Chemical analyses revealed differences in abundance of CECs between the 2 field sites and the controls, with GWR having the highest number and concentrations of CECs and metals. Cell bioassays screening for estrogenic, glucocorticoid, progestin, and dioxin-like activities were near or below detection limits in all river water samples, indicating a low potential for endocrine-related toxicity and tissue damage. Cell bioassay results were corroborated by the in vivo analyses. Field-exposed fish exhibited no changes in plasma hormones (e.g., estradiol), vitellogenin, or gonad maturation, but gene biomarkers of chemical exposure (cytochrome p450 1A and metallothionein) were significantly elevated, confirming exposure of the fish to complex chemical mixtures. The results demonstrate the value of a tiered monitoring approach to assess the sublethal effects of chemical mixtures on aquatic life. Environ Toxicol Chem 2021;40402-412. © 2020 SETAC.Sleep disruption in older adults living with Alzheimer's disease and related dementias (ADRD) is debilitating and contributes to increased institutionalization, reduced cognitive function, and accelerated disease progression. Furthermore, sleep disruption is linked to poor health outcomes in caregivers, such as decreased quality of life and increased caregiver burden. Given the potentially harmful side effects of pharmacologic treatment, nonpharmacologic approaches, such as music, may provide a safer alternative to reducing sleep disruption in this vulnerable population. A growing body of literature suggests that calming tailored music may improve sleep quality in older adults with memory loss, but its efficacy has not been demonstrated in older adults with ADRD in the community, where most older adults with ADRD live. If shown to be feasible and acceptable, tailored music interventions can then be tested for efficacy in reducing sleep disruption. This protocol details a wait-list randomized controlled trial (NCT04157244), the purpose of which is to test the feasibility, acceptability, and examine the preliminary efficacy of a tailored music listening intervention in older adults with ADRD who report sleep disruption. Music selections will be tailored to the music genre preferences of older adults with ADRD and account for known sleep-inducing properties. The feasibility of processes that is key to the success of the subsequent study will be examined. Preliminary efficacy of the intervention will be assessed using objective (actigraphy) and subjective (proxy-reported) sleep quality measures. In addition, qualitative data will be solicited, examining the acceptability and satisfaction with the intervention by individuals with ADRD.Cardiac resynchronization therapy (CRT) is one of the most effective therapies for heart failure with reduced ejection fraction and leads to improved quality of life, reductions in heart failure hospitalization rates and all-cause mortality. Nevertheless, up to two-thirds of eligible patients are not referred for CRT. Many forensic laboratories utilize gas chromatography-mass spectrometry following fatty acid methyl esterification to identify triglyceride residues in fire debris extracts. This study explores an alternate approach, which involves the identification of intact triglycerides by liquid chromatography-mass spectrometry-mass spectrometry (LC-MS/MS). 52 triglyceride-based oils and fats (22 different types) were analyzed by LC-MS/MS using multiple reaction monitoring to investigate variation in triglyceride content between different brands and types of oil and fat. Selected oils were then degraded by exposure to air, typical cooking conditions and/or fire conditions to simulate samples that are typically encountered by Fire Debris Analysts in fire investigation cases. Triglycerides were identified in all pristine and degraded oil samples, and relative peak areas for degraded samples often resembled their pristine oil counterparts. In samples where relative peak area differences were noted, more predominant degradation was observed for triglycerides with a higher proportion of poly-unsaturated fatty acids. Variability in triglyceride content between different brands and types of oil are discussed, as well as factors affecting the identification of triglyceride peaks in commercial oil samples, as compared to the corresponding analytical standard.Selenium (Se) toxicity to fish is primarily manifested via maternal transfer to the eggs, which may result in adverse effects on larval survival and development. The present study assessed the effects of egg Se concentrations derived via maternal transfer on early life-stage development, survival, and growth of Arctic grayling (Thymallus arcticus), a salmonid species not previously assessed for Se sensitivity. Fish gametes were collected from 4 streams in Alaska known to exhibit a range of egg Se concentrations. Eggs were fertilized and reared in the laboratory from hatch through post-swim-up. Larvae were assessed for survival, length, and weight, as well as deformities (skeletal, craniofacial, fin-fold) and edema based on a graduated severity index. Eggs from a total of 47 females were collected, with egg Se concentrations ranging from 3.3 to 33.9 mg kg-1 dry weight. No relationships were observed between larval endpoints evaluated and parent females' egg, muscle, or whole-body Se concentrations. Therefore, Se 10% effective concentrations (EC10s) were defined as the maximum measured Se concentrations >33.
