continuously carried out to extend and to improve its capabilities, to fulfil the growing requirements of specific industries and hierarchical levels. Consistent issues that have to be approached are related to the latest specifications and the real-time context that could extend the applicability of the protocol and bring significant benefits in terms of speed, data volumes, footprint, security. The real-time context is essential in the automotive sector and it is highly developed within some specific protocols. The current work approaches first the conceptual analysis to improve the OPC UA interfacing using the Publish-Subscribe mechanism, focusing on real-time constraints and role distribution between entities, and considering some well-founded interfacing strategies from the automotive sector. The conceptual analysis is materialized into a solution that takes OPC UA Publish-Subscribe over User Datagram Protocol (UDP) mechanism to the next level by developing a synchronization algorithm and a multithreading broker application to obtain real time responsiveness and increased efficiency by lowering the publisher and the subscriber footprint and computational effort, reducing the difficulty of sending larger volumes of data for various subscribers and the charge on the network and services in terms of polling and filtering. The proof of concept is evaluated and the results prove the efficiency of the approach and the solution.
