We found that the apo form undergoes substantial conformational changes in the β4-α4 loop, α7-α8 helices, and C-terminus loop among the four subunits comprising the tetramer. Furthermore, a comparison of the apo structure with the binary (NAD+)-complex and ternary (NADH and 7-oxoglycochenodeoxycholic acid)-complex Eco-7α-HSDH structures revealed that only the ternary-complex structure has a fully closed conformation, whereas the binary-complex and apo structures have a semi-closed or open conformation. This open-to-closed transition forces several catalytically important residues (S146, Y159, and K163) into correct positions for catalysis. To confirm the catalytic activity, we used alcohol dehydrogenase for NAD+ regeneration to allow efficient conversion of chenodeoxycholic acid to 7-ketolithocholic acid by Eco-7α-HSDH. These findings demonstrate that apo Eco-7α-HSDH exhibits intrinsically flexible characteristics with an open conformation. This structural information provides novel insight into the 7α-HSDH reaction mechanism.Research into the biosynthesis of C11-oxy C19 steroids during human fetal development, specifically fetal adrenal development and during the critical period of sex differentiation, is currently lacking. This review will consider the role of cortisol, 11KT and 11β-hydroxysteroid dehydrogenase type 2 (11βHSD2) in determining the sex of teleost fish, while these hormones and 11βHSD2 will also be discussed with regards to murine mammals. The focus of the review will shift to highlight the potential role of C11-oxy steroids in human fetal development based on the timely expression of steroidogenic enzymes in the adrenal, testes and ovary, as well as in the placenta; summarising reported evidence of C11-oxy steroids in neonatal life.A slug is a shell-less terrestrial gastropod mollusk. During evolution, slugs have lost their mineralized external shell but some of them have retained an internal shell (IS). Unlike external shells, which have been widely investigated, the ISs have been poorly studied. We report for the first time the compositional and complete morphological characterization of Ariolimax californicus' IS. According to literature, this shell calcifies and decalcifies depending on the animal's needs. Its composition is mostly organic, consisting of proteins and β-chitin. The internal shell is organized in layers and membranes in which CaCO3 crystal formation occurs in specific areas. In the two faces of the IS we observed different morphologies and aggregations of calcite bio-crystals along with a different organization of the organic matrix. SNDX-5613 Dorsally, the mineral forms a thick layer composed of misaligned crystal aggregates of large dimensions, separated by thin organic layers. This suggests a protective purpose and the use of this layer as a long-term calcium storage system. The whole ventral mineral layer is covered by a membrane, identified as the hypostracum. This face is proposed to be a short-term calcium storage system. In vitro crystallization experiments suggest massive calcium ions sequestration from the solution for the precipitation of calcite crystals inside the organic matrix. However, exposure to environmental pollutants as a contribution to kidney stone formation remains insufficiently studied. Lead (Pb2+) is of particular interest as epidemiological data indicate that low-level exposure (BLL = 0.48-3.85 μM) confers a 35 % increased risk of developing CaOx nephrolithiasis. However, mechanisms underlying this association have yet to be elucidated. Drosophila melanogaster provide a useful genetic model where major molecular pathophysiological pathways can be efficiently studied. Malpighian tubules (MT) were isolated from either Wild-Type or InsP3R knockdown flies and treated with oxalate (5 mM) ± Pb2+ (2μM) for 1 h. Following exposure, MTs were imaged and crystals quantified.
