However, the usefulness of this technique has been limited by its relative complexity when compared to the classical two-hybrid system, since an additional selection step is required to eliminate the high background of uninformative truncation mutants. We have developed a new method that combines the classical and reverse two-hybrid systems to select loss-of-binding missense mutations in a single step. The strategy used to select against truncation mutants is based on the two-hybrid interaction between a C-terminal fusion peptide and the Tsg101 protein. We have applied this method to identify mutations in human glucokinase (GK) that disrupt glucokinase regulatory protein (GKRP) binding. Our results indicate that this method is very efficient and eliminates all the truncation mutants and false positives. The mutated residues identified in GK are involved in the GKRP binding interface or in stabilizing the super-open conformation of GK that binds GKRP. This technique offers an improvement over existing methods in terms of speed, efficiency and simplicity and can be used to study any detectable protein interaction in the two-hybrid system.Impulse control disorders are recognized as one of the behavioral and psychological symptoms of dementia (BPSD). Majority of studies on the treatment of BPSD related to impulsivity have rather focused on the aggression and agitation. In particular, it has not been investigated how cognitive declines are associated with impulsivity in community-dwelling elderly people. Here, we have measured the cognitive and memory functions and impulsivity of 212 elderly community-dwelling people using a psychometric test battery and analyzed the correlation between their level of impulsivity and cognitive functions by multiple regression analysis. Remodelin We found an elevation of impulsivity, which was evaluated by the Barratt Impulsiveness Scale-11, closely related to decline of cognitive functions, which were evaluated by the Montreal Cognitive Assessment and the Mini-Mental State Examination, and Logical Memory function, which were evaluated by the Wechsler Memory Scale-Delayed Recall. Then we have divided them into groups based on the severity of cognitive decline and conducted an analysis of each group, the result of which showed that as this tendency was particularly noticeable in the suspected dementia group. Therefore, we have concluded that heightened impulsivity is negatively associated with cognitive and memory functions in community-dwelling elderly people.In order to remediate Zn-contaminated livestock and poultry sewage, a zinc-resistant bacterial strain was screened and isolated from the manure of livestock and poultry and identified by molecular biology. The optimal conditions for removing zinc(II) from strain XZN4 were determined by single-factor experiments as follows within 3 times of repeated use, pH value was 5, initial concentration of zinc(II) was 100 mg/L, the amount of bacteria was 6 g/L, the temperature was 25-30 °C, and the removal equilibrium time was 60 min. Then, through adsorption isotherm model, scanning electron microscope image, energy dispersive spectrum analysis, infrared spectrum analysis and sterilization control experiment, it was found that the removal of zinc(II) by bacteria was single-molecule layer adsorption, which was carried out in coordination with degradation. The influence of different concentrations of copper(II), ammonia nitrogen, phosphorus, and chlortetracycline on the removal of zinc(II) from livestock and poultry sewage by XZN4 strain in the actual application was discussed. The bacteria can reduce the concentration of zinc(II) from the complex livestock and poultry waste water to below the discharge standard, and has a strong environmental tolerance, the highest removal rate reached 88.6% and the highest removal amount reached 10.30 mg/L. The screening and application of XZN4 strain can thus be of great significance for the microbial treatment of zinc(II) in complex livestock and poultry sewage. The results will provide guidance for the microbial remediation of heavy metal pollution.Control of heterointerfaces in advanced composite materials is of scientific and industrial importance, because their interfacial structures and properties often determine overall performance and reliability of the materials. Here distinct improvement of mechanical properties of alumina-matrix tungsten-carbide composites, which is expected for cutting-tool application for aerospace industries, is achieved via interfacial atomic segregation. It is found that only a small amount of Zr addition is unexpectedly effective to significantly increase their mechanical properties, and especially their bending strength reaches values far beyond those of conventional superhard composite materials. Atomic-resolution STEM observations show that doped Zr atoms are preferentially located only at interfaces between Al2O3 and WC grains, forming atomic segregation layers. DFT calculations indicate favorable thermodynamic stability of the interfacial Zr segregation due to structural transition at the interface. Moreover, theoretical works of separation demonstrate remarkable increase in interfacial strength through the interfacial structural transition, which strongly supports reinforcement of the interfaces by single-layer Zr segregation.Downhill folding has been defined as a unique thermodynamic process involving a conformations ensemble that progressively loses structure with the decrease of protein stability. Downhill folders are estimated to be rather rare in nature as they miss an energetically substantial folding barrier that can protect against aggregation and proteolysis. We have previously demonstrated that the prokaryotic zinc finger protein Ros87 shows a bipartite folding/unfolding process in which a metal binding intermediate converts to the native structure through a delicate barrier-less downhill transition. Significant variation in folding scenarios can be detected within protein families with high sequence identity and very similar folds and for the same sequence by varying conditions. For this reason, we here show, by means of DSC, CD and NMR, that also in different pH and ionic strength conditions Ros87 retains its partly downhill folding scenario demonstrating that, at least in metallo-proteins, the downhill mechanism can be found under a much wider range of conditions and coupled to other different transitions.
