A recent publication reported an experimental 3D complex structure of the S protein of SARS-CoV-2 showed that the ectodomain of the SARS-CoV-2 S protein binds to the peptidase domain (PD) of human ACE2 with a dissociation constant (Kd) of ~ 15 nM. In this study, we focused on inhibitors for ACE2 S protein complex using virtual screening and inhibition studies through molecular docking for over 200,000 natural compounds. Toxicity analysis was also performed for the best hits, and the final complex structures for four complexes were subjected to 400 ns molecular dynamics simulations for stability testing. We found two natural origin inhibitors for the S protein human ACE2 complex (Andrographolide and Pterostilbene) which displayed better inhibition potential for ACE2 receptor and its binding with the S protein of SARS-CoV-2. Comparative studies were also performed to test and verify that these two drug candidates are also better than hydroxychloroquine which is known to inhibit this complex. However, we needed better potential drug candidates to overcome the side effects of hydroxychloroquine. Supplementary experimental studies need to be carried forward to corroborate the viability of these two new inhibitors for ACE2 S protein complex so as to curb down COVID-19.To simultaneously detect two metabolites of Aspergillus flavus, namely, cyclopiazonic acid (CPA) and aflatoxin (AFT), an ultrasensitive monoclonal antibody (mAb) YTT-2 against CPA was developed and characterized, with sensitivity to CPA of 1.32 ng mL-1. Along with the previously homemade mAb 1C11 against AFT, two mAbs were used to develop time-resolved fluorescence immunoprobes or gold immunoprobes. We developed two multiple-analyte paper immunosensors including time-resolved fluorescent immunochromatographic assay (TRFICA) and gold immunochromatographic assay (GICA) for the simultaneous determination of CPA and AFT. The TRFICA showed limits of determination (LODs) of 0.21 and 0.004 ng mL-1, while the GICA showed LODs of 0.33 and 0.01 ng mL-1 for CPA and AFT, respectively. To validate the specificity of the two rapid immunoassays, rice, corn and peanut samples were spiked with different concentrations of CPA and AFT. The two methods showed satisfactory recoveries (76.39~90.82% for CPA and 84.60~94.45% for AFT) and coefficients of variation of 3.50~7.80% for CPA and 4.12~13.90% for AFT. The results indicated that the TRFICA could complete the test within 5 min and had lower LODs and linear ranges, compared with that of GICA. The method developed in this work can be widely applied to the rapid and quantitative simultaneous determination of multiple harmful metabolites in fungi for food safety and health care. Graphical abstract.This proof-of-concept study aimed to assess the ability of a mobile application and cloud analytics software solution to extract facial expression information from participant selfie videos. This is one component of a solution aimed at extracting possible health outcome measures based on expression, voice acoustics and speech sentiment from video diary data provided by patients. DNQX chemical structure Forty healthy volunteers viewed 21 validated images from the International Affective Picture System database through a mobile app which simultaneously captured video footage of their face using the selfie camera. Images were intended to be associated with the following emotional responses anger, disgust, sadness, contempt, fear, surprise and happiness. Soils polluted with animal charcoal from skin and hide cottage industries harbour extremely toxic and carcinogenic hydrocarbon pollutants and thus require a bio-based eco-friendly strategy for their depuration. The effects of carbon-free mineral medium (CFMM) amendment on hydrocarbon degradation and microbial community structure and function in an animal charcoal-polluted soil was monitored for 6 weeks in field moist microcosms consisting of CFMM-treated soil (FN4) and an untreated control (FN1). Hydrocarbon degradation was monitored using gas chromatography-flame ionization detector (GC-FID), and changes in microbial community structure were monitored using Kraken, while functional annotation of putative open reading frames (ORFs) was done using KEGG KofamKOALA and NCBI's conserved domain database (CDD).

Gas chromatographic analysis of hydrocarbon fractions revealed the removal of 84.02% and 82.38% aliphatic and 70.09% and 70.14% aromatic fractions in FN4 and FN1 microcosms in 42 days. Shotgun metagenomilso revealed that intrinsic bioremediation of the polluted soil could be enhanced via addition of water and aeration.
This study revealed that though CFMM amendment slightly increases the rate of hydrocarbon degradation, it negatively impacts the structural and functional properties of the animal charcoal-polluted soil.
