Plants of the genus Pulicaria (family Asteraceae) are widely used in central Asia and the Middle East for treatment of different human diseases. Ultra performance liquid chromatography coupled to high resolution mass spectrometry (UPLC/MS) was utilized to establish the metabolic profiles of two Pulicaria species P. crispa and P. incisa. 122 metabolites were identified including flavonoids (37), phenolic acids (22), sesquiterpenes (17), diterpenes (7), and fatty acids (27), with enrichment in methoxylated flavonoids (20), caffeoylquinic acid conjugates (14) xanthane sesquiterpenes (9) and hydroxylated fatty acids (20) in both Pulicaria species. The metabolite profile of P. incisa was characterized by the presence of tri- and tetra-methoxylated flavonoids while xanthane sesquiterpenes were the main chemical markers of P. crispa. Additionally, a novel sesquiterpene acid (dihydropulicaric acid) was annotated in both species based on its MS fragments. Antioxidant activity for P. crispa and P. incisa methanol extraco its bioactive metabolites.Leishmaniasis is a serious health problem that needs a suitable vaccine delivery system to control the disease. Cationic lipids such as 1, 2-dioleoyl-3-trimethylammonium-propane (DOTAP) have been widely used in nanoliposomes' formulation to deliver antigen and adjuvant at the same time to induce protection against Leishmaniasis. Therefore, it is necessary to accurately quantify DOTAP concentration in the formulation and biological materials. Due to the poor UV absorbance of DOTAP, the use of the conventional HPLC-UV method was impossible. Currently, an evaporative light scattering detector (ELSD) or MS/MS detector in conjunction with HPLC is used to quantify DOTAP. These methods have several disadvantages, including time- consuming during extraction procedure and decrease or/and even remove some components of the formulation. According to the advantages of the quantitative 1H Nuclear Magnetic Resonance (1H-NMR) spectroscopic method, a free extraction approach was developed to the assay of DOTAP in nanoliposomes containing Leishmania antigens. This method was carried out based on the relative ratio of signal integration of DOTAP [CH2 (CH2-CH = CH-CH2)] in δ 2 ppm to a definite amount of an internal standard called dimethyl sulfone (DMSO2). The q1H-NMR method showed good precision (intra-day RSD = 1.8 % and inter-day RSD = 2.5 %), linearity (in the ranges of 1.3-7.8 mg. mL-1 with correlation coefficients at 1), repeatability (RSD ≤ 2.39 %), and stability (RSD ≤ 2.32 %) for the quantification of the DOTAP without any extraction method. Considering all the experiments conducted in this study, NMR can be a feasible alternative to other traditional techniques for the simultaneous quantification of lipids in liposome formulations.Although cardiac pathologies are the major cause of death in the world, it remains difficult to provide a reliable diagnosis to prevent heart attacks. Rapid patient care and management in emergencies are critical to prevent dramatic consequences. Thus, relevant biomarkers such as cardiac troponin and natriuretic peptides are currently targeted by commercialized Point-Of-Care immunoassays. Key points still to be addressed concern cost, lack of standardization, and poor specificity, which could limit the reliability of the assays. Consequently, alternatives are emerging to address these issues. New probe molecules such as aptamers or molecularly imprinted polymers should allow a reduction in cost of the assays and an increase in reproducibility. In addition, the assay specificity and reliability could be improved by enabling multiplexing through the detection of several molecular targets in a single device.The knowledge about the age effects in speech acoustics is still disperse and incomplete. This study extends the analyses of the effects of age and gender on acoustics of European Portuguese (EP) oral vowels, in order to complement initial studies with limited sets of acoustic parameters, and to further investigate unclear or inconsistent results. A database of EP vowels produced by a group of 113 adults, aged between 35 and 97, was used. Duration, fundamental frequency (f0), formant frequencies (F1 to F3), and a selection of vowel space metrics (F1 and F2 range ratios, vowel articulation index [VAI] and formant centralization ratio [FCR]) were analyzed. To avoid the arguable division into age groups, the analyses considered age as a continuous variable. The most relevant age-related results included vowel duration increase in both genders; a general tendency to formant frequencies decrease for females; changes that were consistent with vowel centralization for males, confirmed by the vowel space acoustic indexes; and no evidence of F3 decrease with age, in both genders. This study has contributed to knowledge on aging speech, providing new information for an additional language. The results corroborated that acoustic characteristics of speech change with age and present different patterns between genders.
To report clinical outcomes and survival at a minimum of 10 years of a consecutive series of uncemented total hip arthroplasty (THA) with fourth-generation ceramic-on-ceramic (CoC) bearings and determine the incidence of articular noises, their risk factors, and impact on quality of life (QoL).
From a series of 456 CoC THAs, there were 25 revisions, 38 deceased, and 36 lost to follow-up. The remaining 357 hips were assessed at 11.1 ± 0.7 years (range, 10-14) using the Forgotten Joint Score, Oxford Hip Score, and a questionnaire on articular noises. Epigenetics inhibitor Kaplan-Meier survival was calculated, and multivariable analysis was performed to determine associations between articular noise and patient/surgical factors.
The 10-year survival was 96.3% for the cup, 96.5% for the stem, and 94.2% for all components. The Forgotten Joint Score was 79 ± 26 (range, 0-100), and Oxford Hip Score was 16 ± 7 (range, 12-60). The mean impact of articular noises on QoL was 1.8 ± 2.9 (range, 0-9) considered negligible in 60 hips (16%), moderate in 27 hips (7%), and severe in only 13 hips (4%). Multivariable analyses revealed that the incidence of articular noise decreased with age (odds ratio [OR], 0.94; P= .001), body mass index (OR, 0.85; P= .001), and in hips implanted with 32-mm (vs 36 mm) heads (OR, 0.18; P= .030).
Of 100 hips (28%) that reported some articular noises 10 years after CoC THA, the impact of articular noises on QoL was negligible for most patients. Although larger heads can improve mobility and reduce risks of dislocations, surgeons must be aware that increasing head size could also increase risks of articular noises, notably in young and active patients.
