CFA dissolution often occurs in conjunction with declining dissolved calcium concentrations. Therefore, managed aquifer recharge (MAR) operations using deionised or low calcium source water are at risk of disturbing the naturally persisting geochemical equilibrium between CFA and the ambient groundwater and induce fluoride mobilisation. In this study, we employ reactive transport modelling to investigate how an engineered manipulation of the MAR source water composition might mitigate such groundwater fluoride contamination. Based on a previously developed and calibrated model for Australia's largest groundwater replenishment operation, we investigate the efficiency of (i) raising aqueous calcium concentration through the addition of CaCl2 or Ca(OH)2 amendment, (ii) raising aqueous sodium concentrations through the addition of NaCl or sea salt amendment and (iii) raising the pH. The modelling results illustrate in detail how the geochemical zonation around injection boreholes evolves over time and how this affects fluoride release and attenuation for the different amendment types. Treatments involving the addition of calcium and sodium in the source water are both found to be effective at reducing maximum groundwater fluoride concentrations during MAR, with calcium generally producing the greatest reduction in maximum fluoride concentrations. In contrast, increasing the injectate pH was found to be inefficient in reducing fluoride concentrations significantly due to the strong pH buffering effect of the aquifer sediments.Anaerobic digestion (AD) is a key technology at many wastewater treatment plants (WWTPs) for converting primary and surplus activated sludge to methane-rich biogas. However, the limited number of surveys and the lack of comprehensive datasets have hindered a deeper understanding of the characteristics and associations between key variables and the microbial community composition. Here, we present a six-year survey of 46 anaerobic digesters, located at 22 WWTPs in Denmark, which is the first and largest known study of the microbial ecology of AD at WWTPs at a regional scale. For three types of AD (mesophilic, mesophilic with thermal hydrolysis pretreatment, and thermophilic), we present the typical value range of 12 key parameters including operational variables and performance parameters. High-resolution bacterial and archaeal community analyses were carried out at species level using amplicon sequencing of >1,000 samples and the new ecosystem-specific MiDAS 3 reference database. Here, we developed a magnetic bead-mediated selective adsorption strategy (MagExo) for easy-to-operate EV isolation. Benefited from the presence of an adsorption window between EVs and proteins under the effect of a hydrophilic polymer, EVs tend to adsorb on the surface of magnetic beads selectively and can be separated from biological fluids with high purity by simple magnetic separation. The proposed method was used for EV isolation from plasma and cell culture media (CCM), with two times higher yielisolated EVs; this makes the isolation of EVs an essential challenge in EV research. In the present work, we proposed a simple and easy-to-operate method (MagExo) for the separation and purification of EVs based on the phenomenon that EVs can be selectively adsorbed on the surface of magnetic microspheres in the presence of a hydrophilic polymer. The performance of MagExo was comparable to or even better than that of gold standard methods and commercial kits, with two times higher yield and comparable purity of the harvested EVs to that achieved with ultracentrifugation (UC); this could meet the requirements of various EV-associated downstream applications. Parameters as effective dead space, rebreathing, pressure, or temperature field distribution are quantified and analysed in detail in order to study the performance and feasibility of such devices to relieve the effects of respiratory infections.
