An indoor microcosm experiment was designed with temperature variation (20 °C and 30 °C), salinity (no addition, 2.5 g/L NaCl) and the insecticide chlorpyrifos (no addition, 1 μg/L) as treatments. The impact of these stressors was evaluated on water quality variables and on the zooplankton comunity (structure, diversity, abundance and taxa responses) for 28 days. This study shows that temperature is the main driver for zooplankton community change, followed by salinity and chlorpyrifos. The three stressors contributed to a decrease on zooplankton diversity. The increase of temperature contributed to an increase of zooplankton abundance. Salinity generally affected Cladocera, which resulted in a Copepoda increase at 20 °C, and a reduction in the abundance of all major zooplankton groups at 30 °C. The insecticide chlorpyrifos affected primarily Cladocera, altough the magnitude and duration of the direct and indirect effects caused by the insecticide substantially differed between the two temperature scenarios. Chlorpyrifos and salinity resulted in antagonistic effects on sensitive taxa (Cladocera) at 20 °C and 30 °C. This study shows that temperature can influence the direct and indirect effects of salinity and pesticides on zooplankton communities in Mediterranean coastal wetlands, and highlights vulnerable taxa and ecological responses that are expected to dominate under future global change scenarios.A gallic acid-affinity molecularly imprinted polymer (G-MIP) was first used as an adsorbent for selective identification and capture of luteolin (LTL) in herbal medicine samples. The G-MIP was prepared by using LTL as the template, gallic acid (GA) as the functional monomer, ethylene glycol dimethacrylate (EGDMA) as the crosslinking agent, and 2,2'-azobis(2-methylpropionitrie) (AIBN) as the initiator. The properties of G-MIP were characterized by FT-IR, transmission electron microscope, scanning electron microscope, dynamic light scattering, specific surface area, and X-ray photoelectron spectrum. The adsorption conditions were optimized, and the adsorption equilibrium model and adsorption kinetics model of the adsorbent were investigated under the best experimental conditions. The saturated adsorption capacity is 1.24 mg g-1, which is not only higher than the adsorption capacity of 4-carboxyphenylboronic acid-affinity MIP adsorbent but also superior to those of many reported adsorbents for enriching of LTL. The LTL was quantified by HPLC. The linear range is 0.05-100 mg L-1, the detection limit is 0.020 mg L-1. This method was successfully applied in the selective recognition of LTL in herbal medicines with recoveries of 93.9-114.2%, and the relative standards deviations (RSDs) are 0.4-5.6%. Thus, this work provides a potential possibility and practical platform for the determination of LTL in complex matrices.Gas chromatography-olfactometry-mass spectrometry (GC-O-MS) has been very useful in identifying aroma compounds from within the complex matrix of wine. Supplementary separation can be required to overcome co-elution of volatiles or other sensory-directed chromatographic strategies are needed, including multidimensional chromatography and preparative fraction collection coupled to GC. Studies investigating 'overripe orange' aroma in sweet Sauternes wine and the similar 'apricot' aroma in Viognier wine were conducted. Wines with the targeted aroma attributes were selected and concentrated wine extracts prepared. GC-O found no individual aroma compounds with the targeted aroma attribute. Semi-preparative HPLC was used to obtain less complex fractions of the wine extracts. The fractions were eluted in water/ethanol and, therefore, could be smelled directly. Fractions with the targeted aroma character were further resolved by GC-preparative fraction collection (GC-PFC). Recombinational GC-PFC demonstrated the importance of the components within a 4 min preparative GC fraction to the 'overripe orange' aroma of typical Bordeaux dessert wine. In Viognier wine, monoterpenes linalool, α-terpineol and geraniol as well as benzaldehyde were found to be associated with the 'apricot' character. Thus, several wine aroma compounds interact for these specific aromas to be perceived. This sensory-led combination of separation techniques is a powerful tool for the identification of key compounds responsible for specific aromas across the wine and beverage industries.Accurate quantification of effective antigens of different serotypes is crucial for quality control of multivalent vaccines but challenging. A simple and rapid capillary zone electrophoresis (CZE) method was developed for on-line separation and quantification of foot-and-mouth disease virus (FMDV) antigens in monovalent and bivalent FMDV vaccines. The FMDV peak identity in CZE was demonstrated by the study of FMDV dissociation combined with high performance size exclusion chromatography (HPSEC) analysis. After optimizing CZE conditions including UV detecting wavelength, injection volume, and separation voltage, both serotype A and O FMDV showed good reproducibility (RSD less then 5%) and linear responses (R2=0.999) between the peak area and FMDV content in the concentration range of 15-400 μg/mL. The two serotypes of FMDV with similar size had different migration time in CZE according to their different zeta potential, which allows them to be separated and quantified, with accuracy of less then 10% relative error.
