An important factor when considering the use of interventional cone beam computed tomography (CBCT) imaging during cardiac procedures is the trade-off between imaging dose and image quality. Accordingly, Adaptive CaRdiac cOne BEAm computed Tomography (ACROBEAT) presents an alternative acquisition method, adapting the gantry velocity and projection rate of CBCT imaging systems in accordance with a patient's electrocardiogram (ECG) signal in real-time. The aim of this study was to experimentally investigate that ACROBEAT acquisitions deliver improved image quality compared to conventional cardiac CBCT imaging protocols with fewer projections acquired.

The Siemens ARTIS pheno (Siemens Healthcare, GmbH, Germany), a robotic CBCT C-arm system, was used to compare ACROBEAT with a commercially available conventional cardiac imaging protocol that utilizes multisweep retrospective ECG-gated acquisition. For ACROBEAT, real-time control of the gantry position was enabled through the Siemens Test Automation Control sy system, delivering higher quality images while reducing the number of projections acquired by 90% compared to conventional cardiac imaging protocols.

RNA-binding proteins (RBPs) play an important role in skeletal muscle development and disease by regulating RNA splicing. In myotonic dystrophy type 1 (DM1), the RBP MBNL1 (muscleblind-like) is sequestered by toxic CUG repeats, leading to missplicing of MBNL1 targets. Mounting evidence from the literature has implicated other factors in the pathogenesis of DM1. Herein we sought to evaluate the functional role of the splicing factor hnRNP L in normal and DM1 muscle cells.

Co-immunoprecipitation assays using hnRNPL and MBNL1 expression constructs and splicing profiling in normal and DM1 muscle cell lines were performed. Zebrafish morpholinos targeting hnrpl and hnrnpl2 were injected into one-cell zebrafish for developmental and muscle analysis. FL118 In human myoblasts downregulation of hnRNP L was achieved with shRNAi. Ascochlorin administration to DM1 myoblasts was performed and expression of the CUG repeats, DM1 splicing biomarkers, and hnRNP L expression levels were evaluated.

Using DM1 patient myoblast cell lines we observed the formation of abnormal hnRNP L nuclear foci within and outside the expanded CUG repeats, suggesting a role for this factor in DM1 pathology. We showed that the antiviral and antitumorigenic isoprenoid compound ascochlorin increased MBNL1 and hnRNP L expression levels. Drug treatment of DM1 muscle cells with ascochlorin partially rescued missplicing of established early biomarkers of DM1 and improved the defective myotube formation displayed by DM1 muscle cells.

Magnetic resonance imaging (MRI)-guided transperineal prostate biopsy has been practiced since the early 2000s. The technique often suffers from targeting error due to deviation of the needle as a result of physical interaction between the needle and inhomogeneous tissues. Existing needle guide devices, such as a grid template, do not allow choosing an alternative insertion path to mitigate the deviation because of their limited degree-of-freedom (DoF). This study evaluates how an angulated needle insertion path can reduce needle deviation and improve needle placement accuracy.

To explore wastage of hospital-supplied medications for patients discharged to residential care facilities (RCFs).

Telephone interviews with staff at 52 RCFs and nine community pharmacies after patients were discharged from three hospitals in metropolitan Victoria, Australia, with medication supplied in original packs.

Hospital-supplied medication was used by most RCFs, for a median of 48hours, while waiting for community pharmacies to deliver medications packed in the RCFs' preferred dose administration aid system (unit-dose or multi-dose blister packs or sachets). All RCFs reported sending unused hospital-supplied medications to their community pharmacy. Six of the nine community pharmacies (managing 83% patients) indicated they did not reuse hospital-supplied medications, with the exception of select difficult-to-source medications.

There was significant wastage of hospital-supplied discharge medications. Changes to the way hospital discharge medications are funded and quantities supplied are needed to ensure continuity of medication administration while minimising financial and environmental impacts of medication wastage.
