PrEP stigma is a frequently reported barrier, while social disclosure of PrEP use may be an important facilitator. We explored how PrEP users managed PrEP use disclosure using a symbolic interactionist approach. We interviewed 32 participants from two PrEP demonstration projects (Be-PrEP-ared, Antwerp; AMPrEP, Amsterdam). We validated qualitative findings through Be-PrEP-ared questionnaire data. A minority of participants had received negative reactions on PrEP. The way PrEP use was disclosed was highly dependent on the social situation. In a sexual context among MSM, PrEP use was associated with condomless sex. Friends endorsed PrEP use as a healthy choice, but also related it to carelessness and promiscuity. It was seldom disclosed to colleagues and family, which is mostly related to social norms dictating when it is acceptable to talk about sex. The study findings reveal that PrEP stigma experiences were not frequent in this population, and that PrEP users actively manage disclosure of their PrEP user status. Frequent disclosure and increased use may have helped PrEP becoming normalised in these MSM communities. To increase uptake, peer communication, community activism and framing PrEP as health promotion rather than a risk-reduction intervention may be crucial.This work aimed to determine the antioxidant properties of identified hydrolyzed oat proteins and peptides, and their capacity to inhibit lipase and α-amylase. The protein hydrolysates retarded the oxidation of peanut oil by reducing peroxide values (up to 2.5-fold), relative to the control oil. Of the five tested peptides, P1 (YFDEQNEQFR), P3 (SPFWNINAH), and P4 (NINAHSVVY) significantly reduced the oxidation of linoleic acid. In the enzyme assays, P3 was the best lipase inhibitor (IC50 85.4 ± 3 µM) while P1 was the most potent inhibitor of α-amylase (IC50 37.5 ± 1.1 µM). The structure-activity relationship assessed using the CABS-dock computational model predicted that interactions between peptides and pancreatic lipase residues of Ser153 , His264 , and Asp177 were important for the inhibition. In the case of α-amylase, interactions with residues of the active sites (Asp197 , Glu233 , and Asp300 ), but not those of calcium- or chloride-binding domains, were important for the inhibition. In addition, low dose of EGCG also regulated autophagy pathway associated protein (LC3B II and Beclin) expression and this autophagy pathway was blocked by PI3K/AKT inhibitor; moreover, knockdown of ATG5 reversed EGCG-induced apoptosis in 5,637 cells, indicating that EGCG might inhibit the bladder cancer through autophagy pathway. Our findings indicated that EGCG should be considered as a novel therapy for bladder cancer treatment by regulating autophagy pathway. PRACTICAL APPLICATIONS Our research proved EGCG from green tea could be used as an effective anti-tumor ingredient by revealing another mechanism that epigallocatechin-3-Gallate inhibited bladder cancer cells via inducing autophagy-related apoptosis. And green tea could be considered as a kind of tumor-preventing beverage.The fruits and aerial parts of Sechium edule (Jacq.) Sw. (Cucurbitaceae) are a popular cook vegetable being used in different parts of the world with ethnomedicinal and pharmacological values. However, the beneficial health attributes of the tuberous roots have been less exploited. The present study aimed to determine the prebiotic potentiality of the storage carbohydrates from this part. The carbohydrate fractions were harvested by hot-water, cold-water, hot-acid, hot-alkali, and hot 80% ethanol treatments following the standard protocol. The fractions were tested for in vitro prebiotic efficacy, hypocholesterolemic and antioxidant potentials, and in vivo health attributes in Swiss albino mice. The partial characterization was performed by high-performance thin-layer chromatography (HPTLC) and spectroscopic analyses by Fourier-transform infrared spectroscopy (FT-IR) and electrospray ionization mass spectrometry (ESI-MS). The highest prebiotic index was observed in hot-water and ethanol (Et-OH) fractions witherefore, finding out some novel prebiotic from conventional food sources may provide an alternative dietary source to fulfill the consumer demand.
