A recent study reported the propensity of the concomitant inhibition of TcGLK and TcHK by compounds GLK2-003 and GLK2-004, thereby presenting an efficient approach in Chagas disease treatment. We investigated this possibility using atomic and molecular scaling methods. Sequence alignment of TcGLK and TcHK revealed that both proteins shared approximately 33.3 % homology in their glucose/inhibitor binding sites. The total binding free energies of GLK2-003 and GLK2-004 were favorable in both proteins. PRO92 and THR185 were pivotal to the binding and stabilization of the ligands in TcGLK, likewise their conserved counterparts, PRO163 and THR237 in TcHK. Both compounds also induced a similar pattern of perturbations in both TcGLK and TcHK secondary structure. Findings from this study therefore provide insights into the underlying mechanisms of dual inhibition exhibited by the compounds. These results can pave way to discover and optimize novel dual Tc inhibitors with favorable pharmacokinetics properties eventuating in the mitigation of Chagas disease.
Asians develop type2 diabetes at a lower body mass index (BMI) compared with other races, which is partly because of Asian-specific fat depots. Sex plays a role in fat deposition, regardless of race. This retrospective cohort study aimed to investigate the association among fatty liver, sex and type2 diabetes in non-obese Japanese.

The participants in this study (13,596 men and 6,037 women) were aged 30-64years, and had undergone health checkups between 2013 and 2015, in Aichi, Japan. Baseline BMI was categorized as follows <18.5, 18.5-19.9, 20-22.9, 23-24.9, 25-27.4 and ≥27.5kg/m
. Fatty liver was diagnosed by abdominal ultrasonography. The joint effect of BMI and fatty liver on the incidence of type2 diabetes was assessed, stratified by sex.

To develop a working definition of 'clinical credibility' in nursing science.

This is stage 2 of a registered report. Concept mapping methodology, 1. Preparation, 2. Generation of statements (brainstorming), 3. Structuring of statements (prioritising/clustering), 4. Representation of statements, 5. Interpretation of maps and 6. Utilisation of maps were used. Part 1 of our registered report followed the required items on the SPIRIT 2013 checklist. Part 2 was reporting the results of our study adhered to the STROBE reporting guidelines.

Participants (n=67) from three stakeholder groups participated in this study. Nursing students (n=23), nurses and other healthcare professionals (n=23), and patients (within the last 12months) (n=21). Participants (n=62) generated statements (n=429) in response to the question 'What does clinical credibility mean to you?'. Following statement reduction, participants (n=61) prioritised and clustered (n=80) statements. The data were analysed using the concept mapping software, Ariadne. The dominance of Fabaceae Detarioideae species in miombo ecosystems is remarkable but little understood. We therefore compared plant functional traits (PFTs) of common woody species of the Angolan plateau, grouped by life form (trees, geoxyles), lineage (Fabaceae Detarioideae, non-Detarioideae) and symbiont association (ectomycorrhiza, rhizobia). PFTs reflect group-specific adaptations to prevalent environmental conditions. To analyse the impact of environmental drivers, we selected PFTs reflecting ecophysiological aspects of leaf morphology, nutrient content and water transport. Traits were measured following standardized protocols. We found differences in key PFTs between trees and geoxyles reflecting both life form-specific adaptations to environmental conditions and lineage-specific strategies to cope with environmental stresses.
