The people of the United States and the world owe the United States Environmental Protection Agency (U.S. EPA) a debt of gratitude for preserving, protecting, and defending human health and the environment for the past half century. As we celebrate the 50th anniversary of the founding of the U.S. EPA, there are two truths about the agency that are difficult to deny (1) U.S. EPA and its people constitute a renowned agency that has greatly improved both environmental and public health in the United States, and has served as the leading model for nations around the world; and (2) the approaches, tools, structures, and legal frameworks that created the achievements of the U.S. EPA must evolve-and grow-to deal with the issues facing the country and the planet in the next 50 years. Building on the creativity, innovation, and brilliance of individuals and groups working at the U.S. EPA over the course of the last half century, we present 10 recommendations organized in three areas organization, paradigms, and strategies and tools. Underlying these recommendations are the frameworks of sustainability and systems thinking and guiding these recommendations is the goal of evolving the Environmental Protection Agency to the Environmental Prosperity Agency.Clarifying the signaling pathway associated with nitric oxide (NO) and glutathione (GSH) in cardiovascular disease therapy is important for understanding its physiological and pathological processes but is challenging due to the lack of efficient analytical techniques. Herein, we report a BODIPY-based fluorescent probe for recognition of NO and GSH in sequence with high sensitivity and selectivity. The probe exhibits turn-on fluorescence triggered by NO, followed by red-shifted emission in the presence of GSH. The sequentially activated mechanism allows the visualization of NO-induced GSH upregulation in drug-treated endothelial cells and zebrafish for the first time, revealing a signal pathway during the therapy. We hope that it can be used as a convenient and efficient tool for the study of the interplay between NO and GSH and for the screening of effective drugs for cardiovascular disease therapy.Poly(3-hydroxybutyrate-co-3-hydroxyhexanoate), PHBH or PHBHHx, is a novel bio-based polymer that is biodegradable in both soil and marine environments. While bio-based and biodegradability are often celebrated features to mitigate environmental problems of plastics, their life cycle environmental impacts contain uncertainties that are yet to be fully understood. To develop effective introduction schemes for PHBH, this study assessed the life cycle climate change implications of PHBH. We computed the life cycle greenhouse gas emissions (GHG) and fossil resource consumption of produce bags and spoons composed of PHBH and their fossil-based alternatives based on industrial-scale data. The products were assessed against 10 end-of-life scenarios for commercial plastics. As a result, the cradle-to-gate GHG of PHBH ranged between 0.32 and 16.5 kgCO2e/kg-PHBH depending on the land-use change assumed for the biomass production. The product-based comparative analysis presented that PHBH spoons have lower cradle-to-grave GHG emissions over their fossil-based alternatives but not with produce bags because PHBH spoons have a smaller GHG per functional unit than that of its fossil counterpart. The end-of-life scenario analysis conveyed that PHBH should be introduced to a region with a plastic waste management system that avoids methane generation and facilitates energy recovery.NMR studies and X-ray crystallography have shown that the structures of the 99-residue amyloidogenic protein β2-microglobulin (β2m) and its more aggregation-prone variant, D76N, are indistinguishable, and hence, the reason for the striking difference in their aggregation propensities remains elusive. Here, we have employed two protein footprinting methods, hydrogen-deuterium exchange (HDX) and fast photochemical oxidation of proteins (FPOP), in conjunction with ion mobility-mass spectrometry, to probe the differences in conformational dynamics of the two proteins. Using HDX-MS, a clear difference in HDX protection is observed between these two proteins in the E-F loop (residues 70-77) which contains the D76N substitution, with a significantly higher deuterium uptake being observed in the variant protein. Conversely, following FPOP-MS only minimal differences in the level of oxidation between the two proteins are observed in the E-F loop region, suggesting only modest side-chain movements in that area. Together the HDX-MS and FPOP-MS data suggest that a tangible perturbation to the hydrogen-bonding network in the E-F loop has taken place in the D76N variant and furthermore illustrate the benefit of using multiple complementary footprinting methods to address subtle, but possibly biologically important, differences between highly similar proteins.Multifunctional hydrogel with outstanding conductivity and mechanical flexibility has received enormous attention as wearable electronic devices. However, fabricating transparent, ultrastretchable, and biocompatible hydrogel with low-temperature stability still remains a tremendous challenge. ISM001055 In this study, an ultrastretchable, highly transparent, and antifreezing zwitterionic-based electronic sensor is developed by introducing zwitterionic proline (ZP) into gellan gum/polyacrylamide (GG/PAAm) double network (DN) hydrogel. The existence of ZP endows the hydrogel with remarkable frost resistance. The toughness and transparency of zwitterionic Ca-GG/PAAm-ZP DN hydrogel can be maintained down to -40 °C. Also, the zwitterionic hydrogel shows good biocompatibility and protein adsorption resistance. The zwitterionic Ca-GG/PAAm-ZP DN hydrogel-based strain sensor can accurately monitor human motions (such as speaking and various joint bendings) under a broad temperature range from -40 to 25 °C. The zwitterionic Ca-GG/PAAm-ZP DN hydrogel-based strain sensor will be of immense value in the field of wearable electronic devices, especially for extreme environment applications.Interactions between the hypothalamic-pituitary-adrenal axis and the central 5-HT system in the depressive state remain largely unknown. The present study investigated corticosterone (CORT) regulations of extracellular 5-HT in the hippocampal CA3 in a mouse model of depression. Basal dialysate 5-HT, true extracellular 5-HT, 5-HT reuptake efficiency, and time courses of dialysate 5-HT following CORT injections at 10, 20, and 40 mg/kg were determined at baseline, depressive-like state and after subsequent fluoxetine (FLX) treatment using in vivo microdialysis in male C57BL/6 mice. Behavioral tests were used to determine behavioral phenotypes and therapeutic responses to FLX. Depressed mice showed decreased extracellular 5-HT, increased 5-HT reuptake efficiency, and absence of the increase in dialysate 5-HT response to CORT injections, which were all reversed in FLX-responsive mice. Surprisingly, the FLX nonresponsive mice continued to worsen behaviorally and exhibited lower extracellular 5-HT and higher 5-HT reuptake efficiency.
