A boosted regression tree model was developed to predict pH conditions in three dimensions throughout the glacial aquifer system of the contiguous United States using pH measurements in samples from 18,386 wells and predictor variables that represent aspects of the hydrogeologic setting. Model results indicate that the carbonate content of soils and aquifer materials strongly controls pH and, when coupled with long flowpaths, results in the most alkaline conditions. Conversely, in areas where glacial sediments are thin and carbonate-poor, pH conditions remain acidic. At depths typical of drinking-water supplies, predicted pH >7.5-which is associated with arsenic mobilization-occurs more frequently than predicted pH less then 6-which is associated with water corrosivity and the mobilization of other trace elements. A novel aspect of this model was the inclusion of numerically based estimates of groundwater flow characteristics (age and flowpath length) as predictor variables. The sensitivity of pH predictions to these variables was consistent with hydrologic understanding of groundwater flow systems and the geochemical evolution of groundwater quality. The model was not developed to provide precise estimates of pH at any given location. Additionally, NLRP6 may regulate extra-intestinal diseases. In this review, we summarize current knowledge on the NLRP6 inflammasome and its activation and regulation patterns, as well as its effector functions contributing to disease modulation. We discuss current challenges in NLRP6 research and future prospects in harnessing its function into potential human interventions.Fault zones are an important control on fluid flow, affecting groundwater supply, contaminant migration, and carbon storage. However, most models of fault seal do not consider fault zone cementation, despite the recognition that it is common and can dramatically reduce permeability. In order to study the field-scale hydrogeologic effects of fault zone cementation, we conducted a series of aquifer pumping tests in wells installed within tens of meters of the variably cemented Loma Blanca Fault, a normal fault in the Rio Grande Rift. In the southern half of the study area, the fault zone is cemented by calcite; the cemented zone is 2-8 m wide. In the center of the study area, the cemented fault zone is truncated at a buttress unconformity that laterally separates hydrostratigraphic units with a ∼40X difference in permeability. The fault zone north of the unconformity is not cemented. Constant rate pumping tests indicate that where the fault is cemented, it is a barrier to groundwater flow. This is an important demonstration that a fault with no clay in its core and similar sediment on both sides can be a barrier to groundwater flow by virtue of its cementation; most conceptual models for the hydrogeology of faults would predict that it would not be a barrier to groundwater flow. Additionally, the lateral permeability heterogeneity across the unconformity imposes another important control on the local flow field. PROMISE-2 (NCT02974153) was a double-blind, randomized, placebo-controlled, phase 3 study that comprised a screening visit, a 28-day pretreatment period, and a 32-week study duration. Patients in this exploratory analysis of a prespecified subgroup had confirmed diagnoses of both CM and MOH at screening. Patients were randomly assigned to receive intravenous eptinezumab 100, 300mg, or placebo every 12weeks. Efficacy outcomes included mean changes from baseline in monthly migraine days (MMDs) during weeks 1-12, migraine responder rates at week 12, and percentages of patients below International Classification of Headache Disorders thresholds for CM and MOH over weeks 1-24.
