Pigment epithelium-derived factor (PEDF) may play a role in cardiometabolic disorders. The aim of this study was to investigate which biochemical and clinical parameters are independently associated with serum PEDF levels in patients with type 2 diabetes mellitus (T2DM).
We performed a cross-sectional analysis of 124 patients with T2DM who underwent continuous glucose monitoring (CGM) and blood chemistry analysis, including the diacron-reactive oxygen metabolites (d-ROMs) test and serum PEDF measurement (study 1). Then we investigated whether the changes in the studied biochemical and clinical parameters after 24 weeks of treatment (Δparameters) with anti-hyperglycemic agents, including sodium-glucose cotransporter 2 inhibitors, glucagon-like peptide 1 receptor agonists, and/or insulin and anti-hypertensive drugs and statins, were independently correlated with change in PEDF (ΔPEDF) in 52 of the patients with T2DM for whom there was sufficient serum samples to perform the post-treatment analysis (study 2)m PEDF level in patients with T2DM.In this study, we aimed to reveal whether the medial longitudinal arch is formed in the intrauterine period and the structural features of the medial longitudinal arch. The study was conducted on 146 feet of 73 fetuses (38 male, 35 female) aged between 15 and 40 weeks of gestation. This study aims to characterize the early cadmium (Cd) histological and hematological alterations and their corresponding plasma indicators in the Mediterranean sea bass (Dicentrarchus labrax). We also assessed garlic potential to prevent cadmium toxicity. For this purpose, 200 fish of 55 g mean weight were separated into 3 cylindroconical fiberglass tanks of 500-L capacity, each with a stocking density of 4 kg m-3. The fish were regularly hand-fed 0% (control group), 2%, and 6% garlic-supplemented diets to apparent satiation twice a day for 1 month. At the end of the experiment, we injected 22.2 mM cadmium (CdCl2) intraperitoneally to the experimental groups and a placebo solution (9% NaCl) to the control groups; liver, kidney, heart, and blood tissue alterations were monitored with a full screening of their plasmatic indicators, 24 h before and 48 h after Cd injection. Subsequently, whole blood count and blood smears were performed to follow up on Cd-induced vascular damages. Our data showed that Cd induced thrombotic thrombocytopenic purpura, leading to widespread bleeding and cellular alterations in the targeted tissues. These alterations were associated with an obvious normochromic normocytic anemia in favor of microangiopathic hemolytic anemia. Cd injection has also seriously inhibited the overall enzymatic activities triggering a metabolic shift. Although garlic supplementation had little effect on cadmium-induced alterations, it significantly reduced biomass dispersion. Our data is the first evidence of the cadmium versatile toxicity involving vascular alterations as a central and a leading cause of the overall parenchymal lesions. Cd toxicity was associated with a specific enzymatic signature, which must be considered during the interpretation.
To evaluate the effect of repeated administration of zoledronic acid (ZA) on the development and severity of osteonecrosis of the jaws.
In the experiment, 36 rats were used, which were divided into 4 groups group 1 was injected with saline for 6 weeks, group 2 was injected with ZA once, group 3 was injected zoledronic acid for 3 weeks once a week, group 4 was injected with ZA for 6 weeks once a week. While taking medications, the tooth was removed. The volumetric blood flow rate was studied using laser and high-frequency ultrasound Doppler in the area of the periodontium of an extracted tooth in rats with the application of acetylcholine. Bone tissue was examined out using CBCT.
In group 2, there was a violation of blood flow in the mucous membrane, bone microcirculation, but no reliable data was obtained in the bone defect in comparison with group 1. In groups 3 and 4, there was significantly disrupted blood flow. This led to an increase of the osteonecrosis (maximum at the 4th group), which was confirmed by data obtained using CBCT. An inverse relationship was observed between the blood flow of bone tissue and the size of the defect after tooth extraction.
The introduction of ZA in a dose-dependent fashion resulted in impaired endothelial vasodilation and impaired blood flow to extraction sockets. These findings might explain the development of osteonecrosis of the mandible following removal of a first molar.
