Twenty-three natural jamunone analogues along with a series of jamunone-based derivatives were synthesized and evaluated for their inhibitory effects against breast cancer (BC) MDA-MB-231 and MCF-7 cells. The preliminary structure-activity relationship revealed that the length of aliphatic side chain and free phenolic hydroxyl group at the scaffold played a vital role in anti-BC activities and the methyl group on chromanone affected the selectivity of molecules against MDA-MB-231 and MCF-7 cells. Among them, jamunone M (JM) was screened as the most effective anti-triple-negative breast cancer (anti-TNBC) candidate with a high selectivity against BC cells over normal human cells. Mechanistic investigations indicated that JM could induce mitochondria-mediated apoptosis and cause G0/G1 phase arrest in BC cells. Furthermore, JM significantly restrained tumor growth in MDA-MB-231 xenograft mice without apparent toxicity. Interestingly, JM could downregulate phosphatidylinositide 3-kinase (PI3K)/Akt pathway by suppressing protein-tyrosine phosphatase 1B (PTP1B) expression. These findings revealed the potential of JM as an appealing therapeutic drug candidate for TNBC.
Robotic surgical technology has been widely introduced and applied in various fields of surgery. The aim of this study was to analyze long-term oncological and urogenital functional outcomes following laparoscopic/robotic total mesorectal excision (TME) in rectal cancer surgery.
We identified studies that compared oncological and functional outcomes following laparoscopic TME (LTME) and robotic TME (RTME) for treatment of rectal cancer over the past 16 years. Data related to overall survival (OS), disease-free survival (DFS), International Prostate Symptom Score (IPSS), and International Index of Erectile Function (IIEF) were subjected to meta-analysis.
There was no difference in long-term OS and DFS in the pooled data. Compared with LTME, there were significant differences in the score of IPSS at 3, 6 and 12 months for RTME, in the pooled data for male patients. There were significant differences in IIEF score for male patients at 3 and 6 months.
Compared with LTME, RTME has better preservation of urinary and sexual functions and comparable long-term oncological outcome in rectal cancer.

Obesity is amajor health problem in Poland and around the world. Excessive gain in early childhood is an important risk factor for the development of obesity. The aim of the study was to analyze the prevalence of obesity in 2-, 4- and 6-year-old obese children.
The study group 656 overweight and obese children aged 5-18 years old. The patients' height and weight were measured, body mass index (BMI) was calculated. Overweight BMI between 85th-97th percentile and obesity BMI>97th were defined using World Health Organization. Climbazole in vitro BMI<+2 SDS as overweight, BMI ≥ +2 SDS as class I obesity, and BMI≥3 SDS as class II. Measurements from the health books of children aged 2 (n=626), 4 (n=533) and 6 (n=518) years old were analyzed.
Mean age 12.25 ±2.90 years, BMI SDS +2.54 ±0.60. There were 100 overweight (15.2%) and 556 obese (84.8%) children in the group, including 143 patients with class II obesity (21.8%). Children<10 years old comprised 28%. It was established that 36.6% of the patients were overweight or obese at the age of 2 years old. At the age of 4, the percentage was 73.9%, and at the age of 6, it was as high as 84%.

Systematic monitoring of developmental parameters in children is essential from an early age.Globally, the tenth most common cancer is the oral squamous cell carcinoma (OSCC) and the treatment strategy for improving of OSCC patients survival rate still remains a challenging one. Aberrant regulation of cell to extracellular matrix protein interactions leads to progression of human cancers. The focal adhesion kinase (FAK) and its downstream target paxillin have been implicated in cancer growth, migration, invasion and metastasis of different cancers. However, the clinical significance of FAK and paxillin in OSCC is not well characterized so far. In the present work, we showed that relative mRNA and protein expressions of FAK and paxillin are significantly higher in side population (SP) cells of OSCC cell line SCC-55. Concomitantly, the matrix metalloproteinase-11 (MMP-11) level is also significantly elevated in SP cells. The enhanced expression of paxillin is strongly correlated with increased chemoresistance, proliferation rate, migration and invasion potential of SP cells. In addition, inhibition of paxillin expression by RNAi makes SP cells more sensitive to chemotherapy drugs. Therefore, our results suggest that paxillin over expression might play a significant role in cancer progression, invasion and chemoresistance of OSCC.
Hepatocellular carcinoma (HCC) is the sixth most common cancer globally, and a major unresolved medical issue. According to guideline recommendations of European Association for the Study of the Liver in 2018 and American Society for Clinical Oncology (ASCO) Guideline, nivolumab are reasonable options for appropriate advanced stage HCC.
We searched the PubMed, Embase and CNKI (China National Knowledge Infrastructure) databases for all articles within a range of published years from 2010 to 2020 of nivolumab in advanced hepatocellular carcinoma, and carried out this meta-analysis on all published studies to estimate prognostic factors of nivolumab in advanced hepatocellular carcinoma.
Finally, 6 studies with 627 advanced hepatocellular carcinoma patients treated with nivolumab met the inclusion criteria for this study. Our results indicated that α -fetoprotein (AFP), eastern Cooperative Oncology Group (ECOG) performance status, Child-Pugh class, portal vein invasion, protein induced by vitamin K absence-II (PIVKA-II), and albumin-bilirubin (ALBI) score were prognostic factor of nivolumab in advanced hepatocellular carcinoma, but hepatitis C virus (HBV) infection, barcelona Clinic Liver Cancer (BCLC) stage, and extrahepatic metastasis were not significant prognostic factors.
