The detection of anti-platelet antibodies (APA) in plasma, although highly specific, is notoriously insensitive. Specialised clinical platelet laboratories routinely perform a screening test of only one dilution for indirect APA testing by flow cytometry. We evaluated the presence of APA using several dilutions of plasma from 61 ITP patients. Herein, we report that serial dilutions can improve the diagnostic value of indirect APA assay for ITP. We show that performing just two dilutions (12 and 125) would capture over 90% of patients with detectable plasma APA. This method enables indirect testing to become a valuable tool to be incorporated into the management algorithm for ITP.We conducted a multicentre cross sectional observational study of laboratory, public health and hospitalisation data for PCR-confirmed COVID-19 cases within the New Zealand Northern Region, between 12 February and 8 June 2020. The aim of this study was to describe population level SARS-CoV-2 upper respiratory tract (URT) viral load dynamics by stratifying positivity rates and polymerase chain reaction (PCR) cycle threshold (Ct) values of URT samples from COVID-19 cases by days since symptom onset, and to explore utility of Ct values in determining length of time post-infection and thus potential infectivity. Of 123,124 samples tested for SARS-CoV-2 by PCR, 579 samples (407 positive and 172 negative) from 368 symptomatic non-hospitalised individuals with PCR-confirmed infection were included. Sample positivity rate was 61.5% (8/13) for pre-symptomatic samples, rising to 93.2% (317/340) for samples collected during the purported symptomatic infectious period (days 0-10 post-symptom onset), and dropping to 36.3%of time post-infection or to exclude infectivity where date of symptom onset is unavailable.
A patient 3-dimensional virtual representation aims to facilitate the integration of facial references into treatment planning or prosthesis design procedures, but the accuracy of the virtual patient representation remains unclear.

The purpose of the present observational clinical study was to determine and compare the accuracy (trueness and precision) of a virtual patient obtained from the superimposition procedures of facial and intraoral digital scans guided by 2 scan body systems.

Ten participants were recruited. An intraoral digital scan was completed (TRIOS 4). Four fiduciary markers were placed in the glabella (Gb), left (IOL) and right infraorbital canal (IOR), and tip of the nose (TN). Two digitizing procedures were completed cone beam computed tomography (CBCT)(i-CAT FLX V-Series) and facial scans (Face Camera Pro Bellus) with 2 different scan body systems AFT (ScanBodyFace) and Sat 3D (Sat 3D). For the AFT system, a reference facial scan was obtained, followed by a facial scan with the partict 3-dimensional virtual representations obtained using AFT and Sat 3D systems showed trueness values ranging from 0.50 to 1.64 mm and precision values ranging from 0.04 to 0.14 mm. The AFT system obtained higher trueness than the Sat 3D system, but both systems showed similar precision values.
Several manufacturers have developed a digital workflow for removable prosthodontics. The Baltic Denture System is a 2-visit procedure for the digital production of complete dentures, but clinical studies comparing the system with conventional methods are lacking.

The purpose of this randomized, controlled, blinded crossover trial was to evaluate the impact of the digital versus conventional production of complete dentures on oral health-related quality of life (OHRQoL) measures.

Sixteen participants received 2 sets of new complete dentures produced with a digital (2 visits) and conventional (5 visits) workflow. Each complete denture was in clinical service for an observation period of 3 months. The order of the dentures was randomized, starting with digital or conventional followed by changing the set of dentures after 3 months. Givinostat The primary outcome was the time-dependent change in the OHRQoL, assessed by using the Oral Health Impact Profile, German version (OHIP-G49), comparing the digital with the conication method of the complete dentures had no significant influence on OHRQoL. However, digital dentures needed only 2 visits, 1 hour less chair time, and 5 hours less time for the dental laboratory technicians.

Medium- to long-term data for the performance of zirconia crowns with titanium (Ti) bases are sparse, particularly when the crown height space and occlusal loads are high.

The purpose of this invitro study was to assess the effect of the height of zirconia screw-retained implant crowns with a Ti base on the screw joint stability after cyclic loading. A secondary aim was to investigate the survival of zirconia crowns of different heights after cyclic loading.

Twenty-one internal connection implants were secured between fiberglass-reinforced epoxy resin sleeves. Mandibular first molar monolithic zirconia crowns with 3 different heights (6 mm, 10 mm, and 14 mm) were milled and bonded to the Ti bases (n=7). The screws were tightened to 30 Ncm, and a 30-degree 120-N cyclic load was applied to the crowns at 2 Hz for 5 million cycles. After 5 million cycles, the crowns were evaluated for stability, and the same protocol was repeated for 275-N and 435-N loads for 5 million cycles each. After loading, the detorque values were recorded.
