Lotus extract-treated rats showed significantly reduced wake time and increased sleep time in a caffeine-induced model of arousal. The transcription level of GABA
receptor, GABA
receptor, and serotonin receptor tended to increase with dose, and lotus extract showed a strong dose-dependent binding capacity to the GABA
receptor.
The above results strongly suggest that GABA contained in lotus seed extract acts as a sleep potentiating compound, and that sleep-potentiating activity involves GABA
receptor binding.

Alstonia scholaris (L.) R. Br. (Apocynaceae) is a Dai folk medicine for the treatment of lung diseases in China.
The present study investigated the anti-pulmonary fibrosis effects of total alkaloids (TA) and the potential active ingredients and its possible mechanism.
After intratracheal instillation of bleomycin (BLM, 5mg/kg), mice were divided into ten groups, and orally treated with the corresponding samples once daily for 28 days. The effect of indole alkaloids was determined through analysis of cytokines, as well as histopathological examinations and gene expressions.
Severe lung fibrosis was observed in the BLM-treated mice on day 28. However, the administration of TA significantly ameliorated the pathological changes in the lungs, decreased the content of Krebs von den Lungen-6, lactate dehydrogenase, transforming growth factor-β (TGF-β), hydroxyproline, type I collagen, and malonaldehyde, and enhanced the activity of superoxide dismutase in the serum and lung tissues. In addition, the enhanced TGF-β and matrix metalloproteinase-1 (MMP-1) expressions in BLM-induced mice were obviously weakened by indole alkaloids, as well as the ratio of matrix metalloproteinase-1 to tissue inhibitor of metalloproteinase-1 was decreased. Moreover, picrinine and scholaricine yielded markedly better values in the aforementioned indices than those in other samples, indicating that they may be the active ingredients of alkaloids.
TA exerted protective effects against BLM-induced pulmonary fibrosis by reducing collagen deposition through TGF-β/MMP-1 pathway.

The aim of this study was to characterize age-related deficits of mice using different behavioral endpoints, with a focus on executive function and performance heterogeneity.
2month-old and 18month-old C57BL/6J mice were tested in the novelty-based spatial preference Y-maze test and in sequential tasks in the Morris water maze test (reference memory, reversal learning and working memory), before being evaluated for motor skills in the activity meter and accelerating rotarod tests.
Aged mice displayed an almost normal acquisition in the water maze test, however, difficulties were observed in ability to perform reversal learning and working memory tasks. A marked heterogeneity characterized the performances of aged mice in both Morris water maze and Y-maze tests. Good and poor performers were observed in aged mice although the number of these mice varied depending on the cognitive parameter considered.
Aged mice display deficits in executive function and working memory, with varying severity between inds and use of good performers as controls is suggested so that all cohorts of aged mice show similar physical and motor characteristics.Lipids are one of the main components of the organic matter present in the effluents of the food-processing industry. These waste streams can be biotransformed into valuable triacylglycerides (TAGs) and polyhydroxyalkanoates (PHAs), precursors of biofuels and biomaterials alternative to petroleum-based products. These compounds are yielded by mixed microbial cultures, and considering that both TAG and PHA accumulators may coexist within the community, it seems crucial to define those operational strategies that might control the selection of the dominant metabolic pathways (TAG or PHA accumulation). In this work, residual fish-canning oil was used as a carbon source in a two-stage process (culture selection and intracellular compounds accumulation) in which the substrate was simultaneously hydrolyzed in these two stages without the need for a previous fermentation unit. It was pretended to maximize preferential TAG or PHA storage in the accumulation reactor by the imposition of certain selective pressures in the enrichment one. Uncoupling C and N feedings and limiting nitrogen availability in the medium, allowed to maximize PHA production (82.3 wt% of PHAs, 0.80 CmmolPHA/CmmolS). learn more Besides, when low pH in the famine phase was considered as additional selective pressure, it was possible to shift the ratio TAGPHA from 496 obtaining 43.0 wt% of TAGs (0.67 CmmolTAG/CmmolS). Therefore, this novel and simplified process demonstrated versatility and efficiency in the storage of TAGs and PHAs from a unique residual feedstock and using an open culture proving that product selection can be harnessed if choosing the right operational conditions in the enrichment stage.Whether weather plays a part in the transmissibility of the novel Coronavirus Disease-19 (COVID-19) is still not established. We tested the hypothesis that meteorological factors (air temperature, relative humidity, air pressure, wind speed and rainfall) are independently associated with transmissibility of COVID-19 quantified using the basic reproduction rate (R0). We used publicly available datasets on daily COVID-19 case counts (total n = 108,308), three-hourly meteorological data and community mobility data over a three-month period. Estimated R0 varied between 1.15 and 1.28. Mean daily air temperature (inversely), wind speed (positively) and countrywide lockdown (inversely) were significantly associated with time dependent R0, but the contribution of countrywide lockdown to variability in R0 was over three times stronger as compared to that of temperature and wind speed combined. Thus, abating temperatures and easing lockdown may concur with increased transmissibility of COVID-19 in India.
