C-type lectins (CTLs) are a superfamily of proteins found in almost all vertebrates and invertebrates. They play an important role in innate immune defences, development and epidermal structure. Here, a CTL with one carbohydrate-recognition domain containing a highly conserved Gln-Pro-Asp (QPD) motif was identified in Tribolium castaneum and given the name TcCTL5. Spatiotemporal analyses showed that Tcctl5 was highly expressed in the late pupa stage and mainly existed in the central nervous system and haemolymph. The transcript level of Tcctl5 was prominently induced after bacterial infection. Recombinant TcCTL5 proteins (rTcCTL5) were found to bind to lipopolysaccharide, peptidoglycan and tested bacteria and induce microbial agglutination in the presence of Ca2+ . Interestingly, when Tcctl5 was knocked down, the transcript level of antimicrobial peptides (AMPs) (attacin1, defensins3, coleoptericin1 and cecropins3) was prominently downregulated after induction with Gram-negative Escherichia coli. More interestingly, Tcctl5 was knocked down, leading to increased mortality and loss of locomotor activity, which exhibited less travel distances among early adults. These results demonstrate that Tcctl5 plays an important role in the innate immune reaction and the movement of T. castaneum. Thus, it may represent an alternative molecular target for pest control and thus reduce the use of pesticides in agricultural production.
Vaccination to prevent hepatitis B (HB) virus infection is important for children undergoing immunosuppressive treatment. Information on efficacy of vaccination in children with rheumatic disease undergoing immunosuppressive therapy is scarce.

Children with rheumatic disease administered HB vaccine during immunosuppressive treatment between May 2013 and September 2016 were enrolled. Patients were vaccinated three times (primary series). Those who remained seronegative after the primary series received a secondary series of vaccination. Patient baseline characteristics and treatment details in medical records were retrospectively investigated. The proportions of patients that were seropositive for HB virus antibody after primary- and after secondary-series vaccinations were calculated. Associations between immunosuppressants and serostatus were evaluated.

Fifteen out of 26 patients (58%) produced anti-hepatitis B surface antibody (anti-HBs) after primary series vaccination. The proportions of patients taking methotrexate and those taking mycophenolate mofetil (MMF) that showed seropositive were 80% (8/10) and 27% (3/11), respectively. Multivariate analysis adjusted for dosage of PSL per body weight. Multivariate analysis showed MMF was a factor impeding seroconversion (odds ratio 0.093, 95% confidence interval 0.014 to 0.615). In six of seven patients (86%) that received secondary series vaccination, anti-HBs were produced.

Detective flow imaging for endoscopic ultrasonography (DFI-EUS) is a new imaging modality developed for detecting fine vessels without using ultrasound contrast agents. Between January 2019 and January 2020, 28 patients with SELs undergoing DFI-EUS and eFLOW-EUS were enrolled. DFI-EUS and eFLOW-EUS assessing the vascularity in SELs were compared in terms of the rates of identification of intratumoral vessels. We also investigated how large vessels were depicted in both modalities based on surgical specimens as well as the detection rates of intratumoral vessels in gastrointestinal stromal tumors (GISTs) and non-GISTs using either DFI-EUS or eFLOW-EUS.

Among 28 patients, 23 with pathological confirmation by EUS-guided fine-needle aspiration biopsy (EUS-FNAB) specimens were included. Of those 23 patients, the 10 who underwent surgical resection were selected for analysis. The rate of detection of intratumoral vessels in SELs was significantly higher on DFI-EUS (80%) than on eFLOW-EUS (30%) (P=0.03). Comparison with surgical specimens revealed that detection rate for vessels with maximum size of less 1000μm was higher in DFI-EUS (66%) than that in eFLOW-EUS (0%). GIST patients had significantly higher positive rates (90%) of intratumoral vessels than non-GIST patients (31%) on DFI in 23 cases including EUS-FNAB specimens (P=0.045).

In a double-blind, placebo-controlled, early phaseII study, women with stress urinary incontinence and stress urinary incontinence-predominant mixed urinary incontinence were randomized to a placebo or TAS-303 (3 or 6mg) once daily for 8weeks. The main efficacy end-points were mean percentage change in incontinence episode frequency per 24h from baseline to week8 (the primary end-point) and week4.

At week8, the mean percentage change in incontinence episode frequency per 24h was -34.73% in the TAS-303 3mg group, -35.41% in the TAS-303 6mg group and -28.07% in the placebo group (differences vs placebo, not significant). In patients with stress urinary incontinence, or incontinence episode frequency less than two episodes per 24h at baseline, TAS-303 significantly reduced incontinence episode frequency versus placebo after 4weeks; some secondary end-points also showed a tendency to improve in the same subgroups.
