With introduction of ICIs the landscape for cancer treatment has changed significantly. ICIs are known to induce immune-related adverse effects. This research is intended to shed light on ICIs and describe our safety experience with these agents. This study is a retrospective cohort study aimed to determine the safety of ICIs and its related adverse events at a tertiary hospital in Saudi Arabia. The study was conducted in the oncology center at King Abdulaziz Medical City, Riyadh. We identified study participants by using electronic health care system (BestCare)® to involve patients who received ICIs treatment during the study period from January 2016 up to December 2018, to include a total of 53 patients. Most of our patients were on nivolumab (37 patients) followed by atezolizumab (10 patients), and pembrolizumab (6 patients). The averaurological adverse events). In view of ever-escalating use of 2,4-D, risk assessment becomes mandatory to ensure the safety of both human health and the ecosystem. Oxidative injury has been expected as a possible mechanism implicated in 2,4-D toxicity. The present study was planned and conducted to explore the antioxidant potential of selenium (Se) supplementation to moderate the 2,4-D hepatic and renal toxicity in a rat model. The rats were randomly assigned to four equal groups and treated via oral gavage for a period of 4 weeks. Group I received deionized water as a vehicle, group II received 2,4-D (150 mg-1 kg-1 day-1), group III received Se supplement (1 mg-1 kg-1 day-1), and group IV received 2,4-D (150 mg-1 kg-1 day-1) and Se supplement (1 mg-1 kg-1 day-1) simultaneously. After 4 weeks of administration, 2,4-D induced toxicity was observed, as manifested by disrupted levels of plasma urea, creatinine, alkaline phosphatase (ALP), aspartate aminotransferase (AST), and alanine aminotransferase (ALT). Further, 2,4-D caused a considerable increase in tissue malondialdehyde (MDA) levels and decreased activity of antioxidant enzymes, including superoxide dismutase, catalase, and glutathione reductase. Se supplementation exhibited its antioxidant properties by significantly improving urea, creatinine, ALP, AST, and ALT, and MDA levels and antioxidant enzyme activities. The purpose of the current study is to determine if there is a progressive loss of retinal ganglion cells (RGCs) in the mutant over time and whether this loss was associated with macroglial activity changes and elevated intraocular pressure (IOP).Methods Using the Cre-loxP system, we generated a conditional knockout of Tfap2b exclusively in cranial NCC (AP-2β NCC KO). Immunohistochemistry was performed using anti-Brn3a, anti-GFAP and anti-Vimentin antibodies. IOP was measured using a tonometer and the data was analyzed using GraphPad Prism software. Brn3a and DAPI positive cells were counted using Image-J and statistical analysis was performed with GraphPad Prism software.Results Our findings revealed that while no statistical difference in Brn3a expression was observed between wild-type and mutant mice at postnatal day (P) 4 or P10, at P40 (p less then .01) and P42 (p less then .0001) Brn3a expression was significantly reduced in the mutant retina at the region of the ONH. There was also increased expression of glial fibrillary acidic protein (GFAP) by Müller cells in the AP-2β NCC KO mice at P35 and P40, indicating the presence of neuroinflammation.
