Tissue regeneration and wound healing are still serious clinical complications globally and lack satisfactory cures. Inspired by the impressive regeneration ability of the post-injury earthworms and their widely accepted medicinal properties, we screened and identified a novel collagen-like peptide from the amputated earthworms using high-throughput techniques, including transcriptomics, proteomics, and mass spectrum. The identified collagen-like peptide col4a1 was cloned and expressed to comprehensively investigate the wound healing effect and underlying mechanism. It exerted significant effects on wound healing both in vitro and in vivo, including enhanced viability, proliferation, migration of fibroblasts, granulation, and collagen deposition. Moreover, the col4a1 functioned via binding with integrin α2β1 and upregulating the RAS/MAPK signaling pathway. This work demonstrates that the novel collagen-like peptide col4a1 obtained from the amputated earthworms enables enhanced wound healing and provides new opportunities for wound care.Vibrio anguillarum is a globally distributed aquatic pathogen, and its flagellin B (FlaB) protein can evoke innate immune responses in hosts. In order to explore the role of FlaB in V. anguillarum infection, we constructed a FlaB-deficient mutant using overlapping PCR and two-step homologous recombination, and gene sequencing confirmed successful knockout of the FlaB gene. Scanning electron microscopy showed no significant differences in the morphological structure of the flagellum between wild-type and FlaB-deficient strains. The mutant was subsequently injected into the freshwater prawn (Macrobrachium rosenbergii) to explore its pathogenicity in the host, and expression of myeloid differentiation factor 88, prophenoloxidase, catalase, superoxide dismutase and glutathione peroxidase was investigated by real-time PCR. The results showed that deletion of FlaB had little effect on V. anguillarum-induced expression of these immune-related genes (p > 0.05). In general, the FlaB mutant displayed similar flagella morphology and immune characteristics to the wild-type strain, hence we speculated that knockout of FlaB might promote the expression and function of other flagellin proteins. Furthermore, this study provides a rapid and simple method for obtaining stable mutants of V. anguillarum free from foreign plasmid DNA.The chitosan chains were integrated with MCM-48 mesoporous silica in an eco-friendly composite (CH/MCM-48) of enhanced adsorption capacity. The prepared CH/MCM-48 composite was applied in systematic retention of U (VI) as well as Sr (II) ions from water as the commonly detected radioactive pollutants. It displayed promising retention capacities of 261.3 mg/g and 328.6 mg/g for U (VI) and Sr (II) considering the equilibrium time interval that was identified after 420 min. The composite showed the kinetic behavior of the Pseudo-First order model and the isotherm properties of the Langmuir assumption. The thermodynamic assessment of the reactions validated the retention of both U (VI) and Sr (II) ions by spontaneous, favorable, and exothermic reactions. Based on the theoretical values of entropy (-5.94 kJ mol-1 (U (VI)) and -2.93 kJ mol-1 (Sr (II))), Gibbs free energy (less than 20 kJ mol-1), and Gaussian energy (5.77 kJ mol-1 (U (VI)) and 4.56 kJ mol-1 (Sr (II))) the uptake processes are related to physical adsorption reactions. The CH/MCM-48 composite is of significant recyclability and showed considerable affinities for the studied radioactive ions even in the presence of other metal ions (Cd (II), Pb (II), Zn (II), and Co (II)).In this study lipolytic biocatalysts GD-95RM, GDEst-95 and GDEst-lip were immobilized by encapsulation in calcium alginate beads. All three immobilized biocatalysts demonstrated significantly increased thermal stability at 60-70 °C temperatures and the activity of GD-95RM lipase increased by 50% at 70-80 °C following the immobilization. Moreover, encapsulated GDEst-95 esterase retained higher than 50% lipolytic activity after 3 months of incubation with butanol (25%) and ethanol (50%); GDEst-lip enzyme possessed 50% activity after 2 months of treatment with ethanol (25%) and methanol (25%); and GD-95RM lipase displayed higher that 50% activity after two-week incubation with methanol (50%). All three immobilized enzymes displayed long-term storage capability (>50% activity) at least until 3 months at 4 °C. It was also detected that immobilized GD-95RM and GDEst-lip can perform flow hydrolysis of both avocado oil and p-NP dodecanoate in prototype packed-bed column reactor. The analysis of continuous transesterification of avocado or sunflower oil with ethanol or methanol as substrates confirmed that encapsulated GD-95RM and GDEst-lip enzymes is a useful approach to produce fatty acid alkyl esters.
