In this work, a unique nanobranched microelectrode array (NBMEA) platform is developed to achieve high-quality intracellular recording of cultured cardiomyocytes in a minimally invasive manner. The NBMEA is consisting of high aspect ratio conductive nanobranches fabricated on patterned microelectrodes combining hydrothermal growth and standard microfabrication. This NBMEA platform is a promising and powerful tool for electrophysiology that opens up new opportunities for high-quality and stable intracellular recording of cardiomyocytes.In this work, we have prepared peptide-functionalized metal-organic frameworks (MOFs) as signal-amplifying tags for the detection of secreted protein acidic and rich in cysteine (SPARC). Furthermore, enzyme-MOF nanocomposites are fabricated via a coprecipitation strategy between horse radish peroxidase (HRP) and ZIF-90, where ZIF-90 is used as a protective support for HRP immobilization. Meanwhile, the peptide sequence has been designed as SPARC-binding peptide, which imparts biorecognition functionality to HRP@ZIF-90 for performing a colorimetric sensor. Therefore, during the test, HRP molecules can be quickly released from nanocomposites by acidic condition to catalyze chromogenic reaction, enabling the ultrasensitive detection of SPARC with a low detection limit of 30 fg/mL. Moreover, the content of SPARC in colon cancer tissues with different degrees of differentiation can be determined with this sensor, demonstrating that the expression of SPARC is closely related to the occurrence, invasion and metastasis of human colon cancer. Decades of research has focused on optimizing enzymes which utilize natural nicotinamide cofactors, namely nicotinamide adenine dinucleotide (phosphate) (NAD(P)+). Recent findings reignite the interest in engineering enzymes to utilize noncanonical cofactors, the mimetics of NAD+ (mNADs), which exhibit superior industrial properties in vitro and enable specific electron delivery in vivo. We compare recent advances in engineering natural versus noncanonical cofactor-utilizing enzymes, discuss design principles discovered, and survey emerging high-throughput platforms beyond the traditional 96-well plate-based methods. Obtaining mNAD-dependent enzymes remains challenging with a limited toolkit. To this end, we highlight design principles and technologies which can potentially be translated from engineering natural to noncanonical cofactor-dependent enzymes.Synthetic biology, when combined with immunoengineering (designated synthetic immunology), has enabled the invention of an arsenal of genetically encoded synthetic devices and systems to reprogram cells for therapeutic purposes. As the most successful example, cells of the immune system engineered with chimeric antigen receptors (CARs) have shown curative potentials for the treatment of hematological malignancies. We present herein emerging approaches of designing smart CARs to improve their safety, specificity and efficacy in cellular immunotherapy, and describe latest advances in applying CAR-engineered immune cells to target cancer and autoimmune diseases.This follow-up study of 82 children investigated the potential impact of early and recent exposure to mercury and lead on their neurodevelopmental performance at 5-8 years of age (2017-2018). Early exposure of these children to mercury, methylmercury, and lead was assessed during lactation at 3-12 months old, as well as their mother's exposure using measurements from a cross-sectional study (2011-2013). Only infants who failed to pass the neurodevelopment screening tools and/or had elevated mercury were included in this study. Urine and hair were sampled during the follow-up study to assess the children's recent exposure to mercury, methylmercury, and lead. Their cognitive performance and visual-motor integration were also measured using the Test of Non-Verbal Intelligence (TONI) and the Beery-Visual-Motor Integration (Beery VMI), respectively. The association between alterations in urinary porphyrins excretion and exposure to metals was analyzed and their influence on the children's neurodevelopment was explored.
