Behçet's disease is a complex systemic inflammatory vasculitis of incompletely understood etiology. We performed a large genetic study in Behçet's disease in a diverse multi-ethnic population.
A total of 9,444 patients and controls from seven different populations were included in this study. Genotyping was performed using the Infinium ImmunoArray-24 V.1.0 or V.2.0 BeadChip. Analysis of expression data from stimulated monocytes, and epigenetic and chromatin interaction analyses were performed.
We identified two novel genetic susceptibility loci for Behçet's disease, including a risk locus in IFNGR1 (rs4896243, p value= 2.42 X 10
; OR=1.25) and within the intergenic region LNCAROD/DKK1 (rs1660760, p value= 2.75 x 10
; OR= 0.78). The risk variants in IFNGR1 significantly increase IFNGR1 mRNA expression in lipopolysaccharide-stimulated monocytes. In addition, our results replicated the association (p value< 5 x 10
) of six previously identified susceptibility loci in Behçet's disease IL10, IL23R, IL12A-AS1, CCR3, ADO, and LACC1, reinforcing these loci as strong genetic factors in Behçet's disease shared across ancestries. We also identified >30 genetic susceptibility loci with a suggestive level of association (p value< 5 x 10
), which will require replication. Finally, functional annotation of genetic susceptibility loci in Behçet's disease uncovered their possible regulatory roles and suggested potential causal genes and molecular mechanisms that could be further investigated.

We also replicate and extend the genetic associations in other loci across multiple ancestries.Central and peripheral respiratory chemoreceptors are stimulated during voluntary breath holding due to chemostimuli (i.e., hypoxia and hypercapnia) accumulating at the metabolic rate. We hypothesized that voluntary breath-hold duration (BHD) would be (a) positively related to the initial pressure of inspired oxygen prior to breath holding, and (b) negatively correlated with respiratory chemoreflex responsiveness. In 16 healthy participants, voluntary breath holds were performed under three conditions hyperoxia (following five normal tidal breaths of 100% O2 ), normoxia (breathing room air), and hypoxia (following ~30-min of 13.5%-14% inspired O2 ). In addition, the hypoxic ventilatory response (HVR) was tested and steady-state chemoreflex drive (SS-CD) was calculated in room air and during steady-state hypoxia. We found that (a) voluntary BHD was positively related to initial oxygen status in a dose-dependent fashion, (b) the HVR was not correlated with BHD in any oxygen condition, and (c) SS-CD magnitude was not correlated with BHD in normoxia or hypoxia. Although chemoreceptors are likely stimulated during breath holding, they appear to contribute less to BHD compared to other factors such as volitional drive or lung volume.
Healthcare workers are a key occupational group at risk for suicidal thoughts and behaviors (STB). We investigated the prevalence and correlates of STB among hospital workers during the first wave of the Spain COVID-19 outbreak (March-July 2020).
Data come from the baseline assessment of a cohort of Spanish hospital workers (n = 5450), recruited from 10 hospitals just after the height of the coronavirus disease 2019 (COVID-19) outbreak (May 5-July 23, 2020). Web-based self-report surveys assessed 30-day STB, individual characteristics, and potentially modifiable contextual factors related to hospital workers' work and financial situation.
Thirty-day STB prevalence was estimated at 8.4% (4.9% passive ideation only, 3.5% active ideation with or without a plan or attempt). A total of n = 6 professionals attempted suicide in the past 30 days. In adjusted models, 30-day STB remained significantly associated with pre-pandemic lifetime mood (odds ratio [OR] = 2.92) and anxiety disorder (OR = 1.90). Significant modifiable factors included a perceived lack of coordination, communication, personnel, or supervision at work (population-attributable risk proportion [PARP] = 50.5%), and financial stress (PARP = 44.1%).

Addressing adolescent depression has proven challenging using traditional face-to-face psychotherapies, with major barriers including workforce shortages internationally and reluctance to seek help among some adolescents. There is substantial evidence to support the use of digital tools to treat mental health problems, with the National Institute for Health and Care Excellence (NICE) now recommending such tools as a first-line treatment. In this article, we outline the evidence base for SPARX, a digital tool named specifically in NICE guideline NG134, and discuss its use in clinical settings. We also consider implementation issues and future research directions.Gestational exposure to environmental pollutants can induce oxidative injury and apoptosis since the fetal organs are sensitively vulnerable to these chemicals. In this work, we have investigated the renal anti-apoptotic efficiency of linseed (LS) against the oxidative stress-mediated upregulation of the fetal apoptosis-related genes following the prenatal intoxication with diesel nanoparticles (DNPs) and/or fenitrothion (FNT). A fifty-six timed-pregnant rats were equally divided to eight groups; control, LS (20% in diet), DNPs (0.5 mg/kg by intratracheal inoculation), FNT (3.76 mg/kg by gavage), DNPs+FNT, LS + DNPs, LS + FNT, and LS + DNPs+FNT. The transmission electron microscope analysis revealed the spherical shape of diesel particles with a homogeneous nanosized range (20-92.3 nm) and the crystallinity was confirmed by electron diffraction microscopy. Administration of DNPs and/or FNT significantly increased fetal renal malondialdehyde, nitric oxide, and glutathione reductase as compared with the control group.
