Mice deficient in Gal-3 exhibit important phenotypes, but it is unclear whether these phenotypes reflect an impairment of the functions of this protein. Gal-3 plays an important role in modulating the immune response to different pathogenic microorganisms. However, the role of Gal-3 in immunity to infection is still poorly understood. Therefore, we investigated the effects of Gal-3 deletion on the expression of genes involved in the innate immune response in the lungs, spleens, and brains of Gal-3 KO mice. Gene profiling expression analysis suggested that Gal-3 deletion resulted in differentially modulated expression of the genes encoding beta-glucan, mannose and chitin-responsive pattern recognition receptors, signal transduction, inflammation, and phagocytosis. Our data thus suggest the importance of Gal-3 expression in the host innate immune system.Intrinsically disordered regions (IDRs) in proteins are characterized by their flexibilities and low complexity regions, which lack unique 3 D structures in solution. IDRs play a significant role in signaling, regulation, and binding multiple partners, including DNA, RNA, and proteins. Although various experiments have shown the role of disordered regions in binding with RNA, a detailed computational analysis is required to understand their binding and recognition mechanism. In this work, we performed molecular dynamics simulations of 10 protein-RNA complexes to understand the binding governed by intrinsically disordered regions. The simulation results show that most of the disordered regions are important for RNA-binding and have a transition from disordered-to-ordered conformation upon binding, which often contribute significantly towards the binding affinity. Interestingly, most of the disordered residues are present at the interface or located as a linker between two regions having similar movements. The DOT regions are overlaped or flanked with experimentally reported functionally important residues in the recognition of protein-RNA complexes. This study provides additional insights for understanding the role and recognition mechanism of disordered regions in protein-RNA complexes.Communicated by Ramaswamy H. Sarma.Objective Although impaired time perception is associated with significant disturbance in the everyday functioning of adult patients with Attention Deficit-Hyperactivity Disorder (ADHD), it is not very well studied. The present study aimed to evaluate both visual and auditory time reproduction (TR) by using eight time intervals in adult ADHD patients compared to healthy controls (HC).Method Wechsler Adult Intelligence Scale-R (WAIS-R), Adult Attention Deficit-Hyperactivity Disorder Self-Report Scale (ASRS), Wender Utah Rating Scale (WURS), Hyperfocusing Scale (HS), Beck Depression Inventory (BDI) and a 2-back task were administered to participants. In TR tasks, participants reproduced the same duration of the auditory/visual stimulus by pressing a key and absolute discrepancy scores (ADS) were calculated. A 2 (Group) x 2 (Task Modality) x 8 (Time Interval 500, 1000, 2000, 4000, 6000, 8000, 12000 and 16000 msec) mixed-design ANOVA was performed.Results All clinical scores of ADHD group were higher than HC (p 8000 msec were more prominent in patients. For both TR tasks, ASRS-hyperactivity/impulsivity scores were the main predictor of ADS in the linear regression analysis, while ASRS-attention deficit and HS scores were excluded.Conclusion Results of the present study indicate significant TR impairment in adult ADHD regardless of task modality (visual or auditory), depressive symptoms and working memory performances. Longer time intervals in TR tasks differentiated patients better and TR impairment was associated with hyperactivity/impulsivity. These findings should be replicated in larger samples and underlying neurobiological components of impaired TR need to be examined in future research.Recent advances in our understanding of epitranscriptomic RNA methylation have expanded the complexity of gene expression regulation beyond epigenetic regulation involving DNA methylation and histone modifications. The instalment, removal, and interpretation of methylation marks on RNAs are carried out by writers (methyltransferases), erasers (demethylases), and readers (RNA-binding proteins), respectively. Contrary to an emerging body of evidence demonstrating the importance of RNA methylation in the diverse fates of RNA molecules, including splicing, export, translation, and decay in the nucleus and cytoplasm, their roles in plant organelles remain largely unclear and are only now being discovered. Thus, unveiling the cellular components involved in RNA methylation in cell organelles is essential to better understand plant biology.Patients with chronic ankle instability (CAI) walk with increased lateral plantar pressure, precipitating future injuries. Gait retraining to medially shift plantar pressure may prevent recurrent injury. We assessed if a multi-axis destabilisation device changed plantar pressure and muscle activity in patients with CAI during walking. Twelve adults with CAI (age 23.6 ± 5.0 years; body mass index 26.7 ± 4.5 kg/m2) participated. Insole plantar pressure and electro-myography were collected synchronously during treadmill walking. The destabilisation device had a half-sphere under both the rearfoot and forefoot. Two 30s walking trials were recorded at baseline, first without and second with the destabilisation device.
